    Benzalacetone synthase (BAS) is a plant-specific chalcone synthase (CHS) superfamily type III poyketide synthase (PKS) that catalyzes a one-step decarboxylative condensation of 4-coumaroyl-CoA with malonyl-CoA. The diketide forming activity of Rheum palmatum BAS is attributed to the characteristic substitution of the conserved active-site Phe215 with Leu (numbering in Medicago sativa CHS). To further understand the intimate three-dimensional structural details of the enzyme catalyzed processes, we carried out X-ray crystallographic analyses of R. palmatum BAS. The crystal structures solved at 1.6Å and 1.8Å resolution suggested that, unlike M. sativa CHS, R. palmatum BAS utilizes an alternative active-site pocket to lock the aromatic moiety of the coumaroyl starter for the diketide formation reaction. These results provided structural insights into the functional diversity of type III PKS superfamily enzymes, and suggest strategies for the engineered biosynthesis of plant polyketides.

    Pentaketide chromone synthase (PCS) from Aloe arborescens is a novel plant-specific type III polyketide synthase (PKS) that produces 5,7-dihydroxy-2-methylchromone from five molecules of malonyl-CoA. On the basis of the crystal structures of wild-type and M207G mutant PCS, F80A/Y82A/M207G triple mutant was constructed and shown to produce an unnatural novel nonaketide naphthopyrone by sequential condensations of nine molecules of malonyl-CoAs. This is the first demonstration of the formation of a nonaketide by the structurally simple type III PKS. A homology model predicted that the active-site cavity volume of the triple mutant is increased to four times of that of the wild-type PCS.

    The structure of human translin at 2.2 A resolution is reported in space group C222(1). Translin forms a tetramer in the asymmetric unit. Although the monomer structure is almost the same as the crystal structure of murine translin in space group P2(1)2(1)2, the relative positions of the tetramers differ between the human and murine translins. This suggests that the multimerization of translin is flexible; the flexibility may be related to the binding to DNA/RNA.

    Human tau-protein kinase I (TPK I; also known as glycogen synthase kinase 3 beta; GSK3 beta) is a serine/threonine protein kinase that participates in Alzheimer's disease. Here, binary complex structures of full-length TPK I/GSK3 beta with the ATP analogues ADP and AMPPNP solved by the X-ray diffraction method at 2.1 and 1.8 A resolution, respectively, are reported. TPK I/GSK3 beta is composed of three domains: an N-terminal domain consisting of a closed beta-barrel structure, a C-terminal domain containing a 'kinase fold' structure and a small extra-domain subsequent to the C-terminal domain. The catalytic site is between the two major domains and has an ATP-analogue molecule in its ATP-binding site. The adenine ring is buried in the hydrophobic pocket and interacts specifically with the main-chain atoms of the hinge loop. The overall structure and substrate-binding residues are similar to those observed in other Ser/Thr protein kinases, while Arg141 (which is not conserved among other Ser/Thr protein kinases) is one of the key residues for specific ATP/ADP recognition by TPK I/GSK3 beta. No residues are phosphorylated, while the orientation of the activation loop in TPK I/GSK3 beta is similar to that in phosphorylated CDK2 and ERK2, suggesting that TPK I/GSK3 beta falls into a conformation that enables it to be constitutively active.

    Human tau-protein kinase I (TPK-I; also known as glycogen synthase kinase-3beta, GSK-3beta) is a serine/threonine protein kinase. Full-length TPK-I/GSK-3beta was expressed in Escherichia coli as a fusion protein with a 6xHis tag at the C-terminus and was crystallized using the hanging-drop vapour-diffusion method. Prismatic crystals of dimensions 0.4 x 0.2 x 0.1 mm were obtained using 12-15%(w/v) polyethylene glycol 6000 as a precipitant at 278 K. The crystals belong to the orthorhombic space group P2(1)2(1)2(1), with unit-cell parameters a = 82.9, b = 86.1, c = 178.1 A measured at 100 K, diffract to 2.3 A resolution and seem to contain two enzyme molecules per asymmetric unit.

    Several enzymes involved in the biosynthesis of natural products, such as plant polyketide synthase and CoA-ligase, exhibit remarkable catalytic versatility and substrate promiscuity. This makes the enzymes an excellent platform for enzyme engineering and further production of structurally divergent molecules. In this study, on the basis of the X-ray crystal structures of the enzymes, we succeeded in production of chemically and structurally divergent unnatural natural products, by structure-based enzyme engineering and precursor directed biosynthesis. In addition, we succeeded in elucidation of biosynthetic machinery of complex natural products including calyculin, teleocidin, and antimycin.
