    Abstract

    The Papanicolaou stain, consisting of five dyes, provides extensive color information essential for cervical cancer cytological screening. The visual observation of these colors is subjective and difficult to characterize. Direct RGB quantification is unreliable because RGB intensities vary with staining and imaging conditions. Stain unmixing offers a promising alternative by quantifying dye amounts. In previous work, multispectral imaging was utilized to estimate the dye amounts of Papanicolaou stain. However, its application to RGB images presents a challenge since the number of dyes exceeds the three RGB channels. This paper proposes a novel training-free Papanicolaou stain unmixing method for RGB images. This model enforces (i) nonnegativity, (ii) weighted nucleus sparsity for hematoxylin, and (iii) total variation smoothness, resulting in a convex optimization problem. Our method achieved excellent performance in stain quantification when validated against the results of multispectral imaging. We further used it to distinguish cells in lobular endocervical glandular hyperplasia (LEGH), a precancerous gastric-type adenocarcinoma lesion, from normal endocervical cells. Stain abundance features clearly separated the two groups, and a classifier based on stain abundance achieved 98.0% accuracy. By converting subjective color impressions into numerical markers, this technique highlights the strong promise of RGB-based stain unmixing for quantitative diagnosis.

    PURPOSE: The color of Papanicolaou-stained specimens is a crucial feature in cytology diagnosis. However, the quantification of color using digital images is challenging due to the variations in the staining process and characteristics of imaging equipment. The dye amount estimation of stained specimens is helpful for quantitatively interpreting the color based on a physical model. It has been realized with color unmixing and applied to staining with three or fewer dyes. Nevertheless, the Papanicolaou stain comprises five dyes. Thus, we employ multispectral imaging with more channels for quantitative analysis of the Papanicolaou-stained cervical cytology samples. APPROACH: We estimate the dye amount map from a 14-band multispectral observation capturing a Papanicolaou-stained specimen using the actual measured spectral characteristics of the single-stained samples. The estimated dye amount maps were employed for the quantitative interpretation of the color of cytoplasmic mucin of lobular endocervical glandular hyperplasia (LEGH) and normal endocervical (EC) cells in a uterine cervical lesion. RESULTS: We demonstrated the dye amount estimation performance of the proposed method using single-stain images and Papanicolaou-stain images. Moreover, the yellowish color in the LEGH cells is found to be interpreted with more orange G (OG) and less Eosin Y (EY) dye amounts. We also elucidated that LEGH and EC cells could be classified using linear classifiers from the dye amount. CONCLUSIONS: Multispectral imaging enables the quantitative analysis of dye amount maps of Papanicolaou-stained cytology specimens. The effectiveness is demonstrated in interpreting and classifying the cytoplasmic mucin of EC and LEGH cells in cervical cytology.

    INTRODUCTION: Although the widespread use of screening tests and HPV vaccines for squamous cell carcinoma has led to early detection and treatment, effectiveness is limited for cervical adenocarcinoma. Lobular endocervical glandular hyperplasia (LEGH) corresponds to gastric metaplasia but is regarded as a pathological condition with subtle morphological abnormalities. LEGH is a benign lesion and a precursor to gastric-type adenocarcinoma. We herein developed an objective and quantitative method by applying an image analysis technique to overcome the difficulties associated with the differential diagnosis of LEGH in uterine cervical cytology. This approach is expected to enable the early detection and accurate diagnosis of LEGH. METHODS: We extracted signal values for the nucleus and cytoplasm from microscopic images of cytological specimens of normal endocervical (EC) and LEGH cells. These values were then converted into CIELAB and sRGB values to create color distribution maps, and color unmixing techniques were applied to assess the spectral absorbance of each pigment. RESULTS: The CIELAB signal values extracted from the nuclear images of LEGH cells exhibited lower values than those of EC cells. Furthermore, based on color distribution maps, the cytoplasm of EC cells exhibited shades from purple to pink, while LEGH cells showed a distribution toward yellow. CONCLUSIONS: This study reveals that, compared to EC cells, LEGH cells exhibit lower nuclear signal values and increased nuclear chromatin content. Thus, assessing the relative difference in cytoplasmic color tones between them may become an effective indicator for distinguishing between EC and LEGH cells.

    PURPOSE: Endometrial cancer (EC) is one of the most common types of cancer affecting women. While the hematoxylin-and-eosin (H&E) staining remains the standard for histological analysis, the immunohistochemistry (IHC) method provides molecular-level visualizations. Our study proposes a digital staining method to generate the hematoxylin-3,3'-diaminobenzidine (H-DAB) IHC stain of Ki-67 for the whole slide image of the EC tumor from its H&E stain counterpart. APPROACH: We employed a color unmixing technique to yield stain density maps from the optical density (OD) of the stains and utilized the U-Net for end-to-end inference. The effectiveness of the proposed method was evaluated using the Pearson correlation between the digital and physical stain's labeling index (LI), a key metric indicating tumor proliferation. Two different cross-validation schemes were designed in our study: intraslide validation and cross-case validation (CCV). In the widely used intraslide scheme, the training and validation sets might include different regions from the same slide. The rigorous CCV validation scheme strictly prohibited any validation slide from contributing to training. RESULTS: The proposed method yielded a high-resolution digital stain with preserved histological features, indicating a reliable correlation with the physical stain in terms of the Ki-67 LI. In the intraslide scheme, using intraslide patches resulted in a biased accuracy (e.g., R = 0.98 ) significantly higher than that of CCV. The CCV scheme retained a fair correlation (e.g., R = 0.66 ) between the LIs calculated from the digital stain and its physical IHC counterpart. Inferring the OD of the IHC stain from that of the H&E stain enhanced the correlation metric, outperforming that of the baseline model using the RGB space. CONCLUSIONS: Our study revealed that molecule-level insights could be obtained from H&E images using deep learning. Furthermore, the improvement brought via OD inference indicated a possible method for creating more generalizable models for digital staining via per-stain analysis.

    A Fresnel Zone Aperture (FZA) mask for a lensless camera, an ultra-thin and functional computational imaging system, is beneficial because the FZA pattern makes it easy to model the imaging process and reconstruct captured images through a simple and fast deconvolution. However, diffraction causes a mismatch between the forward model used in the reconstruction and the actual imaging process, which affects the recovered image's resolution. This work theoretically analyzes the wave-optics imaging model of an FZA lensless camera and focuses on the zero points caused by diffraction in the frequency response. We propose a novel idea of image synthesis to compensate for the zero points through two different realizations based on the linear least-mean-square-error (LMSE) estimation. Results from computer simulation and optical experiments verify a nearly two-fold improvement in spatial resolution from the proposed methods compared with the conventional geometrical-optics-based method.

    This study proposes a novel computational imaging system that integrates a see-through screen (STS) with volume holographic optical elements (vHOEs) and a digital camera unit. Because of the unique features of the vHOE, the STS can function as a holographic waveguide device (HWD) and enable the camera to capture the frontal image when the user gazes at the screen. This system not only provides an innovative solution to a high-quality video communication system by realizing eye-contact but also contributes to other visual applications due to its refined structure. However, there is a dilemma in the proposed imaging system: for a wider field of view, a larger vHOE is necessary. If the size of the vHOE is larger, the light rays from the same object point are diffracted at different Bragg conditions and reflect a different number of times, which causes blurring of the captured image. The system imaging process is analyzed by ray tracing, and a digital image reconstruction method was employed to obtain a clear picture in this study. Optical experiments confirmed the effectiveness of the proposed HWD-STS camera.

    A mask-based lensless camera optically encodes the scene with a thin mask and reconstructs the image afterward. The improvement of image reconstruction is one of the most important subjects in lensless imaging. Conventional model-based reconstruction approaches, which leverage knowledge of the physical system, are susceptible to imperfect system modeling. Reconstruction with a pure data-driven deep neural network (DNN) avoids this limitation, thereby having potential to provide a better reconstruction quality. However, existing pure DNN reconstruction approaches for lensless imaging do not provide a better result than model-based approaches. We reveal that the multiplexing property in lensless optics makes global features essential in understanding the optically encoded pattern. Additionally, all existing DNN reconstruction approaches apply fully convolutional networks (FCNs) which are not efficient in global feature reasoning. With this analysis, for the first time to the best of our knowledge, a fully connected neural network with a transformer for image reconstruction is proposed. The proposed architecture is better in global feature reasoning, and hence enhances the reconstruction. The superiority of the proposed architecture is verified by comparing with the model-based and FCN-based approaches in an optical experiment.

    ハイパースペクトルイメージングが抱える空間解像度の課題を計測の際に解決するため,新しい計測手法の検討を中心に行なった.まず,現状の課題解決のためのアプローチを調査し,考慮されていないが実際検討が必要な残された課題の洗い出しにより,サンプリング距離より小さな対象物の情報欠落が確認された.これを解決するため,撮影対象全体の情報を低空間解像度で撮影するのではなく,部分的に高空間・波長解像度の情報を取得し,補助画像を用いることで全体の情報を再構成する新しい計測アプローチに至り,計測原理の構築と再構成手法の提案を行っている.

    To adopt graph signal processing into hyperspectral (HS) imaging, we deeply analyzed the prior knowledge on HS images. Thanks to effective evaluation of spatial architecture, our proposed method can estimate an HS image with high spatial and spectral resolution. From the results, we can see the spatial architecture helps to improve the performance for HS image restoration, so we tried to construct the methodology of spatial architecture into the graph representation for HS image restoration.
    Then, we proposed a new hyperspectral imaging framework near by real imaging systems. As a result, we showed the problem can be resolved by the suitable evaluation of apriori knowledge on HS images.

    本年度は,ハイパースペクトル画像の波長情報の構造を考慮したハイパースペクトル画像復元手法に関する研究を行った.具体的には,ハイパースペクトル画像の先験的情報である空間・波長方向の区分的なめらかさと波長方向の低ランク性を評価する二種類の正則化関数を同時に利用した復元手法の提案を行った.これによって,これまで行ってきた区分的滑らかさのみを用いた手法よりも多くの構造的特徴を利用することができるため,より高精細な復元を達成できる.また,昨年度まで行ってきた研究の精査を行い,その結果をまとめたものを学術論文に掲載した.
