    The ultimate goal of regenerative medicine is the transplantation of a target organ generated by the patient's own cells. Recently, a method of organ generation using pluripotent stem cells (PSCs) and blastocyst complementation was reported. This approach is based on chimeric animal generation using an early embryo and PSCs, and the contribution of PSCs to the target organ is key to the method's success. However, the contribution rate of PSCs in target organs generated by different chimeric animal generation methods remains unknown. In this study, we used 8-cell embryo aggregation, 8-cell embryo injection, and blastocyst injection to generate interspecies chimeric mice using rat embryonic stem (ES) cells and then investigated the differences in the contribution rate of the rat ES cells. The rate of chimeric mouse generation was the highest using blastocyst injection, followed in order by 8-cell embryo injection and 8-cell embryo aggregation. However, the contribution rate of rat ES cells was the highest in chimeric neonates generated by 8-cell embryo injection, and the difference was statistically significant in the liver. Live functionality was confirmed by analyzing the expression of rat hepatocyte-derived drug-metabolizing enzyme. Collectively, these findings indicate that the 8-cell embryo injection method is the most suitable for generation of PSC-derived organs via chimeric animal generation, particularly for the liver.

    <p>Recently, a novel culture system consisting of primary hepatocytes structured over a network of endothelial cells on the Engelbreth-Holm-Swarm (EHS) gel has been reported. This in vitro liver model on the EHS gel (IVLEHS) has been shown to maintain the expression of hepatic genes and their functional activity. Moreover, the IVLEHS was more sensitive to xenobiotics than hepatocyte monocultures, suggesting the potential utility of this culture system for compound hepatotoxicity screening. However, the effect of this three-dimensional structure formation on the cellular metabolic profile of hepatocytes in the IVLEHS is not well understood. To address this concern, we performed metabolome analysis using capillary electrophoresis-time of flight mass spectrometry. Between the IVLEHS and mono-cultured hepatocytes on the EHS gel, there was no significant difference in the levels of metabolites of the urea cycle and the tricarboxylic acid cycle, essential amino acids, and adenylate energy charge (AEC) which is an important indicator of cellular energy status. On the other hand, acetaminophen-dependent decrease of the AEC in the IVLEHS was greater than that in the monoculture, suggesting the higher sensitivity of IVLEHS to acetaminophen-induced hepatotoxicity which is caused by metabolic activation of this drug. Further analysis showed that the levels of taurocholate, one of the major conjugated bile acids, were higher in the IVLEHS than in the monoculture. Considering that the construction of the IVLEHS did not seem to disturb the major cellular metabolism, our findings would strengthen the concept that IVLEHS would have beneficial effects on the maintenance of hepatic functions.</p>

    Our purpose in the present study was to establish a system for in vitro hepatic morphogenesis, consisting of not only hepatocytes but also cell lineages supporting hepatic differentiation, such as cardiomyocytes and endothelial cells, from murine ES cells. We succeeded in establishing a novel system of hepatic morphogenesis from murine ES cells based on naturally occurring embryological events.

    Epigallocatechin-3-gallate (EGCG), exhibited protective effect against rubratoxin B (RB)-induced apoptosis in cultured rat hepatocytes. In this study, the mechanism of hepatoprotection by EGCG was investigated. Hepatocytes were concomitantly treated with RB and EGCG and then the expression levels of mitogen-activated protein kinase (MAPK) p38, caspase-3 and caspase-8 genes were measured by reverse transcriptase-polymerase chain reaction (RT-PCR). p38 was induced by RB, indicating that RB-induced apoptosis was mediated by p38 pathway. On the contrary, the elevation of p38 by RB was diminished by EGCG co-treatment. The analysis of caspase-3 mRNA expression showed the same tendency as observed for the p38. To further elucidate, the expression level of mitogen-activated kinase kinase 6 (MKK6) gene, upstream signaling factor of p38, was analyzed by RT-PCR. EGCG also suppressed the expression of MKK6 mRNA. These results suggest that EGCG protects hepatocytes from RB toxicity by the suppression of the gene expression in p38 and caspase pathway at any points examined.

    The interaction of rubratoxin B (RB) with bovine serum albumin (BSA) was investigated. Circular dichroism spectroscopy shows the interaction of RB with BSA as conformational change of BSA. Fluorescence quenching study suggests that RB binds to BSA at a few sites. At low concentration of RB, RB firstly binds to BSA at any binding sites far from Trp residues. By increasing RB concentration, RB binds to BSA at any binding sites near Trp residues.

    Progressive multifocal leukoencephalopathy (PML) is considered to be caused by the genome mutation of archetype to PML-type JC virus to acquire the potential to infect with astrocytes and oligodendrocytes in brain tissue. So far, SV40 T antigen-producing cell lines only have been infectable with JC viruses, but here we succeeded to be establish SV40 T antigen-independent culture system of JC virus infection by the differentiation of human iPS cells to astrocytes and oligodendrocytes. PML-type JC virus could infect human iPS cell-derived oligodendrocytes significantly higher than astrocytes. Furthermore, we succeeded to develop a microfluidic device for JC virus infection.

    A device, equipping two independent channels separated by a porous membrane where intestinal epithelial (C2BBe1) cells were cultured, and electrodes for measurement of TEER was invented in this study. After C2BBe1 cells formed tight junction in the whole area on the membrane E. coli was supplied from the upper channel. The tight junction of C2BBe1 cells could be maintained with E.Coli for 3 days by the adequate flow rate, however failed by the static condition. Furthermore, L. murinus was succeeded to be co-cultured in the above co-culture fluidic device. On the other hand, after macrophage cell line RAW264 cells were supplied from the lower channel of C2BBe1 cell-culture device, E.Coli was inoculated in the lower channel. The tight junction was disrupted by the E.Coli, but maintained without the E.Coli. By using this system, it is expected to establish not only co-culturing system of intestinal micro-biota and intestinal epithelial tissue on a chip but also enteritis culture model.

    In mammals, digested substances taken from mouth were incorporated and metabolized at the intestine, and then were delivered to liver through hepatic portal vein. Liver plays crucial roles such as energy source accumulation and supply to whole body, detoxification and excretion by conjugation. Because, for drug development, it is necessary to investigate kinetics, metabolism, and toxicity of drugs in intestine and liver, many animal experiments and clinical studies have been carried out. So, we succeeded to establish ES/iPS-derived in vitro drug metabolism assay system closed to real intestine and liver, and these ES/iPS-derived intestine and liver tissues were cultured on our original fluidic devices. These results must be expected to be animal experiment alternatives.

    Proliferation of liver progenitor cells (LPCs) accompanies on many types of liver injury when the replication of hepatocytes is blocked. Although there are enormous number of researches for the characterization of LPCs, their origin and their roles in liver regeneration is still on debate. We recently found that a subpopulation of liver progenitor cells, which situate in periportal area after GalN-induced rat liver damage, express Pdx-1 gene. To address the roles of Pdx-1 in LPCs during liver regeneration, we performed overexpression and gene knockout experiments of Pdx-1 using a LPC cell line. We found that the change of Pdx-1 expression does not alter LPC proliferation. We also performed microarray analysis to compare gene expression of wild type- and knockout- LPCs for the Pdx-1 gene. The data implies that Pdx-1 is involved in the expression of neuroendocrine phenotype of LPC, as well as the suppression of hepatocyte differentiation.

    The vHELP, having heart, endothelium, liver, and pancreatic islet, in vitro living system was succeeded to be constructed from mouse embryonic stem cells. Some biochemical analysises could indicate that these tissue-specific functions in the vHELP. The vHELP was cultured with medium flow in the original fluidic device made from polydimethylsiloxane. Higer hepatic functions were gained by this culture in the fuluidic device.
    Consequently, we have succeeded to construct mouse ES cell-derived vHELP as a minimal mammal in vitro system.

    To construct an in vitro model of hepatic tissue organogenesis by developing synthetic biological systems, we challenged to establish an adequate platform for synthetic approach in mammalian. Gene expression analysis was carried out at several liver developmental stages in mouse. Next, a hepatic endothelial cell strain was established as support cell for hepatic differentiation of mouse ES cells. To prepare in vitro system corresponding to early hepatic development period, conditional expression system of LIF and activin was constructed and was introduced into endothelial cell line to keep undifferentiation and endodermal differentiation of mouse ES cell, respectively. Mouse hepatic progenitor cells were cultured and differentiated to mature hepatocytes or cholagiocytes under the laminar flow-supplied different medium conditions in a same micro-fluidic device. In conclusion, we could show an adequate example as platform in consideration of cross talk for synthetic biology in mammal.

    We present a novel approach to generate parenchymal organs, such as liver and hollow tissues, blood vessels, based on the use of cultured cells, atelo collagen I and cytokines. Dense connective tissues were generated simultaneously by accumulating collagen fibrils and fibroblasts on a sheet of polylactic acid mesh or artificial blood vessels of synthetic polymers simultaneously using a bioreactor system that we designed by ourselves. For artificial liver tissues, hepatic cells were introduced into the reactor following the perfusion of another medium containing atelo collagen I. In order to make artificial vessels, cultured endothelial cells were circulated the bioreactor. Embedded hepatic cells can retain their specific functions, such as the albumin and the urea production. The two-layered artificial vessels were well-defined i.e. the layer of collagen fibrils and the endothelial layer. These results suggest that this novel approach can produce biological tissues mechanically solid for surgical operation.

    We have succeeded to establish to construct in vitro liver model culturing primary hepatocytes, portal vein-ligation-induced hepatic stem cells, or human ES/iPS cell-derived hepatocytes with the networks of endothelial cells on EHS gels. The culture of these in vitro liver models in our original microfluidic system had significantly larger advantages for liver specific potentials compared to the non-fluidic. Our results lead strong expectation of our in vitro liver chips for application of drug metabolism and disposition.

    We have designed a new device to form artificial tissues from culture medium supplemented with collagen I and various cells less than 24 hours. Moreover, we stack up 3 different tissue layers in the same bioreactor by supplementing newly prepared culture medium containing various extracellular matrices, such as collagen and proteoglycans, and different types of cells. Embedded cells restore their specific functions of original tissues, i.e. up-regulation of smooth muscle type myosin the tissues. Using this technique, we can supply artificial tissues applicable to clinical use with relative ease.

  • チタン表面への生体分子および細胞接着挙動のナノレベル・2次元・リアルタイム測定

    チタンは医学・歯学の分野において人工歯根・人工骨の材料として重要な位置を占めており,その用途・規模は年々拡大している。本研究はチタンを用いた人工歯根・人工骨の治療期間短縮ならびに適応範囲拡大を目指し,チタン表面での生体物質の相互作用をnmオーダーでかつ多点で解析するシステムとして,2次元イメージング表面プラズモン解析装置(SPR)に適したイメージングSPR用チタンセンサーの設計・チタンSPRセンサー作製を目指した。
    本研究のチタンSPRセンサーは二酸化チタンを直接表面に被覆せず,金属チタン表面を大気中で酸化させた。測定結果より,本研究で開発したチタンSPRセンサーは,タンパク質吸着量をng/mm^2のオーダーで精密に測定できることが明らかとなった。また,このセンサーを用いることにより,タンパク質の吸着過程を経時的に測定することも可能となった。これより,チタンセンサーは大気中で酸化された金属チタン表面での生体分子の相互作用を求める上で極めて有用であることが示唆された。
    また,今回作製した2次元イメージングSPRは,(1)表面に存在する極微量の物質をpg〜ng/cm^2のオーダーでリアルタイムに定量可能,(2)多数の生体分子間相互作用に関するデータを瞬時に取得可能,(3)2次元平板上で同時に起こる複数(〜数1,000種類)の反応をパラレル分析することが可能であった。さらにユニークな特徴として(4)イメージングSPRでありながら正確な入射光角度-反射光強度のSPRスペクトルを測定可能であり,他のイメージングSPRと比べてより正確な物質吸着量を求めることができるなどの優れた長所を有していることも明らかとなった。
    以上より,2次元イメージングSPRとチタンSPRセンサーはチタン材料の生体適合性の研究に重要な役割を果たすことが期待できる。

  • Development of functional Ti implants with release of growth factors

    In order to develop functional Ti implants with release of growth factors, biodegradable materials as a carrier have to adhere strongly to Ti surface. Lactide copolymers synthesized in this study hardly adhered to TI surface.
    Ti plates were treated with 0.1 and 1 N hydrochloric acid, 37wt% phosphoric acid or kept untreated. The tensile bond strength of the composite cement to the untreated and pre-treated Ti plates was determined without and after 20,000 thermo-cycles. XPS was used to chemically analyze the effect of the three Ti pre-treatments, as well as the interaction of 10-methacryloxydecyl dihydrogen phosphate (10-MDP) with Ti treated with 1 N HCl. Although no significant difference in immediate tensile bond strength was measured, thermo-cycling significantly decreased the bond strength of all experimental groups except for Ti treated with 1 N HCl. XPS demonstrated that H_3PO_4 was strongly adsorbed on the Ti surface. These results clearly suggest that phosphorylation was effective to get strong attachment to the Ti surface. Ti pre-treated with 1 N HCl improved the adsorption of 10-MDP as compared to untreated Ti.
    Cleaned Ti disks were immersed into three different concentrations of polyphosphoric acid solution (0.1,1 and 10 wt%) for 24 hours at 37℃. Ti immersed in distilled water for 24 hours at 37℃ served as control. Degrees of cell attachment (1,3,5 hours after cell seed) and proliferation (1,3,5 and 7 days after cell seed) on each treated Ti disk were evaluated by MTS assay. A significantly higher cell attachment was found on Ti treated with polyphosphoric acid in contrast to untreated Ti (control) for all three culture periods. MTS assay also revealed that cell proliferation levels significantly increased following a polyphosphoric-acid dose dependency. It was concluded that polyphosphoric-acid treatment of Ti enhanced the attachment and proliferation of hBMSCs.

  • 肝臓の再生医学研究のためのモデル動物の開発とES細胞を用いた肝細胞の再生

    昨年度にマウスES細胞から、肝実質細胞のみならず、肝非実質細胞である血管(類洞)内皮細胞と肝実質細胞が共に分化・出現する肝組織分化誘導システムの開発に成功(Ogawa, S, Y.Tagawa, et al.,Stem Cells)した。本年度はこのマウスES細胞由来肝組織分化誘導システムが薬物動態試験への応用が可能であることを示した(Tsutui, M, ・・・, and Y.Tagawa, Drug Metabolism and Disposition 印刷中)。
    まず、このマウスES細胞由来肝組織中の薬物代謝に関連するチトクロームP450遺伝子ファミリーがマウス胎仔肝や成体肝実質細胞と同様に発現していることをRT-PCRで確認した。これらの発現しているCYPが実際に活性を持っているかどうかを調べるために、マウスES細胞由来肝組織のメディウム中にテストステロンを添加し、その水酸化箇所と活性を検討した。成体肝実質細胞の水酸化パターンとは異なったが、マウス胎仔肝と全く同様に6β、16α、2α、2βが水酸化を受け、15α、7α、16βが水酸化を受けなかったことがHPLCによる解析でわかった。この水酸化活性は、胎仔肝や成体肝実質細胞に比べると、非常に高いことが確認できた。このことは、アンモニアの代謝能力と同様であり、in vitroの肝実質細胞の肝機能活性よりも個体における肝臓器官の活性に近いことが示唆された。さらに、CYPの活性は誘導されることも知られているので、フェノバルビタールを添加した後に、テストステロンの水酸化誘導能の変化について検討した。テストステロンの6βと16βに関して約2.5倍の水酸化活性の誘導が確認できた。以上のことから、このマウスES細胞由来肝組織は、CYPに関連する薬物代謝活性を十分に持っていることがわかり、肝臓の薬物代謝能力の獲得メカニズムの解析に有効であることが示唆できた。

    This research project has demonstrated the possibility of fetal cardiac tissues and ES cell derived cardiocytes adding spontaneous contraction to vessels by their ectopic transplantation around vascular tubes and its application for cardiac diseases.
    1.Mouse fetal cardiac tissues were transplanted around the abdominal aorta of the nude mouse. They survived securing blood supply from the testicular artery and showed spontaneous contraction and action potential. Morphological analysis confirmed that the contracted tissues contained cardiocytes. This method will be applied for implanted ventricular assist system.
    2.ES cell-derived cardiocytes were differentiated from ES cells by hanging drop method or a new method using conical tubes and transplanted around the abdominal aorta of the nude mouse. On and after 7 days spontaneous contracted tissues were recognized on the aorta and generated irregular action potentials. Morphological and molecular biological analysis showed these tissues including atrial and ventricular cardiocytes. Moreover, it was confirmed that atrial cardiocytes contained atrial natriuretic peptide by RT-PCR and immunohistochemistry.
    3.To create a new type of implanted lung-heart apparatus, TiO_2 nanotubes with photodecomposition of water into O_2 and H_2 as an Oxygen supplier and fetal cardiac tissues as a pump were implanted under the inguinal region of the nude mouse. After 4 weeks, Sv02 of the surface of the treated skin were measured, showing higher percentage than controls. Transplanted cardiac tissues contracted spontaneously and showed features of cardiocytes ultrastructures.
    4.The surgical method for partial transplantation using cardiac tissues was developed. It was featured by transverse incision of the chest through the intercostals space and sternum and offered the wide exposure for treatment. A part of the greater omentum containing cardiac tissues were sutured easily to the wall of the original heart by this method.

    From our research, following results are obtained.
    1) A quite new strain of spontaneous eosinophilic rat (Matsumoto Eosinophilia shinshu rats: MES/sdi) was produced.
    2) All animals (more than 1500 rats tested) develop eosinophilia and bone marrow cell proliferation also observed.
    3) From the results of anti-IL-5 study, it is suggested that IL-5 plays an important roles in pathogenesis of eosinophilia.
    4) Histopathologically, eosinophilic proliferation and infiltration in the bone marrow, gastrointestinal tract, mesenteric lymph node, and spleen was found.
    5) MES rats were resistant to infection with T. spiralis newborn larvae. This indicates that eosinophils destroy the newborn larvae in vivo.
    From these results, it is concluded that MES rats could be a useful animal model for research on HES (Hypereosinophilic syndrome in humans) because the hypereosinophilia, with many associated eosinophilic lesions seen in the MES rats, is comparable to the features of HES in humans and other animals.

  • 発生工学的手法を応用IFN-γおよびIL-6の肝炎における役割の解析

    コンカナバリンA(Con A)誘導肝炎は、ヒトウイルス性または自己免疫性肝炎のマウスモデルとして知られている。Con Aを投与するとIFN-γやIL-6をはじめとしてIL-1、IL-2、IL-4といった多くのサイトカインが誘導されるために、一般的な肝炎発症におけるIL-6やIFN-γの詳細な解析は難しい。IFN-γKOマウスを用いて、Con Aにより誘導されたIFN-γがFas-FasLシステムを活性化することによって肝臓細胞のアポトーシスを誘導し肝障害を起こすことがわかった。また、IL-6KOマウスや抗IL-6抗体投与実験により、Con A投与直後に誘導されるIL-6は肝炎に対して抑制的に働くが、Con A投与後後半のIL-6は肝炎を悪化させることもわかった。そこで次に、組織特異的誘導型IFN-γやIL-6過剰発現マウスを作成し、循環器系または肝臓特異的にIFN-γやIL-6を過剰発現させるシステムの開発を重点的におこなった。Con A刺激した脾臓細胞からマウスIFN-γおよびIL-6のcDNAをRT-PCRにより得た。これらのcDNAをテトラサイクリン誘導プロモータの下流に連結したレトロウイルスベクターを構築した。このベクターをエンベロープタンパク質産生細胞PT67細胞に導入し、培養上清中に感染性・非増殖性ウイルス粒子を得た。そして、これら外来遺伝子を持ったレトロウイルスをES細胞に感染させ、低コピーの遺伝子導入を行った。薬剤選別をおこない、これらの外来遺伝子が染色体DNAに組み込まれたESクローンを単離した。現在、これらのESクローンからキメラマウスを作成している。
    また、Con Aを投与する前にフルクトースキトサンや酸性糖タンパク質を投与すると、肝障害の誘導が抑えられることがわかった。これらの糖/糖タンパク質は、免疫細胞の活性化に関わっていることが知られており、サイトカイン産生と関わっているかどうかを検討している。

    1.炎症や免疫応答において重要な役割を果たすと考えられている、IL-1α、IL-1β、IL-1α/β、IL-1レセプターアンタゴニスト、およびIFN-γの遺伝子欠損マウスを作製することに成功した。
    2.IL-1欠損マウスを用い、IL-1が末梢だけでなく中枢で発熱やグルココルチコイドの分泌制御に重要な役割を果たしていることを明らかにした。この結果は、IL-1が生体の恒常性の維持に主要な役割を果たしていることを示唆する。
    3.IL-1レセプターアンタゴニスト欠損マウスは、関節リウマチによく似た自己免疫性の関節炎を発症することを明らかにした。この結果は、IL-1が免疫系の恒常性維持にも重要な役割を果たしていることを初めて示したものであり、ヒトの関節リウマチの原因の1つとして、IL-1系の異常が考えられることを示唆した。
    4.関節リウマチの発症機構を、我々が独自に開発したHTLV-Iトランスジェニックマウス関節炎モデルを用いて検討し、IL-1およびIL-6が病態形成に重要な役割を果たしていることを明らかにした。この結果は、関節リウマチの治療に1つの示唆を与えるものである。
    5.同じく、IFN-γ欠損マウスを用い、肝炎の発症にIFN-γが大きな役割を果たしていることを明らかにした。
    6.多くの研究者との共同研究により、Mycobacteria、マウス肝炎ウイルス、Helicobacterpylori、Babesia、HSV-1、Toxoplasma、Lieshmaniaなど種々の感染症におけるIFN-γの役割を検討し、重要な役割を果たしていることを示した。また、サイトカインネットワークにおける役割を検討した。
    7.IL-1についても、炎症や、HIV、マラリア、Leishmaniaなどの感染における役割を検討し、このサイトカインが病態形成に重要であることを示した。

    1.インターロイキン-1(IL-1)遺伝子ノックアウト(KO)マウスの作製とその解析
    昨年度までにIL-1αとIL-1β遺伝子それぞれ単独及びダブルKOマウスを作製しており、今年度はIL-1受容体アンタゴニスト(IL-1ra)遺伝子KOマウスの作製に成功し、IL-1関連遺伝子はすべて破壊した。これらのマウスを用いて、まずLPSによるエンドトキシンショックに対する抵抗性を調べた。IL-1raKOマウスは高感受性を示したが、予想に反してIL-1α/βダブルKOマウスの感受性は野生型と差がなかった。次にテレビン油投与による発熱に対しては、IL-1αKOマウスは野性型と同様に発熱するに対して、IL-1βKOマウスは発熱せず、逆にIL-1raKOマウスは野生型以上に発熱することがわかり、IL-1βが炎症性の発熱を引き起こしていることが明らかとなった。また感染防御では、リステリア菌の感染に対して、IL-1α/βダブルKOマウスは感受性が高いことも明らかとなった。
    2.インターフェロン-γ(IFN-γ)のConA誘導肝炎における役割
    ConA誘導肝炎は、T細胞から分泌されるサイトカインによって引き起こされ、ヒトのウイルス性肝炎のモデルとして重要である。昨年度にIFN-γKOマウスやlprマウスを用いて、この系ではIFN-γがFasシステムを活性化することにより肝実質細胞のアポトーシスを引き起こしていることを明らかにしたが、今年度はTNF-αKOマウスやiNOSの阻害剤を用いて、更に発症の機構を解析した。IFN-γKOマウスは発症が抑制されたのに対して、TNF-αKOマウスやiNOSの阻害剤であるL-NMMA投与マウスでは発症が抑制されず、これらは発症に関与しないことがわかった。また、ConA投与後に誘導されてくるFasとICEがIFN-γKOマウスでは誘導されないことを見いだし、IFN-γによるEasとICEの発現誘導がFasシステムの感受性を高めている可能性が示唆された。

    Con A誘導肝炎は、T細胞から分泌されるサイトカインによって引き起こされる病気で、ヒトのウイルス性肝炎のモデルとして重要である。先に、我々はIFN-γKOマウスや1prマウスを用いて、この系ではIFN-γがFasシステムを活性化することにより肝実質細胞アポトーシスを引き起こしていることを明らかにした。ヒトのウイルス性肝炎の場合もIFN-γの亢進が認められることから、同様の機構が働いている可能性が考えられる。本年度はIFN-γ及びTNF-α,IL-1の各KOマウスを用いて、IFN-γ誘導アポトーシスの機構について検討した。
    Con A誘導肝炎に対してIFN-γKOマウスは低感受性を示したのに対し、TNF-α、IL-1α/βKOマウスでは発症が抑制されなかった。Con A投与のコントロールマウスでは、Fasの下流のプロテアーゼとして知られるICEの発現が2倍に上昇し、IFN-γKOマウスではその上昇は見られなかった。以上のことから、Con A肝炎において、TNF-αやIL-1は発症に関係していないことがわかり、さらに、fasの発現亢進だけでなく、IFN-γによるICEの発現誘導がCon A肝炎におけるFasシステムの感受性を高めている可能性が示唆された。さらに、ウイルス性急性肝炎患者の生検の病理解析よりCon A誘導肝炎同様に肝臓細胞のアポトーシスを確認した。最近、その他の多くのウイルスにおいても感染細胞のアポトーシスが報告されており、ウイルスが感染すると個体を守るためにIFNが産生されるのだが、そのIFNは宿主細胞のアポトーシスを誘導し炎症を引き起こす原因になっている可能性が考えられた。
    また、IFN-γKOマウスはマウス肝炎ウイルス(MHV)感染に対して高感受性を示し、肝臓におけるウイルス感染の防御にIFN-γが重要な働きをしていることもわかった。

    HTLV-1トランスジェニックマウス(Tg)での関節炎発症に関与するサイトカインを同定するために、IL-1α,IL-1β,IFN-γ,TNF-α,IL-4,IL-6のそれぞれの遺伝子ノックアウトマウス(KO)とHTLV-1Tgとを交配して関節炎の発症率を比較した。IFN-γKOマウスは差がなかったが、IL-1α/βダブルKOとIL-6KOマウスでは発症が抑制されることがわかり、この二つのサイトカインが関節炎の発症に重要な役割を果たしていることが明らかとなった。TNF-αとIL-4については現在観察群を作成中である。またコラーゲン誘導関節炎においてもKOマウスを用いた同様の解析を行い、やはりIL-1とIL-6が発症に重要であることがわかった。以上のように種々のサイトカインKOマウスを用いることで、関節リウマチの発症に関与するサイトカインを同定することに成功しつつある。
    昨年度は正常マウスの骨髄細胞をHTLV-1Tgに移植すると関節炎の発症が抑制されることを明らかにしたが、今年度は高発症系のBALB/c背景のHTLV-1Tgの骨髄細胞と脾細胞を正常マウスに移植することで、発症を移入することに成功した。またHTLV-1Tgをヌードマウスの背景にしたら発症が抑制されることも見いだしたので、この関節炎の発症にはT細胞が重要であることがわかった。そこでHTLV-1TgのT細胞のVβレパートリーを調べたところ、関節局所において特定のVβが増加していることがわかった。関節で増加したT細胞が認識する抗原分子を同定するために、脾臓のT細胞をII型コラーゲンやHSP60などの存在下で培養したところ、II型コラーゲンに反応するT細胞のVβが関節T細胞のVβと一致するものがあり、関節で増加しているT細胞がII型コラーゲンを抗原として認識していることが示唆された。

    マウスにコンカナバリンA (Con A)を投与すると肝炎を誘発できることが知られており、ヒトのウイルス性肝炎のモデルとして使われている。この肝炎の発症は、T細胞依存的であり、IL-1, IL-2, IL-6, TNF-αやIFN-γなどのサイトカインの関与が示唆されている。Con A肝炎におけるIFN-γの果たす役割を明らかにするために、IFN-γ遺伝子ノックアウトマウスを作製し、病態に及ぼす影響について検討した。
    IFN-γノックアウトマウスにCon Aを投与したところ、コントロールマウスと対称的に血中のトランスアミナーゼ(GOTやGPT)レベルの上昇がみられなかった。このとき、野生型のマウスでは、肝臓においてアポトーシスが観察されたが、IFN-γノックアウトマウスではアポトーシスが抑制されていた。アポトーシスのシグナル伝達におけるリセプターの1つとして知られているFasは、in vitroにおいてIFN-γによりその発現が誘導される。Con A投与後のマウスの肝臓でFasのmRNA量を調べたところ、IFN-γノックアウトマウスにおいてFas遺伝子の発現低下が確認された。そこで、Fas遺伝子に突然変異を持つlprマウスにCon Aを投与したところ、野生型と比較してトランスアミナーゼのレベルが低かった。以上の結果から、Con A誘導肝炎において、IFN-γがFasシステムの活性化を促進し、肝臓においてアポトーシスを誘導することが明らかとなった。また、IFN-γノックアウトマウスの肝臓ではTNF-αやiNOSの発現誘導も低下していたことから、これらの因子も肝炎の発症に関与していると思われる。

    1.インターロイキン-1(IL-1)遺伝子欠損マウスの作製とその解析
    昨年度にIL-1β遺伝子欠損マウスを作製しており、今年度はIL-1α及びIL-1α/βダブル欠損マウスの作製に成功した。特にαとβ遺伝子は非常に近接して存在しているために、IL-1β遺伝子を欠損したES細胞を用いて更にIL-1α遺伝子を欠損させた後、ダブル欠損マウスを作製した。これで3種類のIL-1遺伝子欠損マウスが揃ったが、現在までのところ健康状態や生殖能力には異常は認められない。それぞれ単独にαまたはβを欠損したマウスにLPSを投与してエンドトキシンショックに対する抵抗性を調べたが、どちらのマウスも野生型と差が見られず、LPSに対する感受性はαとβがお互いに相補している可能性が示唆された。一方、テレビン油を投与して発熱を誘導する実験では、IL-1α欠損マウスとは正常マウス同様に発熱するのに対して、IL-1β欠損マウスは発熱しないことがわかり、初めてαとβに機能の違いがあることを明らかにした。現在IL-1受容体のアンタゴニスト遺伝子の欠損マウスの作製も進めており、ダブル欠損マウスも用いて、様々な炎症・免疫反応におけるIL-1の機能を明らかにしていく予定である。
    2.インターフェロン-γ(IFN-γ)のConA肝炎における役割
    マウスにConAを投与すると肝炎を誘発できることが知られており、ヒトのウイルス性肝炎のモデルとして使われている。IFN-γがこの肝炎の病態に及ぼす影響について、すでに作製したIFN-γ遺伝子欠損マウスを用いて検討した。IFN-γ欠損マウスにConAを投与したところ、肝炎の発症が抑制され、肝臓細胞のアポトーシスが抑制されていた。ConA投与後のIFN-γ欠損マウスの肝臓においてFasの発現誘導が抑えられていた。Fas遺伝子に突然変異を持つlprマウスにConAを投与したところ、同様に肝炎の発症が抑えられたことから、ConA誘導肝炎において、IFN-γがFasシステムの活性化を促進し、肝臓細胞のアポトーシスを誘導することが明らかとなった。
