    Hydrocarbons play key roles in atmospheric and biogeochemistry, the energy economy, and climate change. Most hydrocarbons form in anaerobic environments through high temperature or microbial decomposition of organic matter. Microorganisms can also eat hydrocarbons underground, preventing them from reaching the atmosphere. Using a new technique developed at the Earth-Life Science Institute (ELSI), an international team led by the Tokyo Institute of Technology professors Alexis Gilbert, Naohiro Yoshida and Yuichiro Ueno show that biological hydrocarbon degradation gives a unique biological signature. These findings could help detect subsurface biology and understand the carbon cycle and its impact on climate.

    地球生命研究所の吉田尚弘教授、丸山茂徳教授、黒川顕教授、同大学院理工学研究科の上野雄一郎准教授らの研究グループは、長野県白馬地域の温泉水が無機的に合成されたメタンガスを含むことを突き止めた。これは地球初期の生命誕生のメカニズムを解き明かすことにつながる成果である。この温泉水は、蛇紋岩と呼ばれる特殊な岩石と水が反応することによってできた強いアルカリ性の温泉で、かつ水素濃度が高い。このような温泉は、現在の地球では稀であるが、生命誕生前の初期地球ではありふれた温泉環境であったと考えられている。今回、この温泉で、今まで知られていなかった無機的な化学反応によって生命のもととなる炭化水素が合成されていることを突き止めたことから、同様の化学反応が地球の初期で有機物を作り、それが生命の誕生につながった可能性が示された。

    Abstract

    The atmosphere of Mars is mainly composed by carbon dioxide (CO2). It has been predicted that photodissociation of CO2 depletes 13C in carbon monoxide (CO). We present the carbon 13C/12C isotopic ratio in CO at 30–50 km altitude from the analysis of the solar occultation measurements taken by the instrument Nadir and Occultation for Mars Discovery on board the ExoMars Trace Gas Orbiter (ExoMars-TGO). We retrieve 12C16O, 13C16O, and 12C18O volume mixing ratios from the spectra taken at 4112–4213 cm−1, where multiple CO isotope lines with similar intensities are available. The intensities of the 12C16O lines in this spectral range are particularly sensitive to temperature, thus we derive the atmospheric temperature by retrieving CO2 density with simultaneously measured spectra at 2966–2990 cm−1. The mean δ13C value obtained from the 13C16O/12C16O ratios is −263‰, and the standard deviation and standard error of the mean are 132‰ and 4‰, respectively. The relatively large standard deviation is due to the strong temperature dependences in the 12C16O lines. We also examine the 13C16O/12C18O ratio, whose lines are less sensitive to temperature. The mean δ value obtained with 12C18O instead of 12C16O is −82‰ with smaller standard deviation, 60‰. These results suggest that CO is depleted in 13C when compared to CO2 in the Martian atmosphere as measured by the Curiosity rover. This depletion of 13C in CO is consistent with the CO2 photolysis-induced fractionation, which might support a CO-based photochemical origin of organics in Martian sediments.

    We present a novel technique to analyze d34S, d18O, D'33S, and 34S-18OO double-substitution (D'34S18O) in sulfate by measuring SO2F2 gas with a high-resolution isotope ratio mass spectrometer. The external reproducibility (1 sem, all numbers in per mille) is ± 0.09 (d34S), ± 0.2 (d18O), ± 0.02 (D'33S), and ± 0.08 (D'34S18O) for repeated analyses of our in-house reference sulfate. Furthermore, accuracy of our analytical approach is demonstrated by the analysis of sulfate with distinct negative D'34S18O, produced by mixing 34S and 18OO labeled sulfate solutions.Sulfates from laboratory experiments as well as from different natural environments show a total D'34S18O range of approx. 1.5 per mille and indicate that sulfates that form in low temperature surface environments have distinctively higher D'34S18O than sulfates which form at elevated temperature at hydrothermal conditions.Therefore, D'34S18O may be a promising tracer for sulfate sources.

    Sulfur isotopes have been used to understand the sulfur cycle throughout the Earth's history. The apparent isotopic fractionation between sulfides and sulfates is believed to have increased in response to global oxygenation due in part to changes in the fractionation factors for microbial sulfate reduction. However, accurate estimates of the fractionation factors are difficult due to the lack of sulfate in the Precambrian period, and there are few examples of fractionation factors for 33S and 36S using Proterozoic sedimentary rocks. In this study, we established an analytical method to estimate fractionation factors for microbial sulfate reduction (34epsilon), mass-dependent exponents (33lambda and 36lambda), and initial d34S for sulfate (d34Si) using only pyrite isotopic ratios without sulfate analysis. This method was validated by using isotopic ratios of modern sedimentary sulfides. The estimated d34Si are consistent with measured sulfate values. Re-evaluation of multiple sulfur isotope records of the Proterozoic using this method suggests that the fractionation factors in the Proterozoic are significantly smaller than that in the Phanerozoic and increased from the Ediacaran.

    Minerals and rocks exhibit various isotope compositions depending on their origins and histories. In interpreting their isotopic variations, the equilibrium isotope fractionation factor is a key because it depends on the environment parameters such as temperature. Recent studies have shown that the effect of pressure on the isotope fractionation, which was considered negligible compared to temperature, is significant under the conditions of the Earth's interior. In this article we review recent advances in experimental studies to determine the isotope fractionation of iron and hydrogen at high pressure over several GPa, discussing their issues and future perspectives.

    Photochemistry of sulfur dioxide yields anomalous sulfur isotope fractionation under anoxic atmosphere. The isotope anomaly can be transported into sedimentary sulfide and sulfate, thus is useful to trace atmospheric chemistry before the Great Oxidation Event (GOE) at about 2.3 billion years ago. Based on the isotopic analysis of Archean sedimentary rocks, the δ36S/δ33S trend seems to have changed through time, though the factors controlling the δ36S/δ33S signal are still uncertain. Our new laboratory experiments have shown that the δ36S/δ33S trend are mainly controlled by pSO2 and redox state of the atmosphere even without molecular oxygen. Thus, the observed change of the sulfur isotopic signal may reflect volcanic SO2 flux as well as redox fluctuation before GOE. Furthermore, our results indicate that the Archean atmosphere is not the pure CO2, but should be more reducing to explaining the observed δ36S/δ33S trend in the Archean rocks.

    地球史を通した大気組成の変化は依然定量的に見積もられていないが、太古代堆積岩に硫黄の非質量依存同位体分別が発見された事で大気酸素分圧は23から20億年前を境に急上昇したと考えられるようになった(Farquhar et al., 2000; Holland, 2006)。この同位体異常の原因は正確には不明であるが、紫外線によるSO2の光化学反応の際に非質量依存性同位体分別が引き起こされることは実験的に確かめられている(Farquhar et al., 2001)。PavlovとKasting (2001)による大気モデル計算によれば、気体硫黄分子の光解離が10-5PAL以下の貧酸素大気中で生じると硫酸エアロゾルだけでなくより還元的な元素硫黄が生成するため、同位体異常は化学形態の異なる二種のエアロゾルによって大気から地表まで運ばれうる一方、より高酸素分圧下では火山活動等により大気に流入した硫黄は最終的にほぼ全て硫酸エアロゾルとなるため、光解離による同位体分別効果は相殺され地表に同位体異常が保存される事は無いと説明されている。こうした研究をさらに発展させ、大気組成を制約してゆく試みが注目されているが、地質記録から推定される過去のエアロゾルの四種安定同位体組成と実験およびモデル計算から算出されたものとの間には依然大きな隔たりがある(Ohmoto et al., 2006; Bao et al., 2007; Ueno et al., in review)。不一致の最も大きな原因の一つは光解離による同位体効果が正確に見積もられていない事にある(Farquhar et al., 2007)。そこで我々は分光学的な実験によりSO2同位体分子種それぞれの紫外吸収スペクトルを実測し、それらの吸収断面積を求めた(Danielache et al., in press)。さらにこの同位体分子種ごとの吸収スペクトルを用い、光源のスペクトルを仮定することで光解離時の同位体効果を計算により求めた。この結果、太陽スペクトルによるSO2光解離は大きな同位体異常を作りうる事が明らかになった。またその同位体分別係数は大気中のO2およびO3濃度のみならずCO2やSO2それ自身の分圧によっても敏感に変化する事が示唆された。これらの結果と地質記録を比較するとSO2光解離時の同位体効果は原生代最初期の同位体異常(Kaufman et al., 2007)を説明できるものの、それ以前(太古代)の地質データとは一致しない。このことは太古代/原生代境界(約25億年前)を挟んで大気組成が変化したことを示唆している。またその変化は酸素分圧上昇ではなく、それ以外の要因、例えばCO2分圧の減少等に由来する可能性がある。

    Farquharら(2000)は地層中に含まれる硫化鉱物が20億年以前のものに限って非質量依存性同位体分別の特徴をもつこと発見し、酸素濃度の低い初期地球環境では光化学反応を被った大気由来の硫黄が海洋の硫黄循環に大きく寄与した事が示唆された。それ以来硫黄の四種安定同位体(32S, 33S, 34S, および36S)はフッ化法により精力的に分析が行われるようになった。四種同位体の解析からは従来の34S/32S比からは得られない新たな情報を読み取る事が出来る。講演では四種同位体分析の最近の研究についてレビューし、演者らが行っている過去および現在の熱水系での硫黄循環の解析について紹介する。

    Active submarine hydrothermal activities have been located in Valu Fa Ridge, in the Lau Backarc Basin. In these sites, abundanta sulfide-sulfate deposits were observed associated with active venting of hydrothermal fluids. Among sulfide deposits, Cu-rich sulfides were associted with higher temperature venting (>300C), while Zn-rich sulfides were associted with moderate temperature (250-300C). Amond old deposits, significant amount of barite was notified. We will discuss relationship with fluid chemistry, which showed extremely low acid due to magmatic volatile involvement.

    約8.5億年前の中央オーストラリア、Bitter Spritngs Formationから産出された黒色チャート中の微化石は3次元的に構造が保存されており、光学顕微鏡による形態観察からシアノバクテリアの化石と同定されている。また、顕微赤外分光法による分析から、Bitter Springs微化石を構成する有機物中に鎖状炭化水素のC-H結合や芳香族のC-C結合が存在することが示唆されている。本研究では、Bitter Springs微化石から検出された有機物の起源を調べるために、現生のシアノバクテリアの珪化・加熱実験を行い、シアノバクテリア細胞中の有機物の加熱変化を顕微赤外分光法で調べた。

    原生代から顕生代にかけて、生物相が、単細胞生物から多細胞生物へと変化したことが知られ、当時の地球表層環境を知ることは、生命進化の解明につながる。堆積岩には、堆積当時の地球表層環境の特徴が化学組成や同位体比という形で記録されている。特に堆積岩中の有機分子組成及び分子別炭素同位体比は、当時の炭素循環の情報を記録している。原生代の堆積岩中の炭化水素組成及び分子別炭素同位体比から、当時の水柱の炭素循環は顕生代と異なる可能性が指摘された。しかし、原生代の堆積岩中有機分子の分子別炭素同位体比分析の報告例は少なく、当時の炭素循環について断片的にしか解明されていない。本発表では、未だ報告例のない南中国に産する原生代/顕生代境界層堆積岩中の有機分子組成及び分子別炭素同位体比の分析結果について報告する。

    Abundant organic matter (kerogen) was identified in 3.5 Ga hydrothermal silica dikes from the North Pole area in the Pilbara craton, Western Australia. The silica dikes developed in the uppermost 1000 m of the ancient oceanic crust. Thus, they would have been deposited in the 3.5 Ga sub-seafloor hydrothermal system. The carbon and nitrogen isotopic compositions of the kerogen were analyzed in this study. Their highly 13C-depleted isotopic compositions (δ13C=-38 to-33%0) strongly suggest that they are originally derived from biologically produced organic matter. The remarkable similarity of the δ13C values between the kerogen and modern hydrothermal vent organisms may suggest that the kerogen was derived from chemoautotrophic organisms. This idea is also consistent with their nitrogen isotopic compositions (δ15N =-4 to + 4‰). The silica dikes consist mainly of fine-grained silica with minor pyrite and sphalerite. These mineral assemblages indicate that the silica dike was deposited from relatively low-temperature (probably less than 150 °C) reducing hydrothermal fluid. Thus, anaerobic thermophilic/hyperthermophilic organisms could have survived in the hydrothermal fluid, which formed the silica dikes. Therefore, it is plausible that a chemoautotrophic-based biosphere (possibly methanogenesis) probably existed in the Early Archean sub-seafloor hydrothermal system.

    Hydrogen isotopic compositions of Archean basalts were determined by TCEA-IRMS technique. The analysis of more than 200 samples from 3.2 Ga Cleaverville Formation showed that dD values of hydrous minerals are clearly correlated with metamorphic grade which reflect ocean-floor metamorphism at that time based on metamorphic mineral assemblages. Also, the dD values are correlated with water content of the basaltic rocks as observed in modern seafloor basalts. These observations all indicate that hydrous minerals in the Cleaverville basalts preserve the isotopic composition of Archean seawater. The estimated dD value of the 3.2 billion-years-old seawater is -21±5‰, indicating deuterium content of Earth's seawater increased through the history of the Earth, potentially owing to hydrogen escape into space. Furthermore, we found igneous amphibole from the most D-depleted sample, suggesting not only Archean seawater but also Archean mantle was depleted in deuterium.

    A nobel micro-fluorination method has been developed for rapid high precision quadruple sulfur isotope analysis (Ueno et al., 2015 Chemical Geology). In contrast to the previous F2 method, reaction with solid CoF3 reagent allows us to fluorinate sulfide samples into SF6 gas within several second. Futhermore, the analytical protocols were successfully optimized for analyzing small samples down to 100 nmolS with the highest precision that has made by conventional F2 method. In addition, the same technique was shown to be applicable to fluorinate pyrites directly.

    地層に記録された同位体情報から古大気組成を復元する目的のため、大気化学反応を模擬した光解離実験を行った。本年の実験は、まず分光器を備えた光化学反応セルを作成および改良し、波長150~300nm領域を0.1nm分解能で紫外スペクトルを取りつつ反応を行えるシステムが完成した。また生成物の回収ラインとガスクロマトグラフ質量分析計を導入し、生成物の同定および定量分析が可能になった。このシステムを用いて模擬古大気中でのSO2光解離反応を行ったところ、CO/CO2比の高い雰囲気下では乖離したSO2がCOSに変換されることが実験的に確かめられた。これは還元大気中で硫黄の化学種はCOSの状態で存在したとする予測を支持する結果である。また、SO2分子の吸収スペクトルについて、これまでの同位体種に加えて36SO2のスペクトル測定を行うことに初めて成功し、データ解析を行っている。これにより36S/33S比が古大気組成の制約にとって最も強力な指標となることが期待される。また、インド・ダルワール地塊での地質調査および試料採取を行った。その結果、大気・気候変動期にあたる30-23億年前の層序を一部復元した。今後採取試料の年代決定・同位体分析に移行する。一方、1次元大気化学モデルは従来数値モデルの改良では不適当と判断し、反応の数を減らすことで根本から組み直す選択をし、コード開発を進めている。

    We completed the experiments for effect of H2-consuming and. producing metabolisms on stable hydrogen isotopic equilibrium, effect of S0-reducing metabolisms on multiple sulfur isotopic equilibrium and fractionation and effect of sulfur-disproportionating metabolisms on multiple sulfur isotopic fractionation. All the data have provided very new insights into understanding the systematics of stable isotopic fractionation and equilibrate effects during metabolisms of extremophiles. Several research papers are now under preparation.

    Field works in Kerala, southern India and Adirondack Mountains, NY were done. Petrographic observations of samples from these localities were done. It has been confirmed that CO_2 fluid inclusions in garnet and zircon were common in these lower-crust rocks. A numerical model of the CO_2 cycle connecting the solid Earth and the surface environment was constructed. It has been demonstrated that an expected decarbonation event in the Proterozoic had extensive effect to the Earth's surface temperature. A new method to estimate a thermal structure of the subduction zone was developed. These results were presented in 6 talks and published as 13 papers in academic journals.

    Changes in atmospheric concentrations of greenhouse gases (GHGs) and aerosols are drivers of climate change. N_2O and CH_4 are important GHGs and H_2S is related to aerosol formation. To develop the source-sink analyses of these gases using their isotopomer signals, we quantified relationships between some microbial processes and their isotopomer signals in pure culture and co-culture conditions.

    直接・間接的に地球の放射エネルギー収支に寄与するメタン(CH_4)と硫化カルボニル(COS)は、海洋表層-大気下層において生物活動に起因して生成するが、好気的CH_4生成の有無とその機構、COSの物質収支については不明な点が多く残されている。本研究では安定同位体比を利用して、植物プランクトンの培養実験による酸化的CH_4生成の検証と生成機構の解明、およびCOSの高感度・高精度硫黄同位体比測定法の開発と大気・海水試料への適用を試み、本年度は以下の成果が得られた。
    1. 研究協力者(学外研究者および大学院生)の協力も得て木崎湖、深見池、諏訪湖で植物プランクトンを採取し、無菌的に室温自然光条件下で培養した。CH_4の生成を確認するとともにその炭素同位体比を測定した。前年度北太平洋において採取した海水試料の分析結果の解析を進め、この海域で観測された亜表層の過飽和溶存メタンは微小粒子内の還元環境下でメタン生成菌によって生成されたものと推定した。今後、種々のプランクトンについて純粋培養して生物種、温度、光条件などに対する依存性についての研究を展開することにより、酸化的CH_4生成の重要性が明らかになると期待される。
    2. フラグメンテーションを利用したCOSの高精度硫黄同位体比測定法の開発を進めた。研究協力者(大学院生)の協力を得て、COS硫黄同位体比の標準試料の国際標準物質(CDT)に対する較正を行うために、COSの標準ガスを精製後、SF6に変換して4種硫黄同位体比を測定した。並行して大気・海水中に存在する微量COSを抽出、濃縮、分離するための前処理装置を試作し、GC/FPDと組み合わせて基礎実験を行った。今後、分析精度の向上、海水試料の分析法開発に取り組むことにより海洋表層-大気下層におけるCOSの挙動解明が期待される。

    昨年度確立した流体包有物中メタンガスの同位体分析法を用い、本年度は太古代熱水岩脈中試料の分析を行った。その結果、35億年前の海底を循環した熱水中にメタンガスが存在し、それらは有機物の熱分解過程とメタン生成菌活動の二つの起源を有することを示した。この結果は約35億年当時、メタン生成菌が活動したことを示す世界初の証拠と考えられる。その成果は今夏行われた進化学会にて公表された。
    さらに太古代硫化鉱物の硫黄同位体分析のために抽出した硫黄試料については、カーネギー研究機構において分析を行い、約35億年前の熱水岩脈および熱水性堆積物試料が硫黄同位体比異常を有することを突き止めた。またこの同位体組成は大気・海洋由来の成分と熱水・マグマ起源の成分の混合によって決まることを示し、当時の硫黄循環を知る上で重要な知見を加える事が出来た。また、より34Sに乏しい黄鉄鉱が硫酸塩岩試料中に存在することを突き止めた。これは約35億年当時の硫酸還元菌活動を示唆するため、今後更に研究を発展させる必要がある。
