    Abstract

    In drug development, systematically characterizing a compound’s mechanism of action (MoA), including its direct targets and effector proteins, is crucial yet challenging. Network-based approaches, unlike those focused solely on direct targets, effectively detect a wide range of cellular responses elicited by compounds. This study applied protein covariation network analysis, leveraging quantitative, morphological, and localization features from immunostained microscopic images, to elucidate the MoA of AX-53802, a novel ferroptosis inducer. From the candidate targets extracted through network analysis, GPX4 was verified as the direct target by validation experiments. Additionally, aggregates involving GPX4, TfR1, and F-actin were observed alongside iron reduction, suggesting a ferroptosis defense mechanism. Furthermore, combination therapies targeting GPX4 and FAK/Src were found to enhance cancer cell death, and MDM2, ezrin, and cortactin were identified as potential ferroptosis inhibitor targets. These findings highlight the effectiveness of network-based approaches in uncovering a compound’s MoA and developing combination therapies for cancer.

    "Trim-Away" technology enables rapid degradation of endogenous proteins without prior modification of protein-coding genes or mRNAs through delivery of antibodies that target proteins of interest. Although this approach can be readily applied to almost any cytosolic protein, strategies for cytosolic antibody delivery have been limited to microinjection or electroporation, which require skill-dependent operation or specialized equipment. Thus, the development of antibody delivery methods that are convenient, scalable, and preferably do not require detachment of adherent cells is required to extend the versatility of the Trim-Away method. Here, we developed a cell resealing technique optimized for Trim-Away degradation, which uses the pore-forming toxin streptolysin O (SLO) to permeabilize the cell membrane and delivered the antibodies of interest into HEK293T, HeLa, and HK-2 cell lines. We demonstrated the ability of Trim-Away protein degradation using IKKα and mTOR as targets, and we showed the availability of the developed system in antibody screening for the Trim-Away method. Furthermore, we effectively coupled Trim-Away with cyclic immunofluorescence and microscopic image-based analysis, which enables single-cell multiplexed imaging analysis. Taking advantage of this new analysis strategy, we were able to compensate for low signal-to-noise due to cell-to-cell variation, which occurs in the Trim-Away method because of the heterogenous contents of the introduced antibody, target protein, and TRIM21 in individual cells. Therefore, the reported cell resealing technique coupled with microscopic image analysis enables Trim-Away users to elucidate target protein function and the effects of target protein degradation on various cellular functions in a more quantitative and precise manner.

    p52SHC (SHC) and GRB2 are adaptor proteins involved in the RAS/MAPK (ERK) pathway mediating signals from cell-surface receptors to various cytoplasmic proteins. To further examine their roles in signal transduction, we studied the translocation of fluorescently-labeled SHC and GRB2 to the cell surface, caused by the activation of ERBB receptors by heregulin (HRG). We simultaneously evaluated activated ERK translocation to the nucleus. Unexpectedly, the translocation dynamics of SHC were sustained when those of GRB2 were transient. The sustained localization of SHC positively correlated with the sustained nuclear localization of ERK, which became more transient after SHC knockdown. SHC-mediated PI3K activation was required to maintain the sustainability of the ERK translocation regulating MEK but not RAF. In cells overexpressing ERBB1, SHC translocation became transient, and the HRG-induced cell fate shifted from a differentiation to a proliferation bias. Our results indicate that SHC and GRB2 functions are not redundant, but that SHC plays the critical role in the temporal regulation of ERK activation.

    MicroRNAs (miRNAs) are cargo carried by extracellular vesicles (EVs) and are associated with cell-cell interactions. The response to the cellular environment, such as disease states, genetic/metabolic changes, or differences in cell type, highly regulates cargo sorting to EVs. However, morphological features during EV formation and secretion involving miRNA loading are unknown. This study developed a new method of EV loading using cell resealing and reconstituted the elementary miRNA-loading processes. Morphology, secretory response, and cellular uptake ability of EVs obtained from intact and resealed HeLa cells were comparable. Exogenously added soluble factors were introduced into multivesicular endosomes (MVEs) and their subsequent secretion to the extracellular region occurred in resealed HeLa cells. In addition, miRNA transport to MVEs and miRNA encapsulation to EVs followed a distinct pathway regulated by RNA-binding proteins, such as Argonaute and Y-box binding protein 1, depending on miRNA types. Our cell-resealing system can analyze disease-specific EVs derived from disease model cells, where pathological cytosol is introduced into cells. Thus, EV formation in resealed cells can be used not only to create a reconstitution system to give mechanistic insight into EV encapsulation but also for applications such as loading various molecules into EVs and identifying disease-specific EV markers.

    RTK-RAS-MAPK systems are major signaling pathways for cell fate decisions. Among the several RTK species, it is known that the transient activation of ERK (MAPK) stimulates cell proliferation, whereas its sustained activation induces cell differentiation. In both instances however, RAS activation is transient, suggesting that the strict temporal regulation of its activity is critical in normal cells. RAS on the cytoplasmic side of the plasma membrane is activated by SOS through the recruitment of GRB2/SOS complex to the RTKs that are phosphorylated after stimulation with growth factors. The adaptor protein GRB2 recognizes phospho-RTKs both directly and indirectly via another adaptor protein, SHC. We here studied the regulation of GRB2 recruitment under the SHC pathway using single-molecule imaging and fluorescence correlation spectroscopy in living cells. We stimulated MCF7 cells with a differentiation factor, heregulin, and observed the translocation, complex formation, and phosphorylation of cell signaling molecules including GRB2 and SHC. Our results suggest a biphasic regulation of the GRB2/SOS-RAS pathway by SHC: At the early stage (<10 min) of stimulation, SHC increased the amplitude of RAS activity by increasing the association sites for the GRB2/SOS complex on the plasma membrane. At the later stage however, SHC suppressed RAS activation and sequestered GRB2 molecules from the membrane through the complex formation in the cytoplasm. The latter mechanism functions additively to other mechanisms of negative feedback regulation of RAS from MEK and/or ERK to complete the transient activation dynamics of RAS.

    Phosphatidylinositol-3-phosphate (PI3P) is a lipid that accumulates in the early endosomal membrane, and acts as a scaffold to recruit proteins that contain a PI3P-binding domain, such as the FYVE domain. In this study, we examined the effect of PI3P depletion on the insulin response in rat hepatoma-derived H4IIEC3 cells. We found that insulin treatment induced the transient formation of an actin domain structure, a mesh-like tangled network of actin filaments where phosphorylated Akt, endosomal proteins, and PI3P accumulated. Actin domain formation was repressed by the depletion of PI3P by SAR405, an inhibitor of the class III PI3 kinase, Vps34, by the inhibition of PI3P function by the competitive binding of an excess amount of GST-fused 2xFYVE protein to intracellular PI3P, and by the use of diabetic model cells, in which PI3P was depleted. SAR405 did not affect the phosphorylation level of Akt, and the transcriptional regulation of gluconeogenic and cholesterol synthetic genes after insulin treatment. Interestingly, insulin-induced DNA synthesis was specifically inhibited by SAR405, cytochalasin B, and also in diabetic model cells. These results suggest that PI3P is required for the formation of actin domains, which affected a signaling pathway downstream of Akt associated with DNA synthesis in H4IIEC3 cells.

    Morphology of Golgi apparatus changes frequently and diversely depending on various cellular conditions and these changes correlate with the balance between membrane inflow and outflow at the Golgi via vesicular transports. In a previous study, we introduced a semi-intact cell system suitable for the reconstitution of morphological changes that organelles undergo as well as the vesicular transport between them. Semi-intact cells are cells that have undergone plasma membrane permeabilization by the cholesterol-dependent pore-forming cytolysin, streptolysin O (SLO). Permeabilization enables the introduction of various molecules into the cells, as well as the substitution of the original cytosol with an exogenously made cytosol prepared from cells in various stages of cell cycle, differentiation, and disease progression. Coupled with a green fluorescent protein(GFP)-visualization technique, this cell-based system enables the analysis of the molecular mechanisms underlying biological processes that are highly dependent on the integrity of the intracellular architecture. In this chapter, we present a variety of reconstitution assays concerning biological reactions pertaining to the Golgi apparatus.

    Cell-based assays have become increasingly important in the preclinical studies for biopharmaceutical products such as specialty peptides, which are of interest owing to their high substrate specificity. However, many of the latter are membrane impermeable and must be physically introduced into cells to evaluate their intracellular activities. We previously developed a "cell-resealing technique" that exploited the temperature-dependent pore-forming activity of the streptococcal toxin, streptolysin O (SLO), that enabled us to introduce various molecules into cells for evaluation of their intracellular activities. In this study, we report a new cell resealing method, the listeriolysin O (LLO)-mediated resealing method, to deliver mid-sized, membrane-impermeable biopharmaceuticals into cells. We found that LLO-type resealing required no exogenous cytosol to repair the injured cell membrane and allowed the specific entry of mid-sized molecules into cells. We use this method to introduce either a membrane-impermeable, small compound (8-OH-cAMP) or specialty peptide (Akt-in), and demonstrated PKA activation or Akt inhibition, respectively. Collectively, the LLO-type resealing method is a user-friendly and reproducible intracellular delivery system for mid-sized membrane-impermeable molecules into cells and for evaluating their intracellular activities.

    Apoptotic death of pancreatic β cells is a major cause of type 2 diabetes mellitus (T2D) progression. Two isoforms of pyruvate kinase, PKM1 and PKM2, have been reported to participate in cell death in several cell types; however, little is known about their causal pathways in pancreatic β-cell death. We examined whether the suppression of PKM1 or PKM2 affects endoplasmic reticulum (ER) stress-induced apoptosis in a pancreatic β-cell line, MIN6, and Beta-TC-6 and found that knockdown of PKM1, but not of PKM2, leads to the induction of ER stress-induced apoptosis in these cells. We also investigated the mechanism by which PKM1 inhibits ER stress-induced apoptosis. We confirmed that PKM1 interacts with A-Raf, an upstream regulator of the MEK/ERK pathway, and that this interaction contributes to MEK1 phosphorylation by A-Raf. PKM1 knockdown suppresses the phosphorylation of MEK, ERK, and caspase-9 (Thr125), which is phosphorylated by the MEK/ERK pathway, thereby inhibiting the cleavage and activation of caspase-9. Thus, PKM1 knockdown activates the caspase-9/caspase-3 pathway under ER stress conditions and leads to apoptosis.

    Cancer cells are under chronic endoplasmic reticulum (ER) stress due to hypoxia, low levels of nutrients, and a high metabolic demand for proliferation. To survive, they constitutively activate the unfolded protein response (UPR). The inositol-requiring protein 1 (IRE1) and protein kinase RNA-like ER kinase (PERK) signaling branches of the UPR have been shown to have cytoprotective roles in cancer cells. UPR-induced autophagy is another prosurvival strategy of cancer cells, possibly to remove misfolded proteins and supply nutrients. However, the mechanisms by which cancer cells exploit the UPR and autophagy machinery to promote survival and the molecules that are essential for these processes remain to be elucidated. Recently, a multipass membrane protein, Yip1A, was shown to function in the activation of IRE1 and in UPR-induced autophagy. In the present study, we explored the possible role of Yip1A in activation of the UPR by cancer cells for their survival, and found that depletion of Yip1A by RNA interference (RNAi) induced apoptotic cell death in HeLa and CaSki cervical cancer cells. Intriguingly, Yip1A was found to activate the IRE1 and PERK pathways of the UPR constitutively in HeLa and CaSki cells. Yip1A mediated the phosphorylation of IRE1 and also engaged in the transcription of PERK. The activation of these signaling pathways upregulated the expression of anti-apoptotic proteins and autophagy-related proteins. These events might enhance resistance to apoptosis and promote cytoprotective autophagy in HeLa and CaSki cells. The present study is the first to uncover a key prosurvival modulator, Yip1A, which coordinates IRE1 signaling with PERK signaling to support the survival of HeLa and CaSki cervical cancer cells.

    RalGDS is one of the Ras effectors and functions as a guanine nucleotide exchange factor for the small G-protein, Ral, which regulates membrane trafficking and cytoskeletal remodeling. The translocation of RalGDS from the cytoplasm to the plasma membrane is required for Ral activation. In this study, to understand the mechanism of Ras-Ral signaling we performed a single-molecule fluorescence analysis of RalGDS and its functional domains (RBD and REMCDC) on the plasma membranes of living HeLa cells. Increased molecular density of RalGDS and RBD, but not REMCDC, was observed on the plasma membrane after EGF stimulation of the cells to induce Ras activation, suggesting that the translocation of RalGDS involves an interaction between the GTP-bound active form of Ras and the RBD of RalGDS. Whereas the RBD played an important role in increasing the association rate constant between RalGDS and the plasma membrane, the REMCDC domain affected the dissociation rate constant from the membrane, which decreased after Ras activation or the hyperexpression of Ral. The Y64 residue of Ras and clusters of RalGDS molecules were involved in this reduction. From these findings, we infer that Ras activation not merely increases the cell-surface density of RalGDS, but actively stimulates the RalGDS-Ral interaction through a structural change in RalGDS and/or the accumulation of Ral, as well as the GTP-Ras/RalGDS clusters, to induce the full activation of Ral.

    Forkhead box O1 (FOXO1) is an important target for insulin. It is widely accepted that insulin-induced phosphorylation of FOXO1 by Akt leads to its nuclear exclusion and results in the inhibition of FOXO1-mediated transcription of the gluconeogenic gene phosphoenolpyruvate carboxykinase 1 (PCK1) in hepatocytes. However, many results that contradict this model have accumulated. Here, we provide a new mechanism for insulin-dependent repression of FOXO1-mediated transcription. We showed insulin-induced translocation of endogenous Ser256-phosphorylated FOXO1, which is essential for regulation of FOXO1-mediated transcription, from nuclear speckles to the nuclear periphery. This insulin-dependent translocation of FOXO1 regulated transcriptional repression of PCK1 concomitant with the formation of the FOXO1-euchromatic histone-lysine N-methyltransferase2 (EHMT2) complex and histone modifications of the PCK1 promoter region. Notably, our results suggest that FOXO1 uses nucleoporin 98 kDa NUP98 for this transcriptional regulation. These results provide a new insight into various FOXO1-mediated transcriptional regulation and FOXO1-mediated essential biological pathways.

    Rab2A, a small GTPase localizing to the endoplasmic reticulum (ER)-Golgi intermediate compartment (ERGIC), regulates COPI-dependent vesicular transport from the ERGIC. Rab2A knockdown inhibited glucose-stimulated insulin secretion and concomitantly enlarged the ERGIC in insulin-secreting cells. Large aggregates of polyubiquitinated proinsulin accumulated in the cytoplasmic vicinity of a unique large spheroidal ERGIC, designated the LUb-ERGIC. Well-known components of ER-associated degradation (ERAD) also accumulated at the LUb-ERGIC, creating a suitable site for ERAD-mediated protein quality control. Moreover, chronically high glucose levels, which induced the enlargement of the LUb-ERGIC and ubiquitinated protein aggregates, impaired Rab2A activity by promoting dissociation from its effector, glyceraldehyde-3-phosphate dehydrogenase (GAPDH), in response to poly (ADP-ribosyl)ation of GAPDH. The inactivation of Rab2A relieved glucose-induced ER stress and inhibited ER stress-induced apoptosis. Collectively, these results suggest that Rab2A is a pivotal switch that controls whether insulin should be secreted or degraded at the LUb-ERGIC and Rab2A inactivation ensures alleviation of ER stress and cell survival under chronic glucotoxicity.

    The ER-Golgi intermediate compartment (ERGIC) is an organelle through which cargo proteins pass and are being transferred by either anterograde or retrograde transport between the endoplasmic reticulum (ER) and the Golgi apparatus. We examined the effect of 80 different kinase inhibitors on ERGIC morphology and found that rottlerin, a PKCδ inhibitor, induced the dispersion of the perinuclear ERGIC into punctate structures. Rottlerin also delayed anterograde transport of vesicular stomatitis virus G protein (VSVG) from the ER to the Golgi and retrograde transport of cholera toxin from cell surface to the ER via the Golgi. RNA interference revealed that knockdown of PKCδ or ε resulted in the dispersion of the ERGIC, but unexpectedly did not inhibit VSVG and cholera toxin transport. We also found that rottlerin depolarized the mitochondrial membrane potential, as does carbonylcyanide-p-trifluoromethoxyphenylhydrazone (FCCP), an uncoupler, and demonstrated that a decrease in the intracellular adenosine triphosphate (ATP) levels by rottlerin might underlie the block in transports. These results suggest that PKCδ and ε specifically regulate the morphology of the ERGIC and that the maintenance of ERGIC structure is not necessarily required for anterograde and retrograde transports.

    Phosphatidylinositol-3-phosphate (PI3P) is a lipid that is enriched specifically in early endosomes. Given that early endosomes containing PI3P act as a microdomain to recruit proteins that contain a PI3P-binding domain (FYVE domain), the equilibrium between the production and degradation of PI3P influences a variety of processes, including endocytosis and signal transduction via endosomes. In the study reported herein, we have developed a novel analytical method to quantify the amount of PI3P in endosomes by introducing a GST-2xFYVE protein probe into semi-intact cells. The GST-2xFYVE probe was targeted specifically to intracellular PI3P-containing endosomes, which retained their small punctate structure, and allowed the semi-quantitative measurement of intracellular PI3P. Using the method, we found that treatment of HeLa cells with H(2)O(2) decreased the amount of PI3P in endosomes in a p38 MAPK-dependent manner. In addition, H(2)O(2) treatment delayed transport through various endocytic pathways, especially post-early endosome transport; the retrograde transport of cholera toxin was especially dependent on the amount of PI3P in endosomes.     Membrane trafficking is an important cellular process that enables the precise localization of membrane proteins. The disturbance of membrane trafficking results in various disease states. To explore systematically the defects in trafficking pathways that cause these disturbances or disease states, we developed an automated high-throughput fluorescence-based imaging system and carried out visual screening for kinase-regulated trafficking pathways of the cation-independent mannose 6-phosphate receptor (CI-M6PR) in HeLa cells. As the result of our visual screening, which examined the effect of kinase inhibitors and a kinase siRNA library, we identified five kinases (CDC42BPB, PRKACA, PRKACG, GSK3 beta and CSNK2A1) that regulate CI-M6PR trafficking. Moreover, we focused on Alzheimer's disease (AD) to study the relationship between the five kinases and a disease state. Notably, two trafficking pathways, which were regulated by PRKACG and GSK3 beta, respectively, induced high levels of secretion of Abeta, the hallmark of AD. In addition, we found that the modulation of GSK3 beta activity affected the microtubule plus end tracking function of cytoplasmic linker protein-associating protein 2 and resulted in the perturbation of BACE1 localization/trafficking and extensive A beta secretion. Our systems provide new approaches for the analysis of spatially-regulated membrane trafficking and related disease states.

    Recent advances in microscopic techniques have shed light on the roles of specific subcellular structures in the regulation of gene expression. One such structure is the stress granule (SG), which is engaged in stress-triggered translational arrest by sequestering pre-initiation complexes of translation. Recent studies revealed the spatial, compositional, and functional linkage of the SG to the processing body (P-body), another cytoplasmic structure that has been implicated in mRNA degradation and siRNA- or miRNA-mediated gene silencing. In this study, we report that PCBP2, a facilitator of IRES (Internal Ribosomal Entry Site)-mediated translation, is a novel constituent of the SG and P-body. Immunofluorescence studies revealed that while PCBP2 is diffusely distributed throughout the nucleoplasm and the cytoplasm, the protein is enriched in a subset of P-bodies under normal conditions. Upon exposure to heat and arsenic stress, PCBP2 became predominantly accumulated at the SG, but was still present in Dcp1a-positive P-bodies. Live-cell imaging revealed the dynamic association of PCBP2-enriched P-bodies and the SG, and FRAP experiments demonstrated that PCBP2 actively moves in and out of the SG and P-body. Taken together, these results suggest that PCBP2 shuttles between the cytoplasm and the two structures under stress. We propose that PCBP2 may be involved in stress-induced remodeling of mRNP complexes and that it may also play a role in the rapid transition of certain silenced mRNAs into a translationally active state. Additionally, given the property of PCBP2 as a nuclear-cytoplasmic shuttling protein, PCBP2 may play a role in directly targeting nascent mRNPs to specific P-bodies for storage.

    The mRNA-binding protein CUGBP-1 is a multi-faceted factor, involved in a wide range of biological processes including splicing, translation initiation and mRNA degradation. Here we show that CUGBP-1 is a novel constituent of stress granule (SG), the translational silencing machinery assembled in response to environmental stress. CUGBP-1 was rapidly routed to SGs upon exposure to a variety of environmental stress, and actively shuttles between the nucleus and SGs. The linker domain located between the second and third RNA recognition motifs (RRMs) was found to be essential for the recruitment of CUGBP-1 to SGs. Importantly, we discovered that the linker domain is also required to direct CUGBP-1 to another subcellular structure, perinucleolar compartment (PNC). These results demonstrate the dynamic behavior of CUGBP-1 during stress response and that the linker region, in concert with RRMs, plays a significant role in defining its subcellular localization and dynamics.

    Newly synthesized secretory and transmembrane proteins are folded and assembled in the endoplasmic reticulum (ER) where an efficient quality control system operates so that only correctly folded molecules are allowed to move along the secretory pathway. The productive folding process in the ER has been thought to be supported by the unfolded protein response (UPR), which is activated by the accumulation of unfolded proteins in the ER. However, a dilemma has emerged; activation of ATF6, a key regulator of mammalian UPR, requires intracellular transport from the ER to the Golgi apparatus. This suggests that unfolded proteins might be leaked from the ER together with ATF6 in response to ER stress, exhibiting proteotoxicity in the secretory pathway. We show here that ATF6 and correctly folded proteins are transported to the Golgi apparatus via the same route and by the same mechanism under conditions of ER stress, whereas unfolded proteins are retained in the ER. Thus, activation of the UPR is compatible with the quality control in the ER and the ER possesses a remarkable ability to select proteins to be transported in mammalian cells in marked contrast to yeast cells, which actively utilize intracellular traffic to deal with unfolded proteins accumulated in the ER.

    In mammalian cells, the Golgi apparatus is disassembled at the onset of mitosis and reassembled at the end of mitosis. This disassembly-reassembly is generally believed to be essential for the equal partitioning of Golgi into two daughter cells. For Golgi disassembly, membrane fusion, which is mediated by NSF and p97, needs to be blocked. For the NSF pathway, the tethering of p115-GM130 is disrupted by the mitotic phosphorylation of GM130, resulting in the inhibition of NSF-mediated fusion. In contrast, the p97/p47 pathway does not require p115-GM130 tethering, and its mitotic inhibitory mechanism has been unclear. Now, we have found that p47, which mainly localizes to the nucleus during interphase, is phosphorylated on Serine-140 by Cdc2 at mitosis. The phosphorylated p47 does not bind to Golgi membranes. An in vitro assay shows that this phosphorylation is required for Golgi disassembly. Microinjection of p47(S140A), which is unable to be phosphorylated, allows the cell to keep Golgi stacks during mitosis and has no effect on the equal partitioning of Golgi into two daughter cells, suggesting that Golgi fragmentation-dispersion may not be obligatory for equal partitioning even in mammalian cells.

    NSF and p97 are ATPases required for the heterotypic fusion of transport vesicles with their target membranes and the homotypic fusion of organelles. NSF uses ATP hydrolysis to dissociate NSF/SNAPs/SNAREs complexes, separating the v- and t-SNAREs, which are then primed for subsequent rounds of fusion. In contrast, p97 does not dissociate the p97/p47/SNARE complex even in the presence of ATP. Now we have identified a novel essential factor for p97/p47-mediated membrane fusion, named VCIP135 (valosin-containing protein [VCP][p97]/p47 complex-interacting protein, p135), and show that it binds to the p97/p47/syntaxin5 complex and dissociates it via p97 catalyzed ATP hydrolysis. In living cells, VCIP135 and p47 are shown to function in Golgi and ER assembly.

    Membrane fusion plays a crucial role in intracellular vesicular transport, intercellular fusion and viral infectivity. The fusion reaction has been supposed to underlie the following three steps ; (i) specific docking of two membranes, (ii) perturbation of lipid bilayer at fusion point (s) and formingfusion pore (s), (iii) dilation of fusion pore (s). For the intracellular membrane fusion in membrane trafficking pathways, the recent convergence of genetic and biochemical approaches has led to the identification of a number of proteins that involved in specific docking of two membranes (step (i)). However, the details in step (ii) have not been clarified. In this review, I will focus on putative "fusion peptide" of viral fusogenic proteins as an activator of the step (ii). Through the works on the fusion of liposomes mediated by several synthetic peptides that are derived from the fusion peptide of influenza virus hemagglutinin, we found that the fusion is triggered when the peptides become more hydrophobic under fusion-competent conditions, and also found that the common structural features of the fusogenic peptides is not "amphiphilic helical structure". The amphiphilic peptides appearto require other factor (s) to cause fusion. I propose that uneven distribution of the side-chain bulkiness of peptide in ordered structure is important factor for fusion active peptide.

    Protein-mediated membrane fusion plays a crucial role in the eukaryotic world. Especially, steps such as endocytosis and exocytosis require strongly regulated and coordinated fusion events. The fusion protein, however, is still not identified in spite of increased sophistication of cell-free assay systems for measuring the intracellular fusions because "multi-subunit" protein complex seems to be involved in the fusions. In this review we describe some of our physicochemical approaches for searching and understanding the "fusion machinery" in living cells.

    低・中分子・バイオ医薬品の上市には、正常または病態の細胞内環境で実際に働く薬剤標的の同定や、その作用・副作用を細胞内の蛋白質ネットワークの頑健性・撹乱として捉え、その制御の鍵分子(群)の精密な機能同定が必要不可欠になってきている。われわれのグループは、①「超解像蛍光プローブ技術」を利用した高精度蛍光抗体法、②高精度細胞染色画像ビッグデータを用い、蛋白質の性質・挙動変化の時間同調性をもとに作成する共変動蛋白質ネットワーク解析技術、③マルチオミクス解析とのデータ統合による形態情報を特長とする新規ネットワーク解析と数理モデルなどによるネットワークの検証技術、④そこから抽出される鍵蛋白質の正常・病態モデル細胞内での超解像・長時間イメージング技術などの実験的検証、により上記課題を解決し、汎用的で、日本発の技術を統合した世界に類を見ない創薬支援プラットフォームを創成し、創薬・細胞医薬の精緻化とスピード化を目指す。

    われわれは、細胞膜を透過性にして細胞質交換を可能にした「セミインタクト細胞リシール技術」を開発し、細胞内への生体物質導入法や導入した物質の様々な細胞内生命現象への影響などを光学顕微鏡下で可視化解析するマルチスケール解析を中心とした様々な解析システムを構築している。この細胞内操作技術は、多種類のタンパク質成分や膜不透過性の(天然)化合物・低分子化合物・中分子(特殊機能ペプチドや機能性核酸など)を、同時に定量的に細胞内に導入することが可能であり、かつオルガネラや細胞骨格の構造とトポロジーが保持されているため、導入されたタンパク質(群)の様々な動的平衡状態を「真の細胞環境内に近い状態で」計測できる。今年度は、研究計画に沿って、2種類の細胞内導入法(SLO型及びLLO型リシール法)を構築し、その最適化と性能比較を行った。その結果、2種類のリシール法を用い、膜不透過性の低分子・中分子・タンパク質・細胞質など、広いサイズ範囲の物質を、細胞質依存的に(SLO型)、細胞質非依存的に(LLO型)に細胞内に導入するシステムを完成させることができた。また、導入した物質の細胞機能に与える影響を生化学的に検定すル様々な生化学的・細胞生物学的手法の加え、導入物質によって影響を受ける細胞の「状態」変化を、細胞内のタンパク質分子ネットワーク(正確には、共変動分子ネットワーク)として記述するための解析手法(マルチスケール解析法)の応用環境の整備も行った。さらに、SLO型リシール法を用いて作成した、正常または病態モデル細胞に対し、LLO型リシール法を使って様々な低分子・中分子を追加導入することで、低分子・中分子を病態環境でスクリーニングし構造・機能解析するシステムも構築した。これら知見を基に、2種類のリシール法の性能・特長を比較し、幾つかの和文・英文の総説として発表した。

    われわれは、細胞膜を透過性にして細胞質交換を可能にした「セミインタクト細胞リシール技術」を開発し、細胞内への生体物質導入法や導入した物質の様々な細胞内生命現象への影響などを光学顕微鏡下で可視化解析するシステムを構築している。この細胞内操作システムは、多種類のタンパク質成分や膜不透過性の(天然)化合物を、同時に定量的に細胞内に導入することが可能であり、かつオルガネラや細胞骨格の構造とトポロジーが保持されているため、導入されたタンパク質(群)の様々な動的平衡状態を「真の細胞環境内に近い状態で」計測できる。昨年度までに、in cell NMR測定に必要と思われる浮遊状態の細胞(基質からトリプシン処理により遊離させたHeLa細胞、T細胞系の株化細胞や個体から調製した血球系細胞など)をターゲットにしたリシール細胞法の基本プロトコルを作成した。今年度は、その基本プロトコルをさらに進化させ、(1)リシール操作後の細胞のviabilityや生存率の向上を目指す条件検討を行った。その結果、リシール効率及び生存率を飛躍的に増加させる化合物を数種類発見した。当該化合物の作用機序を解明することで,リシール操作に必要な細胞質成分の探索系を構築した。この他、(2)解析対象となるタンパク質や生体分子を導入したリシール細胞に、後から膜不透過性の中分子を、簡単にしかもリシール細胞にダメージを与えない状態で導入する新規の中分子導入法法を開発した。(3)これら膜不透過性中分子の細胞内での機能解析を生化学的手法または光学顕微鏡下の単一細胞レベルで定量的に解析する基本解析プロトコルを構築した。

    We established a neural cell line N14.5-Tom5 cells, in which mitochondrial protein GFP-Tom5 is stably expressed. By using N14.5-Tom5, we found that activation of GSK3beta or Akt was involved in mitochondrial fission during neural differentiation, and identified the phosphorylation of Drp1, dynamin-related GTPase, by GSK3beta regulated the fission. Interestingly, the phosphorylation of Drp1 is reported be required for the mitochondrial fission at the onset of mitosis in mammalian cells. Next, we constructed model cells that replicated the conditions in neurons of Alzheimer's disease by using the resealing-cell technique and cytosol prepared from ApoE(-/-) mouse. As a result, we found that inactivity of Akt in the model cells caused the elongation of mitochondria and resulted in depressed mitochondrial function. Collectively, the persistent phosphorylation of both GSK3beta and Akt plays a crucial role for the maintaining normal mitochondrial morphology and function.

    タンパク質の細胞内局在の攪乱は、細胞恒常性の攪乱(=病態)を誘起する。申請者は、スライドグラス型の「生細胞アレイチップ作成・アッセイ自動化装置」と「チップ自動可視化装置」を構築し、タンパク質の細胞内局在の攪乱(=病態)を誘起する「キナーゼ(群)」を、ヒトキナーゼsiRNAライブラリーから網羅的に可視化スクリーニングする「キナーゼネットワーク可視化解析システム」を構築している。本解析システムを用い、リソソーム酵素の輸送に関わるカチオン非依存性マンノース6リン酸受容体の細胞内局在を攪乱(=病態)するキナーゼ群を、ヒトキナーゼsiRNAライブラリーから網羅的に可視化スクリーニングした。そのうちの一つGSK3βの活性低下やその基質タンパク質である微小管結合タンパク質CLASP2の脱リン酸化が、ポストゴルジ輸送を撹乱し、アルツハイマー病の原因因子であるアミロイドβペプチド(Aβ)の産生を亢進させることを明らかにした。今年度は、そのポストゴルジ輸送攪乱の原因として、GSK3βの活性低下により脱リン酸化したCLASP2がゴルジ体から解離することで、(i)ゴルジ体の断片化、(ii)CLASP2による微小管(+)端動態の撹乱、(iii)ゴルジ体由来微小管中心の形成の撹乱、が同時に生起することで、Aβのプロセッシング酵素であるBACE1の細胞内輸送が撹乱され、エンドソーム内で基質であるAβ前駆ペプチド(APP)とBACE1が共局在化してAPP→Aβへのプロセッシングが活性化されるというモデルを提唱できた。

    マウス・クローディン(CL3)依存的な細胞-細胞間接着を撹乱するキナーゼ阻害剤を網羅的に可視化スクリーニングした。そのため、上皮系培養細胞・Eph4にクローディン-YFP(CL3-YFP)を恒常的に発現するstable transfectan t (Eph4-CL3-YFP)を樹立した。96wellプレートの各wellに、コンフルエントに培養したEph4-CL3-YFP細胞に対し、10μM及び100μMの二種類の濃度のキナーゼ阻害剤ライブラリー(90種類のキナーゼ阻害剤のライブラリー)を、1時間または3時間、37℃でインキュベーションした後の細胞間接着(網目状の細胞接着像)の様子を観察した。観察にはINCell Analyzer (GEヘルスケア社)を用い、各wellから任意の5点を選択し、画像の取得及び解析に供した(ハイスループット可視化スクリーニング)。又、同時に、CL3-YFPの裏打ちタンパク質・ZO-1(内在性)の様子を間接蛍光抗体法により観察し、CL3-YFPの画像と並び比べることにより真に細胞接着のみが撹乱するキナーゼ阻害剤を抽出することができた。その結果、CL3/ZO-1構造の両構造が撹乱を受けるキナーゼ阻害剤(PKC阻害剤を含む9種類)、CL3構造のみが攪乱するキナーゼ阻害剤(1種類)、ZO-1構造のみが撹乱するキナーゼ阻害剤(5種類)を得た。さらに、これらの阻害剤に対し、キナーゼ阻害剤処理濃度、処理時間などを様々に変化させ、共焦点レーザー顕微鏡を用いて細胞接着部分を詳細に観察・検討した結果、細胞-細胞間接着のみに効果を示し、かつ、細胞-基質間接着に影響を与えないキナーゼ阻害剤として、CL3/ZO-1の両構造共に攪乱する阻害剤2種、CL3構造のみ攪乱するもの1種、ZO-1構造のみ攪乱するもの2種を絞り込むことができた。

    ER exit sites(ERES)は、小胞体(ER)からゴルジ体へ向かう輸送小胞の形成プラットフォームである。われわれは先行研究により、膜タンパク質であるYip1AをERESのマーカー膜タンパク質とし、M期におけるERES構造体分解の制御機構を明らかにしている。しかし、本研究実施の過程で、Yip1AはGFP-tag依存的にERESの存在し、多くはゴルジ体に局在化することを発見した。そこで、本研究では、ゴルジ体におけるYip1Aの機能探索を中心に研究を進めた。そのため、小胞体-ゴルジ体間の小胞輸送(順行輸送と逆行輸送)をゴルジ体に局在化するGT-GFP(ガラクトシルトランスフェラーゼのGFP融合タンパク質)をプローブとしFRAP法を用いてそのキネティックスを含めて定量的に解析する手法を構築した。次に、セミインタクト細胞内で細胞質依存的に生起する小胞体→ゴルジ体間輸送(順行輸送)とゴルジ体→小胞体間輸送(逆行輸送)素過程を分割してアッセイする方法を構築した。次に、Yip1Aの細胞質ドメインのペプチド領域(N末から1-109番目)及び(1-65番目)部分のGST融合タンパク質を調製し、細胞質とともにセミインタクトCHO細胞に加えて順行・逆行輸送アッセイを行った。その結果、興味深いことに、逆行輸送のみがそれらペプチドにより有意に阻害されることが判った。これは、Yip1Aの細胞質側ドメインが細胞質またはゴルジ体表面の輸送装置に作用して、ゴルジ体からのGT-GFPの逆行輸送を競争的に阻害したことを意味する。この他にも、この細胞質ペプチド領域は、VTC/ERGICからのp58の逆行輸送を阻害しなかったこと,RNAi法によりHeLa細胞の内在性のYip1AをノックダウンするとShigatoxinのエンドソーム→ゴルジ体→小胞体の逆行輸送が阻害されることなどから、Yip1Aはゴルジ体からのCOPI依存的な逆行輸送に関わることを示唆する結果を得た(論文投稿中)。

    細胞内コレステロール排出活性を持つABCタンパク質ABCA1の一アミノ酸置換変異体ABCA1-QRは通常は小胞体に留まったままだが、小胞体ストレスに敏感に反応して小胞体を離脱し形質膜へと輸送されることを見出した。実際、ABCA1-QR-GFPを恒常的に発現しているstable transfectant(HEK-QR)に小胞体ストレス(DTT処理又はタプシガルジン処理など)を負荷したところ、小胞体に蓄積していたQR-GFPはおおよそ30分以内で核周縁領域に移行し蓄積する様子が観察された。つまり、小胞体に蓄積しERAD経路を取って分解されていたQR-GFPは、小胞体ストレス負荷により通常の小胞輸送経路を取って細胞形質膜へと輸送されるており、同一の膜タンパク質が小胞体内でERAD経路と小胞輸送経路を選別しうることが明らかとなった。そこで、申請者は小胞体ストレス依存的なQR-GFPの小胞体からの輸送に関わるキナーゼの網羅的同定を試みた。始めに20種類のキナーゼ阻害剤をHEK-QRに作用させた上で小胞体ストレスを負荷し、ジアシルグリセロールキナーゼおよびGSK3beta阻害剤などによって輸送が阻害されることを明らかにした。さらに、阻害剤の結果に基づいて当研究室が保持する細胞チップ自動作成装置を用いて網羅的キナーゼRNAiを行い、数種類のキナーゼが小胞体ストレス依存的なABCA1-QRの輸送に関与することを見つけた。今後はこのキナーゼ群の関与の確認、および詳細な分子メカニズムの同定を行う予定である。

    本研究では、細胞膜を透過性にした「セミインタクト細胞」に、ストレスにさらされた細胞から調製した細胞質を導入し、ストレス負荷時に翻訳が活性化される転写因子ATF4の発現の可視化・再構成を行い、ストレス依存的な翻訳制御の分子機構を明らかにすることを目的とした。先ず、1)ATF4mRNAを恒常的に発現する細胞株の樹立した。2)生細胞におけるATF4-GFPのストレス応答反応の可視化した。CHO-ATF4細胞に小胞体ストレス(タプシガルジン、DTT処理など)や酸化ストレスを負荷することにより、ATF4-GFPがストレス依存的に翻訳され、核内に移行することを確認した。この細胞株のATF4mRNA量はストレス負荷時、ストレスのない場合においても殆ど変化していないことをRT-PcRやノーザンプロットにより確認した。3)セミインタクト細胞を用いたストレス依存的ATF4-GFPの翻訳制御の可視化・再構成系の構築した。CHO-ATF4細胞をセミインタクト細胞処理し、小胞体ストレス(タプシガルジンやDTT等)を負荷したCHO細胞(WT)より調製した細胞質を添加し、ATF4-GFPの翻訳制御の再構成を試みた。その結果、ストレス負荷細胞質依存的にATF4-GFPが翻訳され、かつそれが核内に効率よく輸送されることを確認できた。ATF4-GFPは分子量約70kDaでの転写因子であり、例え発現したとしてもセミインタクト細胞の穴より細胞外へ拡散してゆくことが危惧されていたが、非常に効率よく核に移行することより、本再構成アッセイ系が翻訳制御の可視化再構成系として十分使用できること、また、セミインタクト細胞内では転写因子のような核移行する分子は何らかの方法で(恐らく微小管依存的に)核にターゲットできることが明らかとなった。これらの結果より、各種ストレス依存的な翻訳制御ネットワークの可視化解析を行う準備ができた。

    本研究では小胞体ストレスや酸化ストレス等の多様な細胞ストレス付加によって翻訳制御を受けている転写因子ATF4の翻訳制御機構を明らかにするため、ストレス依存的に細胞質内にできてくるストレスグラヌルとATF4のmRNAの相互作用について研究を行った。目的のmRNAを生きた細胞内で可視化するためMS2-tag法を利用した。ATF4、アクチン及び膜タンパク質CD4のmRNAの3‘末端にMS2-配列を挿入したプラスミドとGFP-MS2融合タンパク質のプラスミドをCHO細胞に通常の方法で同時にtransfectionした。この時、GFP-MS"タンパク質には核移行シグナルを挿入し、細胞質内でMS2-tag付きmRNAと結合できなかったGFP-MS2は核へ移行し、細胞質内のmRNAが観察しやすいように工夫した。その結果、生きたCHO細胞内において、ATF4、アクチン及びCD4のmRNAの動態を可視化することに成功した。興味深いことに、アクチンmRNAは細胞底面の周縁部のアクチンフィラメントに沿って局在し、ストレス負荷時にもその局在に影響はなかった。ATF4-mRNAは細胞質全体に散在していたが、ストレス負荷によってもその局在に変化はなかった。これに反し、CD4-mRNAは、ストレス負荷により局在を細胞質からストレスグラニュルに大きく変化させた。これらの結果より、ストレス負荷時に翻訳活性化されるATF4は、細胞質に残り、通常翻訳抑制を受けるCD4等のタンパク質はストレスグラニュルにmRNAが集積し翻訳阻害されると予想された。アクチン等ストレス負荷によって翻訳制御を受けないタンパク質も、そのmRNAの局在を変化させない仕組みがあることが判った。このmRNAの可視化システムは、mRNAのストレスグラニュル局在化による翻訳制御メカニズム解明のための基本アッセイ系となることが判った。

    Vero毒素(O-157毒素)は細胞のエンドサイトーシス経路によって細胞内に取り込まれ、リソソーム経路を回避しながらゴルジ体へと移行し、さらに分泌経路を遡り小胞体(ER)内へ移行する。そして、小胞体内腔から毒素活性のあるAサブユニットが細胞質へ移行することで毒性を発揮する。その輸送経路・制御機構を明らかにすることは毒性発現の予防や疫学的見地から有用であるばかりか、本来、厳格な選別機構で制御されているはずのエンドサイトーシスとエキソサイトーシス経路の曖昧さと両者のクロスロードを制御する分子機構を明らかにできる。本研究では、細胞膜を一部透過性にしたセミインタクト細胞系と細胞内小胞輸送可視化技術を駆使して、哺乳動物細胞における外来細胞毒素(Vero毒素)の細胞内進入機構とその毒性発現制御機構を明らかにすることを目的とした。本年度までに、Vero毒素をリコンビナントタンパク質として大腸菌により大量調製する系を確立した。生細胞内輸送過程を追跡するためCy2標識したを蛍光標識毒素をプローブとして細胞内輸送過程追跡に用いた。その結果、毒素受容体を持つVero生細胞における蛍光標識毒素の細胞内輸送過程(細胞膜→エンドソーム→ゴルジ体→小胞体)を可視化することに成功した。さらに、後期エンドソーム→TGN間の輸送過程をより詳細に検討する目的で、マンノース6リン酸受容体(cation-dependent)のGFP融合タンパク質(GFP-Man6-P受容体)の細胞内動態の可視化と輸送キネティックスの解析法の樹立した。また、セミインタクトVero細胞を用い、蛍光標識毒素及びGFP-Man6-P受容体の細胞内移行過程の可視化再構成し、毒素及びGFP-Man6-P受容体の後期エンドソーム→ゴルジ体(TGN)間輸送過程を細胞質依存的に再構成することができた。

    In mammals, endoplasmic reticulum (ER) stress is sensed and transduced by three ubiquitously expressed transmembrane proteins in the ER (IRE1, PERK and ATF6). Physiological significance and roles of the IRE1 and PERK pathways have been clarified by making their knockout mice. We have succeeded in creating ATF6alpfa and ATF6beta knockout mice in this study. Thus, we now know phenotypes of knockout mice deficient in three major signal transduction pathways operating in the mammalian unfolded protein response (UPR).
    Double knockout mice of ATF6alpfa and ATF6beta caused embryonic lethality at very early days (before embryonic day 8.5), which represents an earliest phenotype observed in knockout mice of UPR signaling molecules. ATF6alpfa was required for transcriptional induction of not only ER chaperones but also components of ER-associated degradation. Although it was previously shown that PERK knockout decreased the levels of ER chaperones, we were able to show that PERK knockout affected activation of ATF6. A group in USA reported previously that knockdown of ATF6alpfa and ATF6beta had almost no effect on induction of UPR target genes, however, we were able to show that it is not the case and that rather the ATF6 pathway is most important in the three signaling pathways.
    In contrast to the IRE1 and PERK pathways, ATF6 itself is involved in all steps in signal transduction, from sensing ER stress to activation of transcription. By analyzing activation mechanisms of ATF6, we showed the presence of ATF6-escort protein through identification of its recognition sites and discovered a very interesting regulation, ER stress-induced reduction of disulfide bonds, that occurs in the oxidative ER.
    We also showed that ATF6 is a transcription factor specializing in transcriptional induction of ER quality control proteins and that ATF6 has gained function in the UPR only in higher eukaryotes such as mammals. These constitute very important information in thinking of evolution of the UPR.

    われわれが独自に開発してきた「セミインタクト細胞系」及び「細胞内タンパク質動態可視化技術」を駆使し、小胞体ストレスに応答する細胞内小胞輸送の影響を研究し下記の結果を得た。ABCトランスポーターの一種ABCA1の変異体QRは、従来の活性発現場所である形質膜へは移行せず、小胞体内に蓄積することが知られている。1.QRのGFP融合タンパク質(QR-GFP)を恒常的に発現しているHEK293細胞株(以下、HEK(QR))を構築し、細胞内挙動をタイムラプス装置を持った共焦点レーザー顕微鏡で観察したところ、網目状の典型的な小胞体ネットワークに蓄積していることが判った。2.この細胞に還元剤(DTT)やタプシガルジン(Tg)などよる小胞体ストレスを付加すると、QR-GFPは小胞体から速やかに放出され核近傍のオルガネラに移行した。更にインキュベーションを続けるとQR-GFPのシグナルは形質膜にも観察された。細胞生物学的研究により、小胞体ストレスによってQR-GFPが通常の分泌経路を通り小胞輸送によって形質膜まで運ばれることを確認した。3.通常、小胞体ストレス不可により、小胞体-ゴルジ体間輸送は阻害される(VSVGts045蛋白質の輸送で確認)が、QR-GFPはむしろストレス応答的に開始されるacuteな反応であることが判った。4.この輸送は、{小胞体-ゴルジ体間輸送を阻害するBFAにより阻害されることより、COPI依存的な輸送であることも明らかにした。5.興味深いことに、この輸送は、ARF1ドミナントネガティブ蛋白質(通常の輸送は阻害される)によって活性化されることが判った。6.この輸送経路はセミインタクト細胞系では小胞体ストレスの有無にかかわらず進行した。これらのことより、この小胞輸送は、小胞体の小胞形成過程におけるQR-GFPの小胞内積み込みの為の選別過程での制御によるものであることが明らかになったため、現在、その細胞質因子、小胞体内腔の因子についてその制御に関わる因子を探索中である。

    細胞増殖信号を処理する細胞内分子システムEGF-Ras-MAPKシステムは、細胞増殖信号を処理する細胞内分子システムである。このシステムの内、新規遺伝子発現に至るメインストリーム、すなわちEGFと受容体の結合から、低分子量G蛋白質Rasの活性化、MAPキナーゼカスケードの情報伝達をへてERKが核移行するまでを再構成することを目的とし研究を行った。
    1)セミ・インタクト細胞システムの最適化
    StreptolysinO処理により調製したセミ・インタクト細胞内でEGFによるEGF受容体のリン酸化を指標として、情報伝達反応が、非処理細胞と同様に起こるような処理法の最適化をおこなった。セミ・インタクト細胞内に外来タンパク質が効率よく導入されることを確認した。
    2)タイパク質発現系、標識法の確立
    再構成系の要素となる各種情報伝達蛋白質を蛍光標識、精製するための発現系の構築を行った。EGF-Ras-MAPKシステムの構成要素であるMEK,ERKの蛍光蛋白質融合体の作製と培養細胞における発現の確認を行おこない、両蛍光蛋白質融合体が正常な局在と分子間相互作用を示すことが確認できた。CFP-MEK1とYFP-ERK2を共発現したHeLa細胞にEGF信号が入ると、細胞質に局在していたERKが核へ局在変化する。単独発現系ではMEKは細胞質、ERKは核内に発現し、共発現系(無刺激時)では、両蛋白質とも細胞質に局在した。これは、静止時の細胞ではERKとMEKが複合体を形成して、ERKの核移行が抑えられていることによる。セミ・インタクト系の実験でタンパク質濃度を制御するために、MEK,ERKの大腸菌発現系を構築し、MEK,YFP-ERKの精製法を確立した。

    小胞体(ER)ストレスに対する哺乳動物細胞の小胞輸送システムの応答とそのダイナミズムを明らかにするため、われわれが独自に開発した「セミインタクト細胞系」及び「細胞内タンパク質動態可視化技術」をカップルさせたアッセイ系を駆使し下記の結果を得た。細胞の脂質代謝に関わるABCトランスポータの一つABCA1とその変異タンパク質(QR)のGFP融合タンパク質を恒常的に発現しているHEK細胞株を樹立しそれらタンパク質の動態を可視化解析した。その結果、ABCA1は細胞形質膜へ輸送され機能するが、QRはER内腔に留まりERのERAD経路によってプロテアソーム分解されることを見いだした(プロテアソーム阻害剤存在下でのQRのアグリソーム形成)。興味深いことに、QRは特定の細胞外ストレス(還元剤添加など)により、ERAD系から逃れ、通常の小胞輸送経路に乗ってゴルジ体→形質膜へと輸送されることを発見した。しかも、この輸送は、通常の分泌タンパク質が利用するCOPII小胞を介してERからERGIC(ER-Golgi中間構造体)を経由してゴルジ体、形質膜まで運ばれた。このER→ゴルジ体小胞輸送とERAD経路の選択の制御因子をセミインタクト細胞系などを用いて調べたところ、small GTP結合タンパク質の一つでるARF1のGTP活性化フォームが増加することによりERAD経路から小胞輸送経路へのスイッチが行われることを見いだした。これらの結果は、ERストレスが今まで知られていたERAD経路やUPD経路を活性化するだけでなく、新しく小胞輸送の活性化によって蓄積したタンパク質を細胞外へ先ずは吐き出すという回避経路を併せ持つこと、その制御には、ARF1またはCOPIと言うゴルジ層板間やゴルジ体→ERの逆行輸送に関わるとされる分子が関わっていることを強く示唆した。

    哺乳類の概日リズム(サーカディアンリズム)発振の分子メカニズムを明らかにするため、マウス時計遺伝子mPer遺伝子上流配列(プロモーター領域を含む)にレポーター遺伝子としてd1-GFP遺伝子(改変型GFP)を繋いだ遺伝子(mPer-d1-GFP)を作成し、mPer-d1-GFP遺伝子を持ったトランスジェニックマウスを作成した。d1-GFPとは、哺乳類細胞内で半減期約1時間で分解を受ける改変型のGFPである。そのため、mPer遺伝子を発現する臓器の細胞内で、約一時間だけ蛍光を発することができる。つまり、トランスジェニックマウスのSCNニューロン及びmPer遺伝子を発現している臓器(の細胞)は、概日リズムと同期して蛍光を発することになる。作成したトランスジェニックマウスの初期培養SCNニューロンは、自家蛍光が強いがそれをバックグラウンドとして周期的な(概日リズムに対応した)蛍光を発することを確認した。今回の研究期間中には、マウスの作成、その系統維持。初代培養細胞での概日リズム確認までを行った。この間に、いろいろな時計関連遺伝子が同定されまたその遺伝子産物間の相互作用などが次々と明らかのなってきている。今後、これら同定された遺伝子産物のGFP融合タンパク質(または、GFP変異体融合タンパク質、CFP,YFPなど)を無細胞系タンパク質発現系を用いて発現・精製し、セミインタクトSCN細胞を用いた単一細胞レベルでの「時計」の再構成系を構築する予定である。現在、共同研究により、gene chipを用いた時計遺伝子の網羅的解析と変動遺伝子の抽出、抽出遺伝子産物の無細胞タンパク質発現系を用いた発現システムの構築を進めている。

    1. メタノール資化性酵母におけるオルガネラ動態のビジュアル化と応用機能開発
    (1)代謝及び蛋白質輸送機能
    メタノール酵母に発達しているペルオキシソーム代謝、特に抗酸化代謝機能に関わる2つの蛋白質、カタラーゼとPmp20について、その生理生化学的機能を明らかにした。特にPmp20については、ペルオキシソームに局在するペルオキシレドキシンとして過酸化脂質を基質としたグルタチオンペルオキシダーゼであることを初めて明らかにし、カタラーゼの持つ抗酸化反応との差異を明確にした(J.Biol.Chem,276,14279)。また、これらの蛋白質のペルオキシソームへの局在化効率には差があることを示し、その差は局在化シグナルとそのレセプターとの相互作用の強さに依存していることがわかった(J.Bacteriol.,183,6372他)
    (2)ペルオキシソーム分解の分子機構
    ペルオキシソームの発達した細胞をその不要な培地に移すとペルオキシソームが選択的に分解される。この過程を2重蛍光染色法により1細胞で追跡することにより、液胞が分断化しながらペルオキシソームを包み込みこんで行くことを明らかにした。さらにその過程を支配する20以上のPAZ遺伝子を同定しペルオキシソーム分解の分子モデルを提出した(Genes Cells,7,75他)。
    (3)蛋白質生産系への開発と応用
    蛋白質やペルオキシソーム分解に関わるPEP4及びPRB1遺伝子をCandida boidiniiiからクローニングして高蛋白質生産用の宿主を創生した。さらに臨床診断酵素として有用なペルオキシソーム酵素であるD-アミノ酸オキシダーゼの高生産に成功した(Biosci, Biotechnol.Biochem.,65,627;ibid.,66 628)。
    平成13年度に予定されていた本研究は、申請者が基盤研究(S)を受けることになったため、辞退することになった。しかしながら、その決定通知を受け取るより以前の段階において、上記成果を論文発表するための図表作製に必要な消耗品として、本研究費の一部を充当することになった。

    We have established CHO mutants defective in the late endocytic pathway (LEX mutants). We have found that one of the mutants, LEX2, was defective in late endosomal sorting due to the lack of an enzyme involved in the later stage of cholesterol biosynthesis, and showed that (1) cholesterol is required for the sorting from MVBs to the Golgi and that (2) MVB reorganization is important for this late endosomal sorting. To study the dynamics of GFP-tagged organelles or transports/targeting of GFP-tagged proteins in single cell under a microscope, we have developed a novel microphotometric assay system by using GFP-visualization techniques coupled with semi-intact cell system. Using the assay system, (1) we have visualized and investigated the morphological changes of the ER-network in CHO cells during mitosis using the GFP-tagged HSP47. Time-lapse images and photobleaching experiments revealed that the ER-network remains almost continuous throughout the cell cycle, but is partially severed during mitosis. The cell cycle-dependent behavior of the ER-network, disruption followed by reformation, was reconstituted in SLO-permeabilized CHO cells using mitotic and interphase cytosol. Remarkably, the reformation of the disrupted ER-network was achieved through two sequential fusion reactions. The first process was mediated by NSF/α-SNAPs, and typical membranous intermediate structures were generated to connect the disrupted ER-tubules. The second fusion was mediated by p97/p47/VRF135, the same minimal components required for the homotypic fusion events in Golgi cisternae regrowth after mitosis. (2) We have visualized and reconstituted the vesicular transports between the Golgi and ER in semi-intact CHO cells. Based on the kinetic analysis, we found that, in the presence of mitotic cytosol, anterograde transport from the ER to Golgi was inhibited depending on cdc2 kinase but the retrograde transport was not. The transitional-ER (tER), which is a platform of ER-derived transport vesicles, was found to be disrupted in the presence of mitotic cytosol. Such perturbation of ER-Golgi transports might result in the relocation of the Golgi components to the ER during mitosis.

    極性細胞内の小胞輸送におけるVIP17等の蛋白質の機能を研究するための新規な細胞内小胞輸送アッセイ系を確立した。まず、ゴルジ体局在化シグナルを持つガラクトシルトランスフェラーゼと緑色蛍光蛋白質(GFP)との融合蛋白質(GFP-GT)を、恒常的に発現しているCHO細胞・MDCK細胞を得た。蛍光性膜蛋白質(GFP-GT)をプローブとすることにより、共焦点レーザー蛍光顕微鏡・超高感度ビデオ付き蛍光顕微鏡。エバネセント蛍光顕微鏡下で、「単一」の細胞内のGFP-GTの輸送過程を経時的に追跡できた。また、フォトカウントシステムを用い、小胞体→ゴルジ体間の輸送のキネティックスを「定量的」に解析することができた。この結果、輸送小胞には少なくとも3種類の小胞があり、それぞれが微小管依存的にダイナミックに動く様子が世界で初めて観察できた。また、ストレプトリシンO(SLO)を用いて形質膜を部分的に透過性にしたセミインタクト細胞内で、この輸送過程を、ATP再生系とL5178Y細胞の細胞質を用い再構成することに成功した。セミインタクト細胞内での輸送過程を上記顕微鏡システムを用いて追跡・解析した結果、小胞体からゴルジ体への輸送過程をATP-依存的な過程、ATP-細胞質-依存的な過程の、少なくとも2つの素過程にdissectionすることができた。また、輸送に関わる中間体としてダンベル状の大きさ約0.3-0.5μmの輸送小胞が頻繁に観察され、ATPをdepletionした場合、その数は激減した。この中間体が、今まで報告されている「intermediate compartment」又は「pre-Golgi structure」である可能性が高い。この再構成システムを用いることにより、それぞれの過程に関与する細胞質因子や、輸送中間体(小胞)のダイナミクスを制御する因子の検索が可能となった。

    The methylotrophic yeast is widely used as a heterologous gene expression system in academic and applied fields. When cells are grown on methanol, a large volume of cells (up to 80%) was occupied by a single-membrane bound organelle, the peroxisome. The study was conducted to produce toxic oxidases within peroxisomes of the methylotrophic yeast, since many oxidases are useful for clinical diagnosis. To this end, the protein traffic was manipulated for efficient protein transport, and this strategy (or concept) was named "sorting engineering" (later biotraffic engineering). Since genetic diorders deficient in peroxisome assembly are known, the molecular mechanism for protein traffic to peroxisomes was in important in medical field and investigated using the methylotrophic yeast as a model organism. In this study, the information obtained from the basic research was applied to production of fungal fructosyl amino acid oxidase, which is useful for determination of glycated proteins. In diabetic patients, the level of glycated proteins is high. Using (i) an efficient targeting signal to peroxisomes and (ii) an alcohol-oxidase depleted strain, and (iii) optimizing codon usage and culture conditions, we could improve the enzyme productivity approximately 47-fold from the orginal transformant. The produced protein accumulated within membrane-bound peroxisomes up to nearly 20% of the toal soluble protein. This is the first successful example which was conducted based on the concept "sorting engineering" ("biotraffic engineering"), and a similar approach will be possible in any other eukaryotic cells for heterologous gene expression, especially for the production of toxic or membrane proteins.

    セミインタクト極性細胞内で、細胞周期依存的なのゴルジ体の小胞化・リアッセンブリー過程の再構成系を確立した。まず、ゴルジ体局在化シグナルを持つ膜蛋白質とGFPとの融合蛋白質を恒常的に発現しているMDCK細胞株を樹立した。ポリカーボネートトランスウエル上で培養したこのMDCK細胞をストレプトリシンO(SLO)で処理し、形質膜を部分的に透過性にしたセミインタクト細胞を作成した。この細胞に、イリマキノン(IQ)を作用させ、ゴルジ体を小胞化させた(IQ依存的小胞化過程)。次に、IQをよく洗い流し、新たに外部よりL5178Y細胞の細胞質とエネルギー再生系(ATP再生系)を加えて小胞化したゴルジ体の核周囲へのリアッセンブリーを再構成した(リアッセンブリー過程)。この再構成系を用い、小胞化過程・リアッセンブリー過程それぞれを2つの素過程にdissectionすることに成功した。次に、それぞれの過程のキネティックスを解析するため「morphometric analysis法」を開発し、中間状態としてゴルジ体が大きめの小胞に分裂し、細胞質中に分散する状態(punctated Golgi membarane)を同定できた、また、それぞれの素過程に必要な因子・阻害剤を検索した結果、ゴルジ体の小胞化前期過程には三量体G蛋白質、リアッセンブリー後期過程にはsmall GTP結合蛋白質の関与を示唆する結果を得た。セミインタクトMDCK細胞に、M期カエル卵抽出液を用いることによってもIQの場合と同様にゴルジ体を小胞化させることに成功した。この過程も、2つの素過程にdissectionできATP再生系を必要とすることが判った。また細胞分裂時に活性化されるキナーゼに対する阻害剤を用いた実験より、過程前期はMAPキナーゼキナーゼ(MEK)が、そして過程後期にはcdc2キナーゼが段階的に効いていることが判った。この再構成系の確立によって、今後、ゴルジ体の細胞周期依存的なグイナミクスの研究とそれぞれの素過程に必要な因子の検索が可能になった。

    我々は、イヌ腎臓上皮由来の培養極性細胞であるMDCK(Madin-Darby canine kidney)細胞のトランスゴルジネットワーク(TGN)での蛋白質・脂質の選別と小胞への組み込みに関わるソーター蛋白質の同定とその選別メカニズムを、TGN由来の分泌小胞膜蛋白質VIP17の細胞内機能解明を中心に研究している。今年度は、(1)極性細胞というコンテクストの中で分子の種々の機能を生化学的に探るため、連鎖球菌の酵素感受性毒素Streptolysin O(SLO)を用いて形質膜を部分的に透過性にした「セミインタクト細胞系」を完成した。これを用い、イリマキノン(海綿の代謝産物)により引き起こされた細胞分裂時におけるゴルジ体の小胞・断片化->再集合を、L5187Y培養細胞質とATP再生系を用い再構成することができた。(2)多重蛍光顕微鏡解析装置を開発した。本装置により、光学顕微鏡下で培養細胞・セミインタクト細胞の形態変化・カルシウム濃度変化・膜電位変化・pH変化等とともに、「細胞内の局在」で生起するイベント(蛋白質相互作用、膜動過程など)を高空間分解能・高時間分解能で定量化し、イメージングできる。今年度は、蛍光標識短鎖セラミド(C5-DMB-セラミド)を用い、生細胞中のゴルジ体からの分泌小胞形成の定量的アッセイ法を確立した。

    Caveolae are non-clathrin-coated surface invaginations present in many cell types and function as a device to trap glycolipids, cholesterol, and other proteins for potocytosis, endocytosis, transcytosis, and intracellular signal transductions. Caveolin (caveolin-1) is a 21-24 kDa integral membrane protein, which is localized to both caveolae and trans-Golgi derived exocytic vesicles. To investigate the possible lipid interactions of caveolin, we reconstituted Esherichia coil-expressed caveolin into proteoliposomes with various lipid compositions and found that the protein was incorporated into DOPC/cholesterol proteoliposomes only when >30 mole % cholesterol was present in the lipid mixture. We also demonstrated that the protein bound cholesterol specifically. A number of proteins are known to interact with cholesterol, but to our knowledge caveolin is the first "membrane protein" that binds cholesterol. The organizing mechanism for the complexes of caveolin and cholesterol molecules has not yet been elucidated. The finding that cholesterol remained bound to the protein was completely unexpected since the final purification step of dog lung caveolin, which isolated from dog lung using biochemically, was sedimentation through a sucrose gradient containing 0.2% SDS, which should have removed associated lipids. Therefore, the binding of cholesterol to caveolin is markedly stable, suggesting lipid-protein interactions with unusual characteristics. This finding indicates that the protein plays a specific role in membrane microdomain (caveolar-related domains) formation and intracellular cholesterol trafficking.

  • MDCK細胞における極性輸送小胞と形質膜との融合装置の探索

    イヌ腎臓上皮細胞であるMDCK細胞のapical形質膜とトランスゴルジネットワーク(TGN)由来の分泌小胞との膜融合に関わるタンパク質を探索するため、次の方法でタンパク質をスクリーニングした。Apical輸送小胞とbasolateral輸送小胞とを、Simonsらの方法でMDCK細胞より単離した。これらのタンパク質組成を二次元電気泳動法により調べ、両者に共通に存在するタンパク質をピックアップした。これらは、両小胞の共通機能である膜融合・膜小胞化などに関係している可能性が大きい。特に、その中でも膜タンパク質は、膜融合・膜小胞化機能に関する細胞装置の基幹となるタンパク質である可能性が大きい。二次元電気泳動ゲルより、膜タンパク質の一つ(VIP17)に注目し、microsequencing・oligo合成・PCR法によってMDCKのcDNAライブラリーよりVIP17のcDNAをクローニングした。VIP17は、膜四回貫通部位を持つ膜タンパク質(proteolipid)であった。ホモロジー検索の結果、ヒトTリンパ球の形質膜タンパク質であるMALや、ラットシュワン細胞のミエリン膜に局在するM14のホモログであった。また、HA-tagを付けたVIP17をMSCK細胞に発現させ、その細胞の内局在を間接螢光抗体法により調べた結果、VIP17-HAtagは、apocal、basolateral形質膜とapica輸送小胞に局在していることが分かった。現在、正確な細胞内局在とその細胞内機能(特に膜融合機能)を研究する目的で、VIP17の融合タンパク質・合成ペプチド・精製タンパク質を抗原とした抗VIP17モノクローナル抗体を作成中である。また同時に、蛍光性セラミドによるTGN由来小胞の蛍光ラベル化を試み、部分的に形質膜をpermeableにしたMDCK細胞を利用した形質膜-輸送小胞の膜融合アッセイ系を作っている。

  • 新しい蛍光ラベル化リガンドを用いたエンドサイトーシス研究とその素過程変異株の分離

    本研究は、真核細胞のエンドサイトーシス過程における細胞内輸送に関与する細胞側因子(細胞装置)を分子遺伝学的に同定し、その分子機構解明を目的としている。本年度は、エンドサイトーシス素過程の内、特に、後期エンドソームからリソソームへのリガンド(低密度リポタンパン質)の移行過程(リガンドのソーティング)に変異を持つ細胞を簡便、迅速に効率よく分取するため、また、そのようにして得た変異「生」細胞中のLDLの分解を定量的にアッセイするための方法を確立できた。具体的には、低密度リポタンパク質(LDL)内にNBD-リノール酸エステル(NBD-LE)とR18(ローダミン系脂質アナログ)を再構成法により封入し、受容体媒体エンドサイトーシスによりCHO細胞に取り込ませた。この時、エンドソーム内のLDLではNBD-LEとR18に蛍光共鳴エネルギー移動(RET)が生起していて2つの蛍光強度化(NBD/R18)は小さい。しかし、リソソーム内に移行してLDLが分解されると、2つの蛍光色素間の相対的距離が大きくなり、RETが解消し、[NBD/R18]の値が大きくなる。蛍光ラベルしたLDLを取り込ませ、その細胞をNBDとR18の蛍光を同時に蛍光測光可能なセルソータを用いて分析すれば、個々の細胞の蛍光はそれから得られる蛍光強度比[NBD/R18]の値に対応して、2次元モニター上に個々の点としてプロットされる。[NBD/R18]のプロットの分布の仕方で、LDLの後期エンドソームよりリソソームへの移行の程度が定量的に解析できた。[NBD/R18]の値が小さい1つのプロットは、つまり、RETが解消されていない細胞1個に相当するので、これをセルソータにより分取すれば、この条件でのこの輸送過程に変異を持つ変異株を生きたまま分離することが可能である。これとは別にコレステロール生合成阻害剤であるプラバスチンとプロモデオキシウリジンを用いたネガティブ選択法により、LDLの細胞内での分解に異常を持つT41細胞を、CHO-K1細胞を野生株として得ることに成功したが、このT41内のLDLの分解の遅さを上記セルソータを用いた方法で定量的に解析できた。
    今後は、このような変異株の分取を本方法にて行い、その分子遺伝学研究を進めたい。

    我々はすでに、エンドサイトーシスを利用した細胞質内物質導入法に用いる人工ウイルスに必要不可欠な「酸性での」膜融合活性を有するペプチドE5の合成に成功し、ペプチドの「両親媒性ヘリカル構造」が膜融合活性発現に必要不可欠であることを明らかにしている。本年度は、さらに新しいペプチド(E5NN,E5CN,E5CC,E5P)を設計、合成した。それらはE5ペプチドをプロトタイプとし、それぞれ異なるα-ヘリックス含量を持つ両親媒性ペプチドである。これらによる中性リン脂質リポソームの膜融合活性、赤血球溶血活性、CDスペクトルなどによる脂質膜との相互作用を調べた結果、ペプチドがもつ両親媒性ヘリカル構造は膜構造にperturbationをおこしたり、bindingしたりすることには必要であるが、膜融合活性には必要十分な構造でないことがわかった。我々は、これらfusion-activeなペプチドに必要なもう一つのfactorとして、α-ヘリカルなペプチドを構成するアミノ酸の側鎖のbulkinessのペプチド1次構造上の不均一な分布に注目した。アミノ酸側鎖のbulkinessの目安として、各残基のsolvent-accessible surface area(A^2)を用い、この値をα-ヘリックスをとった時のペプチドのhelical wheel図上にプロットすると、fusion-activeなペプチド(E5,E5CN)のそれは、大きな残基と小さな残基が円周上に不均一に分布し、ペプチド表面が″rough″であるが、fusion-inactiveなペプチド(E5NN)のそれは、″smooth″であることが判った。″rough″なペプチドは、ヘリックスをとりながらもその表面は完全できれいな両親媒性ではなく、ペプチドが結合した膜界面でdynamicに動き、膜をより一層乱し、融合に導くと考えられる。これに対し″smooth″なペプチドは、チャネルを作ったりporeをつくり、膜(中)上で安定に存安し、界面を乱し膜融合をおこす程にはならないと考えられる。

    To investigate the process of endocytosis in a living cell in real time, we have developed a new microscopic system, in which a time-resolved fluorescence apparatus is combined with a fluorescence confocal microscope. The time resolved fluorescence apparatus is composed of a pulse laser (wave length : 365 nm ; pules width : 15 picosecond ; output power : several tens pJ/pulse) and a synchroscan streak camera which can record decay time of the pulse laserexcited fluorescence emission in real time, and the confocal fluorescence microscope has a spatial resolution of >1mum. Using system, it is possible to investigate the dynamic process of endocytosis of fluorescent ligand in a living cell in real time with a spatial resolution of > 1mum.
    We have developed a novel fluorescence method to monitor the lysosomal disintegration of low density lipoprotein (LDL) particles in living cells. The method is based on the fluorescence resonance energy transfer (RET) between two fluorescent molecules incorporated into LDL particles. RET-LDL particles is endocytosed by cells and relief of the RET upon the disintegration of the LDL in lysosme is monitored by flow cytometory. This method enables us to monitor the process of endocytosis from endosome to lysosome without disruption of the cells. Using this method, we firstly demonstrated that protein phospholylation plays essential role in the transport of LDL particles from endosome to lysosome in CHO cells.
    We isolated a mutant cell line of CHO cells which is deficient in the transport of endocytosed ligand from endosome to lysosome, using the abovementioned method. We analyzed the mutation site by the fluorescence timeresolved confocal microscope and cell fractionation, and showed that the mutation site might be at the point where the pathway of endocytosis overlaps the sorting pathway from trans-Golgi network to the lysosome.

    インフルエンザウイルスの膜融合活性タンパク質(HAタンパク質)の活性部位を模した酸性pHー依存的膜融合活性ペプチド(HAペプチド)及び、そのアナログペプチド数種を合成し、タンパク質・核酸・薬剤等を封入したリポソ-ム表面に吸着・共有結合させた擬似インフルエンザウイルス(以下人工ウイルス)を調製すれば、細胞のエンドサイト-シスを利用したエンドソ-ム経由の全く新しい細胞内物質導入法の開発ができる。本年度は、人工ウイルスのkey pointとなる弱酸性pH(<6.0)での膜融合活性及び、赤血球溶活性などのbiomembraneに対する活性を有する酸性ペプチド(E5,E5L)を合成し、蛍光法・電顕観察を中心とした物理化学的手法により、融合活性に必要不可欠なペプチドの機能モチ-フを研究した。その結果、HAペプチドのN末端10残基程が活性発現に必要な両親媒性ヘリカル構造をつくる為の「核」となっていることを明らかにした。
    また、培養細胞に取り込まれた人工ウイルスの挙動をリアルタイムに近い時間スケ-ルでモニタ-することは、今後の人工ウイルス改良に必要不可欠である。本年度は、リポソ-ムを蛍光共鳴エネルギ-移動(RET)を起こす2種の蛍光性脂質(エネルギ-供与体;NBDーPE,エネルギ-受容体;R_<18>)でラベルし、COSー1細胞に非特異的にエンドサイト-シスさせ、高感度ビデオ付き蛍光顕微鏡などで追跡することができた。また、リポソ-ムがエンドソ-ムやリソソ-ムで破壊されたり、それらの膜と融合すると、蛍光性脂質がそれらエンドソ-ムの膜上で拡散、RETが解消し、エネルギ-供与体(NBDーPE)の蛍光強度が増加し蛍光寿命が長くなることを、時間分解顕微蛍光法やセルソ-タを駆使し、単一細胞内での挙動を定量的に測光、追跡できた。

    薬物・核酸・タンパク質を封入したリポソ-ム表面に、弱酸性pH(<6.0)領域でのみ膜融合活性を持つペプチドを結合させた人工ウイルスを調製し、細胞のエンドサイト-シスを利用した効率の良い、細胞にダメ-ジの少ないドラッグデリバリ-システムの開発を目的とし、本年度は次の(1)(2)の結果を得た。
    (1)人工ウイルス表面のfusogenとして最も重要な弱酸性pHで正確に膜融合活性が制御できるペプチドを、インフルエンザウイルスの膜融合タンパク質(HAタンパク質)の活性部位を模したHAーペプチドをプロトタイプとして5種合成した。これらペプチドによる中性リン脂質(egg PC)よりなるリポソ-ムの融合活性を測定し、また、ペプチドとリポソ-ム・赤血球膜などとの相互作用を円二色性偏光など物理化学的手法を用いて研究した。その結果、各ペプチドは至適pHを5.0付近に持ち、pH6.0以下でのみ効率の良い膜融合活性・赤血球溶血活性などを持つことが判った。また、活性の無い中性pHにおいても強くリポソ-ムの膜表面に結合していること、酸性でのペプチド中の酸性アミノ酸残基のプロトン化と、それによるペプチドの疎水性の増加がペプチドのヘリックス含量を増加させ、活性発現の引金となることなども判った。
    これらのことより、今後、fusogenとなるペプチドは「両親媒性ヘリカル構造」を基本構造とする酸性ペプチドとして設計すればよいことが示された。
    (2)人工ウイルスの生きた培養細胞内での挙動を経時的に追跡する手法の確立は、設計された人工ウイルスの検定に必要不可欠である。我々は、セルソ-タ、時間分解顕微蛍光法などを利用し、培養CHO細胞にエンドサイト-シスさせた蛍光性リポソ-ムを用いて、蛍光の共鳴エネルギ-移動効率の変化により、そのリポソ-ムの細胞内エンドソ-ム、リポソ-ム内での分解・融合の程度を顕微鏡下に生きた細胞を観察しながら定量的に測定する方法を開発した。
