    <jats:title>Abstract</jats:title><jats:p>We report the syntheses of novel marine biodegradable poly(ethylene succinate) (PES)- and poly(butylene succinate) (PBS)-based copolymers containing different dicarboxylic acid (DCA) units with various carbon numbers and different feed ratios. Biochemical oxygen demand tests demonstrated that some of the obtained PES- and PBS-based copolymers were biodegradable in seawater. Specifically, polymers with longer-chain DCA units, even at low contents, exhibited marine biodegradability. The thermomechanical properties of the copolymers, such as their thermal stabilities, melting points, glass transition temperatures, tensile moduli, strains at break, and stresses at break, also varied with the DCA contents. These results indicated that the thermomechanical properties and the marine biodegradabilities of the PES- and PBS-based copolymers were regulated by controlling their structures and DCA contents. The polymers obtained in this study may replace general-purpose polymers. Our approach may also be applicable to other polymeric materials. Furthermore, our findings pave the way for the rational design and preparation of polymeric materials that are biodegradable in environments other than oceans and have good thermomechanical properties.</jats:p>

    <jats:p>Polyhydroxyalkanoates (PHAs) are aliphatic polyesters synthesized intracellularly by microorganisms as a carbon-storage substance. Among the various PHAs, 3-hydroxybutyrate (3HB)-based copolymers are crystalline polymers widely used as biodegradable plastics. Recently, PHAs containing α-carbon-methylated monomers, such as 3-hydroxy-2-methylbutyrate (3H2MB) and 3-hydroxy-2-methylvalerate (3H2MV), have been synthesized and characterized. However, a binary copolymer of 3HB and 3H2MV, P(3HB-co-3H2MV), had not yet been synthesized, and its material properties had not been investigated. In this study, P(3HB-co-11 mol% 3H2MV) (PHBMV11) was synthesized with recombinant Escherichia coli LSBJ, using trans-2-methyl-2-pentenoic acid as the 3H2MV precursor. The thermal properties of PHBMV11 were characterized using differential scanning calorimetry (DSC), and the results were compared with those of P(3HB-co-12 mol% 3-hydroxyvalerate) (PHBV12) to explore the effect of α-carbon methylation in the comonomer unit. PHBMV11 exhibited a higher enthalpy of fusion during the DSC heating process and higher crystallization temperature during the DSC cooling process than those of PHBV12. The half-crystallization time of PHBMV11 was slightly longer than that of the P(3HB) homopolymer and much shorter than that of PHBV12 previously reported. The α-carbon methylation of the 3H2MV unit also has a positive effect on the crystallization of 3HB-based copolymers, as the 3H2MB unit demonstrated previously.</jats:p>

    <jats:p>Polyhydroxyalkanoate (PHA) synthases (PhaCs) are key enzymes in PHA polymerization. PhaCs with broad substrate specificity are attractive for synthesizing structurally diverse PHAs. In the PHA family, 3-hydroxybutyrate (3HB)-based copolymers are industrially produced using Class I PhaCs and can be used as practical biodegradable thermoplastics. However, Class I PhaCs with broad substrate specificities are scarce, prompting our search for novel PhaCs. In this study, four new PhaCs from the bacteria <jats:italic>Ferrimonas marina</jats:italic>, <jats:italic>Plesiomonas shigelloides</jats:italic>, <jats:italic>Shewanella pealeana</jats:italic>, and <jats:italic>Vibrio metschnikovii</jats:italic> were selected <jats:italic>via</jats:italic> a homology search against the GenBank database, using the amino acid sequence of <jats:italic>Aeromonas caviae</jats:italic> PHA synthase (PhaC<jats:sub>Ac</jats:sub>), a Class I enzyme with a wide range of substrate specificities, as a template. The four PhaCs were characterized in terms of their polymerization ability and substrate specificity, using <jats:italic>Escherichia coli</jats:italic> as a host for PHA production. All the new PhaCs were able to synthesize P(3HB) in <jats:italic>E. coli</jats:italic> with a high molecular weight, surpassing PhaC<jats:sub>Ac</jats:sub>. The substrate specificity of PhaCs was evaluated by synthesizing 3HB-based copolymers with 3-hydroxyhexanoate, 3-hydroxy-4-methylvalerate, 3-hydroxy-2-methylbutyrate, and 3-hydroxypivalate monomers. Interestingly, PhaC from <jats:italic>P. shigelloides</jats:italic> (PhaC<jats:sub>Ps</jats:sub>) exhibited relatively broad substrate specificity. PhaC<jats:sub>Ps</jats:sub> was further engineered through site-directed mutagenesis, and the variant resulted in an enzyme with improved polymerization ability and substrate specificity.</jats:p>

    <jats:p>A new polythioester (PTE), poly(3-mercapto-2-methylpropionate) [P(3M2MP)], and its copolymer with 3-hydroxybutyrate (3HB) were successfully biosynthesized from 3-mercapto-2-methylpropionic acid as a structurally-related precursor. This is the fourth PTE of biological origin and the first to be α-methylated. P(3M2MP) was biosynthesized using an engineered Escherichia coli LSBJ, which has a high molecular weight, amorphous structure, and elastomeric properties, reaching 2600% elongation at break. P(3HB-co-3M2MP) copolymers were synthesized by expressing 3HB-supplying enzymes. The copolymers were produced with high content in the cells and showed a high 3M2MP unit incorporation of up to 77.2 wt% and 54.8 mol%, respectively. As the 3M2MP fraction in the copolymer increased, the molecular weight decreased and the polymers became softer, more flexible, and less crystalline, with lower glass transition temperatures and higher elongations at break. The properties of this PTE were distinct from those of previously biosynthesized PTEs, indicating that the range of material properties can be further expanded by introducing α-methylated thioester monomers.</jats:p>

    <jats:title>Abstract</jats:title><jats:p>Recently biobased plastics draw attention as eco‐friendly materials, and it is important to study the effects of substituents, which are popularly found in natural products, on the characteristics of the resulted polymers. In this study, twelve resorcinol‐based semiaromatic polyesters are successfully synthesized using four types of resorcinol derivatives (resorcinol, 2‐methylresorcinol, 5‐methylresorcinol, and 2,5‐dimethylresorcinol) and three types of dicarboxylic acids (glutaric acid, adipic acid, and pimelic acid) to investigate the effect of methylation at the 2‐ and 5‐positions in resorcinol. Glass‐transition temperature of the polyesters is located from ‐3.1 to 37.9 °C, and rises with increasing the number of methyl groups in a resorcinol unit. Crystallinity of the polyesters decreases by methylation to a resorcinol unit, except the polyester from 2‐methylresorcinol and pimelic acid. The mechanical properties of the solvent‐cast films for the polyesters are tunable by the site and number of methyl groups in a resorcinol unit. The major thermal degradation of the polyesters is observed above 300 °C, and the minor thermal decomposition appears between 100 and 200 °C for the polyesters without the resorcinol methylated at the 2‐position and pimelic acid unit.</jats:p>

    <jats:title>Abstract</jats:title><jats:p>Given their ubiquity in modern society, the development of biodegradable and renewably sourced plastics is essential for the creation of an environmentally sustainable society. One of the drawbacks for currently available biodegradable plastics such as poly(<jats:sc>l</jats:sc>-lactic acid) (PLLA) and polyhydroxyalkanoates (PHAs) is that it is difficult to simultaneously achieve mechanical flexibility and certain crystallization behavior in these materials, which limits their use as replacements for established petroleum-based plastics such as isotactic polypropylene (iPP). Here, we report the synthesis and characterization of a new biodegradable plastic, poly(3-hydroxy-2-methylbutyrate) [P(3H2MB)], which is a member of the bacterial PHA family whose members include an α-methylated monomer unit. Biosynthesis of P(3H2MB) was achieved using recombinant <jats:italic>Escherichia</jats:italic><jats:italic>coli</jats:italic> expressing an engineered pathway. Biosynthesized P(3H2MB) exhibited the highest melting temperature (197 °C) among the biosynthesized PHAs and improved thermal resistance. It also exhibited improved crystallization behavior and mechanical flexibility nearly equal to those of iPP. The primary nucleation rate of P(3H2MB) was faster than that of P(3HB), and the spherulite morphology of P(3H2MB) was much finer than that of P(3HB). This crystal morphology may result in more rapid crystallization behavior, increased transparency, and enhanced mechanical properties. The superior physical properties of P(3H2MB) have the potential to open new avenues for the production of high-performance biodegradable plastics for replacing petroleum-based bulk commodity plastics.</jats:p>

    <jats:title>Abstract</jats:title><jats:p>Various copolymers incorporated with β-substituted acrylates, such as alkyl crotonates (e.g., methyl crotonate (MC), ethyl crotonate (EC), isopropyl crotonate (iPC), and <jats:italic>n</jats:italic>-butyl crotonate (nBC)) and methyl cinnamate (MCin), were synthesized by group-transfer polymerization (GTP) using a silicon-based Lewis acid catalyst. In addition to β-substituted acrylates, α-substituted acrylates (e.g., methyl methacrylate (MMA) and <jats:italic>n</jats:italic>-butyl methacrylate (nBMA)) were examined as comonomers. Proton nuclear magnetic resonance (<jats:sup>1</jats:sup>H NMR) spectroscopy and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) characterizations of the obtained copolymers revealed that each monomer component was incorporated sufficiently. The thermal stabilities of the resulting copolymers were investigated by dynamic mechanical analysis (DMA), indicating that the glass-transition temperature (<jats:italic>T</jats:italic><jats:sub>g</jats:sub>) of the copolymers can be widely varied over a relatively high-temperature range by selecting the optimal comonomer. More specifically, the <jats:italic>T</jats:italic><jats:sub>g</jats:sub> values of poly(MC-<jats:italic>random</jats:italic>-EC) (MC/EC molar ratio = 50/50), poly(MC-<jats:italic>random</jats:italic>-nBC) (MC/nBC molar ratio = 50/50), poly(MC-<jats:italic>random</jats:italic>-MCin) (MC/MCin molar ratio = 54/46), and poly(nBC-<jats:italic>random</jats:italic>-MCin) (nBC/MCin molar ratio = 56/44) were 173, 130, 216, and 167 °C, respectively.</jats:p>

    <jats:title>Abstract</jats:title><jats:p>Cinnamic monomers, which are useful chemicals derived from biomass, contain <jats:italic>α</jats:italic>,<jats:italic>β</jats:italic>-unsaturated carbonyl groups with an aromatic ring at the <jats:italic>β</jats:italic>-position. Homopolymers synthesized by addition polymerization of these compounds are expected to be innovative bio-based polymer materials, as they have both polystyrene and polyacrylate structures. However, polymerization of these compounds by many methods is challenging, including by radical methods, owing to steric hindrance of the substituents and delocalization of electrons throughout the molecule via unsaturated π-bonding. Herein we report that homopolymers of these compounds with molecular weights (<jats:italic>M</jats:italic><jats:sub>n</jats:sub>) of ~18,100 g mol<jats:sup>−1</jats:sup> and controlled polymer backbones can be synthesized by the group-transfer polymerization technique using organic acid catalysts. Additionally, these homopolymers are shown to have high heat resistance comparable to that of engineering plastics. Overall, these findings may open up possibilities for the convenient homopolymerization of cinnamic monomers to produce high-performance polymer materials.</jats:p>

    It was examined that the relationship between the structure and thermal properties of the novel alternating copolyesters of 3-hydroxybutyrate and lactate units. Based on the crystalline structure of alternating copolymers with high-heat resistance, it was found that the polymer molecules form the densely chain-packed crystals by reducing the helical radius. In addition, the alternating copolymers of 3-hydroxybutyrate and the other hydroxyacids with chiral center were synthesized, and their structure and thermal properties were determined.

    Synthesis of copolymers with periodic sequence structure was achieved by two-step polycondensation reaction from 3-hydroxybutyrate and aspartate. When the copolymers reacted with alkyl amine to form ion-complex at side-chain of aspartate units, the enzymatic degradation reaction by polyester-degrading enzyme was inhibited. It was found that the enzymatic degradability of copolymers was regained by the washing under the acidc condition. From the obtained results, we proposed new methodology to control the starting point of biodegradation for the periodic copolymers by changing the pH as outer stimulus.

    有限化石資源の有効利用と環境保全の観点から、新たに開発すべき環境循環型プラスチックは、単に自然環境中で分解されることにとどまらず、糖や植物油などの再生産可能資源(バイオマス)から生産できるバイオベースプラスチックでなければならない。さらに、環境循環型高分子の観点から、バイオベースプラスチックと生分解性プラスチックの両方の特性および機能を有する高分子材料の基礎、開発、実用化研究が今後最も重要な研究課題になると考えられる。
    本基盤研究(C)では、「次世代バイオベースプラスチックの創成」を研究する特定領域研究を発足させるために、世界的研究動向と研究ニーズを調査し、この調査研究に基づいて個別研究および共同研究を企画し、研究組織を作り上げることを目的とした。
    平成18年9月15日に、理化学研究所にて本研究に携わる13名が各々のこれまでの研究成果と今後の研究計画を発表し、本研究に関心を持つ一般の研究者・技術者からの意見を収集した。さらに、本企画調査研究への補助金交付決定後ただちに第1回全体会議を東京で開催し、平成19年度新規特定領域研究への申請を行うことを決定した。そのために各研究分担者が、担当する研究分野の世界動向と研究ニーズを調査した。さらに、数回の班長会議および全体会議を行い、追加メンバーの承認、統一テーマの設定、共同研究の開拓、特定領域としての意義等を検討した。
    これらの検討をふまえ、平成19年度発足を目指した特定領域研究「新規高性能バイオベースポリマーの創成と高機能化」(領域代表:岩田忠久)を申請した。

    前年度の成果によって得られたモノマーの分子構造およびオリゴマーの連鎖長の異なる8シリーズ、56種類のポリエステルアミド周期共重合体試料の物性評価を行うとともに、結晶構造・固体構造の解析を行った。
    周期性共重合体の中には、熱処理により複数の融解挙動を示すものが存在することを見出した。周期性共重合体の融解挙動における熱処理効果を詳細に調べた結果、熱処理後に見られる融解温度の急激な増加は、ラメラ結晶の厚化と、それに伴う結晶領域中へのアミドユニットの取り込みに起因することが明らかとなった。すなわち、結晶の大きさをコントロールすることで、周期性共重合体の熱的性質を制御できることを見出した。また、周期性共重合体の力学的性質を評価したところ、周期連鎖の長さに依存して、やわらかい性質から硬い性質まで幅広い物性を示すことが明らかとなった。
    アジピン酸および炭素数が6のジオールとジアミンから合成した周期性共重合体の単結晶および配向結晶性フィルムを調製し、その結晶構造を解析に取り組んだ。電子線及びX線回折測定の結果、周期性共重合体の結晶は、単位格子中に4本のセグメントを有しており、そのセグメントは周期連鎖中の1ユニットに相当することが明らかとなった。さらに、コンピュータプログラムによる結晶構造及びパッキング解析シミュレーションを行ったところ、周期性共重合体の結晶が、エステルユニットに起因した単位格子を有すると同時に、その格子中でアミドユニットに起因した分子鎖折りたたみシート構造を形成していることが明らかとなった。また、同じユニットから構成されたポリエステル、ポリアミド、周期性共重合体結晶の分子モデルの比較から、熱安定性に優れた分子鎖設計に向けた基礎的知見を得ることに成功した。
