    Abstract

    A cutting plotter, a widely used office device, enables users to prepare plastic sheets in arbitrary shapes that have been designed using a computer. Cutting plotters are applied for fabricating microfluidic systems made of thin plastic sheets. For this study, commercially available aluminum tape with an adhesive layer was tested to create a microchannel with electrodes. After optimizing cutting conditions and evaluating accuracy under an optical microscope, the electrodes were used to assemble a pollen sensor. After the electrodes were placed on a solid substrate, nematic liquid crystal 4-cyano-4’-pentylbiphenyl (5CB) was injected between them. The device detected several pollen particles inserted into 5CB through the impedance measurement.

    Abstract

    Polytetrafluoroethylene (PTFE) is an indispensable material for society and industry. PTFE chains have a strong intermolecular force and form strongly bound crystal structures. The strong assembly causes a low surface energy and refractive index, as well as difficulty in processing and recycling. In this paper, we describe how PTFE powders were milled with NaCl crystals using a planetary ball mill. Furthermore, the milled PTFE-NaCl powders were immersed in water to remove NaCl, and filtered powders were obtained. These powders were characterized by X-ray diffraction (XRD) and infrared spectroscopy. The PTFE-NaCl powders had a lower XRD peak intensity than that of processed PTFE powders without NaCl. Even the filtered powders had a lower XRD peak intensity. These results suggest that the intermolecular assembly was loosened by the mechanochemical process with NaCl. Then, PTFE-NaCl and filtered powders were pressed to form pellets. The pellets were successfully sintered, while PTFE powders milled without NaCl were not.

    Abstract

    Surface tension plays an important role in the micrometer-sized fluid interface, but only a few methods can measure it remotely. On spatially restricted surfaces, plural eigenmodes of thermally induced capillary wave resonance are observed, and their resonant frequencies depend on the surface tension. Incident laser light is scattered by the capillary wave with Doppler shift. The resonant peaks corresponding to the eigenmodes are observed in the power spectra. In this study, a spectrum fitting algorithm utilizing the theoretical frequency ratio of the plural eigenmodes is proposed. In addition, the position dependence of the fitting quality on the position along the diameter axis was also investigated. As a result, the algorithm worked well in the circular center area. This algorithm and knowledge will be valuable for various applications such as environmental and biological surface science.

    Abstract

    Monitoring phycotoxin accumulation in marine products such as edible shellfish is a regulatory requirement in many countries. Therefore, a simple and rapid onsite quantification method is sought. Herein, we present a fluorescence polarization immunoassay (FPIA), a well-known one-step immunoassay, using a portable fluorescence polarization analyzer for domoic acid (DA), widely referred to as the primary toxin of amnesic shellfish poisoning (ASP). To establish FPIA for DA, the matrix effect of methanol, which is widely used to extract DA from shellfish, on FPIA was investigated. To validate this method, we performed a spike recovery test using oysters containing DA at a concentration equivalent to the regulatory limits of North America and the European Union (20 mg/kg). The recovery rate was found to be 79.4–114.7%, which is equivalent to that of the commercially available enzyme-linked immunosorbent assay (ELISA). We expect that this FPIA system will enable the quantitative onsite analysis of DA and significantly contribute to the safety of marine products.

    This paper is the first report of a non-competitive fluorescence polarization immunoassay (NC-FPIA) using a peptide as a tracer. The NC-FPIA can easily and quickly quantify the target after simply mixing them together. This feature is desirable for point-of-need applications such as clinical diagnostics, infectious disease screening, on-site analysis for food safety, etc. In this study, the NC-FPIA was applied to detect CD9, which is one of the exosome markers. We succeeded in detecting not only CD9 but also CD9 expressing exosomes derived from HeLa cells. This method can be applied to various targets if a tracer for the target can be prepared, and expectations are high for its future uses.

    Abstract

    Wide and operate safety monitoring of foodstuffs is highly demanded in modern society. This work aims to develop and characterize new test systems for control of phycotoxins, dangerous contaminants of fish and seafood. For rapid and productive testing, 2 methods have been implemented: immunochromatographic assay and fluorescence polarization immunoassay. Various approaches for reducing the limit of detection of target analytes have been considered, including changes in the use of optical and fluorescent labels and varied order of the detected complexes formation. Antibodies and aptamers have been considered as receptor molecules. The developed techniques provide rapid (20–30 minutes, including sample preparation) and sensitive testing. Their effectiveness has been shown for different kinds of fish and seafood. Portable detectors have been proposed that allow testing directly at the sampling points, without transportation to centralized laboratories.

    This paper reports a microchemical chip for electrochemical measurement of ion transport at the water-oil interface during microdroplet formation in a microchannel. For the electrochemical measurement of the interface, the aqueous-organic-aqueous three-phase system was formed in the microchannels. The channel wall was needed to be hydrophobic in order to prevent the aqueous phases from unfavorable intrusion. By using this system, the charging current to the electrical double layer in single microdroplet was measured.

    Lateral bias印加のための電極内蔵型支持体を作製し,この支持体中での膜形成について検討した.ベース素材としてはシリコン(Si)チップとテフロンフィルムの2種を比較した.その結果,いずれの素材においても脂質二分子膜の形成が可能であったが,Siチップでは脂質分子のみで膜形成が可能であるのに対し,テフロンフィルムでは孔周辺にヘキサデカンの塗布が必要であった.次に,これらの膜系にLateral biasを印加したところ,Siチップ系では交流電圧が,テフロンフィルム系では直流電圧が低ノイズ計測に適していることが分かった.さらに,形成した脂質二分子膜中にフラーレン誘導体(PCBM)や生体イオンチャネルを包埋し,Lateral biasの効果について検討した結果,PCBMドープ膜が示す膜貫通方向の光誘起電流や,電位依存性Naチャネルが示す膜貫通電流が,ともにLateral biasによって増大する現象を見出した(Faraday Discussion, in press, 2022).しかし,その作用メカニズムは不明であったため,平面脂質二分子膜に対する蛍光観測系を立ち上げ,Lateral biasの作用メカニズムについて蛍光プローブを用いた検討を進めた.また,上述の電極内蔵型支持体では,Lateral biasの印加に伴って電極材料のチタン(Ti)が酸化チタン(TiO2)へと変化するという課題も判明したが,Ti電極を白金(Pt)で被覆することによって酸化を防止できることも分かった.来年度は,電極内蔵型支持体の量産化も進め,メカニズムの検討および応用探索を加速化していく予定である.

    In this project, a table-top quasi-elastic laser scattering apparatus was developed. The table-top apparatus had a sufficient ability to measure a gas-liquid interface, whose characteristics were comparable to that utilizing a lab-scale laser system. Furthermore, to analyze microscale bubble surface, a microdevice fabrication method for stable bubble formation was demonstrated. Next, an optical trapping method of the bubble was developed. Then, a quasi-elastic laser scattering measurement was demonstrated for the first time.

    Water-in-oil microdroplets formed in microfluidic devices have been studied with eagerness in the past decade because of they can be used as picolitter sized chemical containers and applied to analysis for high-throughput microanalyses such as single cell assay. However, the quantification of the microdroplet contents was difficult because of its short optical path lengths. In order to overcome this difficulty, we developed the selective condensation method for microdroplet contents by utilizing spontaneous emulsification at the interface of the microdroplets. Here, we demonstrated the selective condensation of analyte molecules in microspaces, and their applications to protein determination and crystallization.

    A quasi-elastic laser scattering (QELS) method utilizing capillary wave resonance has been investigated for measuring surface or interfacial tension of microscale liquid interface. In the resonant power spectrum of QELS method, characteristic peaks which are independent of the optical setup were observed and could be utilized for determining the tension. In this study, we have improved the QELS theory for power spectrum prediction, investigated microfluidic devices and optical apparatuses, and discussed 1-micrometer-sized small liquid interfaces.

    CCDを検出器として用いた光熱変換分光法について検討した。波長532 nmの励起光と波長633 nmのプローブ光を、それぞれ音響光学素子で変調し、同軸にした後、顕微鏡ステージ上のガラス製マイクロ流体チップ中の液体試料に照射した。励起光を除去した後、プローブ光をCCD(12bit、プログラム可能)に導入した。励起光の変調周波数を1743 Hz、プローブ光の変調周波数を1748 Hzと5 Hzの差をつけ、CCDを20 Hzで動作させた。励起光による光熱変換現象と、プローブ光照射に5 Hzのうなりが生じ、うなりの一周期中に四枚のCCD画像が得られる。この四枚の画像の四則演算および積算により光熱変換画像を得た。幅220マイクロメートル、深さ100マイクロメートルの流路をもつガラス製マイクロ流路チップを用いた。試料には、色素Sudan IVのトルエン溶液を用いた。励起光強度が数mWの条件で、1.0×10-6 Mの溶液を問題なく検出できることがわかった。 CCDを検出器として用いる場合、光強度を電気信号に変換してすぐに、アナログ/デジタル(A/D)変換するため、計測の感度はA/D変換のbit数に制限される。12bitのCCDを用いる場合、4096階調であり、光強度の10,000分1の信号強度を取得することは原理上できず、本手法の感度上の制約となる。この問題を解決するためには、A/D変換...

    本研究では、マイクロチャネルにより規定された空間で起こるベルーゾフ・ジャボチンスキー反応(BZ反応)について、チャネルサイズ、形状依存性およびチャネルパターンによる伝播特性を、顕微分光法・マイクロ電気化学法を駆使して明らかにすることを目的とする。初年度は、BZ反応を行う場となる種々のチャネル幅、およびチャネル深さのマイグロチャネルの設計・作製を行い、実際にこれらマイクロメートルサイズの微小空間内でBZ反応が起こることを確認するとともに、化学振動がサイズ依存性を示すことを見出した。本年度は、その定量的評価を行うために、熱レンズ検出法を用いた微小吸光度測定を行った。反応系の濃度、特にフェロイン濃度の最適化を行った。これにより、熱レンズ信号の化学振動に伴う強度変化が大きくなり、振動信号からフーリエ変化して得られる振動周期を精度よく決定することができるようになった。その結果200μmまでのサイズにおいては振動周期は短路サイズに比例して大きくなること、KBrO_3に特に鋭敏に依存すること、およびチャネル内壁を疎水処理すると周期が約2倍となることがわかった。これらの結果より、微小空間で起こるBZ反応では、チャネル内壁へのKBrO_3局所濃度変が反応の切り替えを誘起し、その信号がチャネル内部に速やかに伝播し化学振動が起こると結論した。これは、内壁の比界面積が極端に大きくなる微小空間の特性を反映したものと考えられる。これを利用することにより、界面の微小変化を捉えるセンサー等への応用が考えられる。

    化学装置を数cm角の基板上に集積化する研究が、近年急速な勢いで進展している微小流体の研究として、2相以上の相合流・相分離を利用したキレート溶媒抽出・相間移動反応などを現在までに実現した。本研究では、微小流体は通常スケールとは異なる境界条件が必要であるという観点からマイクロチャネル肉の微小流体を解析し、自由に設計・制御する方法を開発することを目的とする。平成14年度には、上面を親水性、下面を疎水性とする表面修飾パターンニング法により幅300μm深さ200μm全長20mmのマイクロチャネル内の安定多相流を実現した。また、液液界面における界面張力波による光散乱を計測する手法である準弾性レーザー散乱法を顕微鏡下で実現した(μQELS法)。
    平成15年度は、表面修飾パターニング法を利用したマイクロ空間における油水対向流(マイクロ向流)を実現し、回収効率を飛躍的に向上できる向流抽出を世界で初めて実現した。また、流体を蛍光粒子可視化法により解析したところ、レイノルズ数が1程度であるにもかかわらず液液界面付近で渦を形成していることを観測した。マイクロ向流中の水相・油相は、従来の流体力学に見られないマイクロ界面における特異な流体力学的挙動を示すという発見があった。
    また、μQELS法により、マイクロ二相流界面の流体特性について検討した。従来のマクロスケール実験では、油水平行層流を形成することは難しく、実現できたとしても大きな剪断応力を印可することは不可能であった。本研究で開発したマイクロチャネル修飾法を利用することにより、液液界面に大きな剪断応力を印可したところ、界面張力が変化するという現象を見出した。このことは、液液界面における新規な境界条件の必要性を強く示唆している。従来実現できなかった実験をマイクロ化学システムの技術を利用して実現した。

    Integration of various chemical and/or physical functions such as electrodes, catalysts, or controlled wettability surfaces is a promising approach for further development of micro total analysis system (μTAS) or lab-on-chip chemistry. The aim of this research is development of novel microchemical processes by using functional microchannel chips. Brief accomplishments are as follows.
    1. Through optimizing bonding procedures in fabrication processes of microchannel chips, we succeed in integration of metal electrodes such as Au, Pt, Ag, and so on, as well as semiconductor electrodes such as TiO_2 in the microchannels without solution leakage. Also, we succeed in modification of the channel surface with silica or titania nanoparticles.
    2. As an analytical application of the electrode integrated microchannel chip, we developed electrochemically assisted thermal lens microspectroscopy. Ultrasensitive detection system of non-fluorescent analytes with electrochemical selectivity was successfully constructed.
    3. Applying the electrode integrated microchannel chip to electrochemical synthesis such as Michael additions, we succeeded in enhancing the current efficiency and selectivity of the reactions.
    4. We developed a novel micromixer based on AC electroosmotic flow induced with meandering electrode structure, which allowed fast mixing of solutions in microchannel in ms time scale.
    5. Photofunctional interfaces in the microchannel were applied to microfluid control. By using photochromism of spirobenzopyran derivatives, flow-switching device was constructed. Titania nanoparticles-hydrophobic molecules modifying surfaces were used to prepare multistep Laplace valves, which were applied to construct micro batch operation system and oil-water phase separation system.
    6. Potential controllable micro photocatalytic reactors were developed by integration of titania film with microchannels. We succeeded in optimization of optical yield of redox-combined photocatalytic reactions such as one-step synthesis of L-pipecolinic acid from L-Lys by applied potential.

    The major purpose of this research project is conducting the microchip technology to organic synthesis. In order to realize this concept, following researches were performed.
    (1)Investigation of the reaction performance of interfacial reaction in microchip
    (2)Development of novel chemical processes using multiphase flow
    (3)Realizing mass-producible synthesis using parallel microscale reactions using plural microchips
    Concerning (1), diazo-coupling reaction, amidation reaction or alkylation reactions were applied for phasetransfer reactions in microchip, and high performance chemical reactions were successfully performed. Especially, alkylation reaction in microchip showed substrate generality, thus the reaction can be widely applicable for general alkylation reactions. Concerning(2), we developed the segmented flow injection-two phase flow formation system for selective extraction of chemical substances in microchip, the liquid membrane-transport system for molecular transport, and the membrane separation system by preparing chemically-functionalized membrane in microchip, all of which can only realized in microchip. These processes can be applicable for separation of synthesized product in microchip. Concerning (3), 3D-microchannel chip for combinatorial synthesis and mass producible synthesis were developed. These microreactors were applied for amido-compound synthesis, and "massive synthesis" including number of compound and amount of compound was successfully performed using these chips.
    In conclusion, initial purpose of this research project was totally achieved by the fundamental investigations listed above. We expect these microreactors to be a versatile tool of future organic synthesis.

    Integration of electrochemical detection and/or reaction fields in a microchannel is one of important techniques for further development of "μTAS" or "lab-on-chip system", In this research, we developed novel methods to fabricate glass-substrate microchannel chips integrated with microelectrodes. The most important technology is bonding of channel and electrodes substrates without solution leakage. We established two methods, one is a method using sodium silicate as adhesive layer and another is thermal bonding. By using the methods, we succeed integration of metal electrodes such as Au, Pt, Ag, and so on, as well as semiconductor electrodes such as TiO_2 in microchannels. To check the electrochemical responses of the fabricated chips, we performed both cyclic voltammetry and chronoamperometry under solution flow conditions. Also, we calculated steady-state current from simulations by using a finite element method. The flow rate dependence of the experimentally obtained steady-state currents agreed very well with the calculated results, which indicated that the microelectrode worked well without solution leakage. As application of the electrode integrated microchannel, electrospectroscopic detection using thermal lens spectroscopy, electrochemical synthesis, and photocatalytic reactions were demonstrated.

    本研究は熱レンズ顕微鏡の更なる超高感度化と実用化を視野に入れ、暗視野光熱変換顕微鏡と可搬性の高いパームトップサイズ熱レンズ顕微鏡を開発することを目的とする。
    1.暗視野光熱変換顕微鏡の開発
    市販の生物顕微鏡を励起光と検出光の2本のレーザー光を導入できるように改良して、暗視野型光熱変換顕微鏡を作製した。作製した暗視野光熱変換顕微鏡を用いて励起光吸収用色素を内包したポリスチレン(80nm)粒子分散水を測定したところ、セル壁面で光が散乱され、原理的にバックグランドフリーである暗視野配置で高いバックグランド信号が発生していることがわかった。これを解決するために対物レンズと光検出器の間にピンホールユニットを作製し、セル壁面からの散乱光を排除して焦点付近からの光のみを通過・検出する(擬似共焦点配置)ようにした。これによりバックグランドを4桁低減することに成功した。
    2.パームトップサイズ熱レンズ顕微鏡の開発
    熱レンズ顕微鏡を小型化するためには、光源や光学系の小型化が必要である。熱レンズ顕微鏡の構成要素である励起光と検出光の焦点差を調整する機構と対物レンズを、色収差を持つ円筒形微小レンズに置き換えた新しい光学システムを設計し、作製した。既存のシステムと比較し、作製したシステムを評価したところ、小型レンズを用いた場合に特有のノイズが発生することを見いだした。今後、微小レンズ固定法の改良によりこのノイズを低減する予定である。

    ガラス基板上に加工した微小流路を化学実験室として利用する研究を進めているが、微小空間の微小信号から分子の振る舞いを知ることは非常に困難である。本研究では、スクイズド光源の低ノイズ特性を活かした、微小空間の溶液界面を観測するための光ヘテロダイン分光法を開発することを着想した。具体的には、スクイズド光ヘテロダイン法を微小空間の液液界面で実現し、輸送現象などを直接観測することを目的とした。
    本研究は、微小流路中における液液界面の計測を目的としているが、ヘテロダイン測定のためには、光の入射方向に対して垂直な界面を形成しなければならない。しかし、微小空間においては界面張力が重力に比べ大きく寄与するため、液液界面全体がメニスカス状になり界面が曲がってしまう。そこで、表面修飾により微小空間における濡れ性を制御する加工法を考案し、光入射方向に垂直な水/ニトロベンゼン(W/NB)界面を微小流路中に形成することに成功した。このW/NB界面をヘテロダイン測定したところ、低周波成分のノイズが除去できず、物性値測定は困難であった。そこで、ローカル光の光周波数を音響光学素子にて80MHzシフトさせた後、ヘテロダイン測定したところ低周波成分のノイズ除去に成功し、流れている液液界面においても物性値測定が可能であった。全体のノイズレベルから考えると、測定はスクイズド光の特性を利用しているとは考えられないが、流れている液液界面というノイズの高い条件下で高い信号が得られる測定セル・光学系を見いだしたことにより、当初想定していた測定が可能となった。本研究で開発した測定装置系により、微小空間における物質輸送・界面吸着ダイナミクスなどの化学プロセスを測定可能であることを示せた。

    The Flow characteristics in a microchannel, operated by pressure-driven flow, are described by a low Reynolds-number flow : laminar flow. In laminar flow, the flow line runs parallel with the cannel-wall, so that the inertial effects on molecular motions and the turbulent mixing of solution do not occur. Owing to such flow characteristics, even when two solutions are introduced separately into a microchannel chip, the two folutions in the single channel can flow witout any mutual mixing. On the other hand, molecular diffusion along the channel-width direction take place, thus, solute molecules dissolved in one solution phase can cdistributed to the other phase : diffusional mixing, while the efficiency is insufficient. It is supposed that the flow characteristics might be influenced by the shape of the channel. In this study, therfore, shape effects of microchannel on a mixing of two miscible solutions and liquid/liquid extraction of two immiscible solutions were explored by usin spatially-resolved microspectroscopy. Polymer-substrate chips having a flat-side-walled channel (symmetrical and unsymmetrical zigzag side walled) were fabricated. In the unsymmetrical zigzag-side walled channel, the interface was sinusoidal reflecting on the shape, while it was flat in flat-or symmetrical-side-walled channel. Since the interface shape affects on contact time of the two solutions, the unsymmetrical zigzag-side walled channel syowed the most efficient mixing or extraction efficiency. In the symmetrical zigzag channel. the efficiency become worse than in flat-shape channel because of retention flow. These results were also supported fro, flow analysis by mathematical calculation.

  • 人工ナノメートル空間中液相分子の高速分光法および反応ダイナミクス制御法の開発

    本研究では、溶液を特長づけるの最小構成単位と考えられているクラスターの反応ダイナミクス計測を光熱変換分光法とマイクロチップ技術の利点を活用して実現することを目指している。溶液クラスターの大きさの数倍から数百倍の大きさと考えられる10〜数100nmの微小空間に液体を閉じこめ、測定することより、通常の大きさでは困難なバルク液体との情報を分離することを着想した。
    1.ナノ空間の作製 ナノ空間の作製には、電子線リソグラフィー・高速原子線加工法を用いた。また、溶液の導入路として幅190μmのチャネルをサンドブラストエッチングで作製した。ナノ空間として完成させるために、厚さ170μmの石英基板をふっ酸接合法にて接合した。溶液の導入には毛細管現象を利用する方法を利用した。ナノ空間として、幅330nm・深さ220nmの流路(330nm空間)・幅850nm・深さ620nmの流路(850nm空間)を、対照のバルク空間として幅250μm・深さ100μmの流路(250,000nm空間)を測定に用いた。
    2.蛍光寿命測定と空間サイズ効果 ローダミンB(RB)水溶液、スルホローダミン101(SR101)水溶液、ローダミン6G(R6G)水溶液を測定した。どの色素の場合もサイズに依存して波形が変化した。RBの場合には空間が小さくなるほど寿命が長くなり、SR101・R6Gの場合には、330nm空間のみ寿命が短くなることがわかった。寿命のサイズ依存は溶液の濃度にはほとんど依存せず、人工ナノ空間の溶媒物性を示していると考えられる。過去の文献等の知見より、SR101・R6Gの寿命の変化は誘電率変化に、RBの寿命変化は粘性変化に対応づけられると考えた。この変化の原因について、ガラス壁面の電荷などの影響により溶液クラスターの性質が顕在化してきているのではないかと考えられる。
    ナノメートルサイズの空間を人工的に加工し、その中で溶液の分光測定した例はこれまでになく、世界にさきがけナノ空間の液相化学が実現できたといえる。

    我々は光反射配置のフェムト秒過渡反射法を用いて、これまで観測された例のない超高速な電荷移動過程を見いだしてきた。この過程は、フェムト秒光パルスにより電極内部に生成した非平衡状態の高エネルギー電子が関与していると考えられる。より詳細な検討のためには、電子を受容するような化学種等で電極を修飾する等の実験が有効である。しかし、これまでの光反射配置では、高い表面精度が求められるために化学修飾電極等の測定が困難であった。そこで本研究では、高い表面精度が必要なく、電極表面での電子移動・エネルギー移動過程などを高感度に測定できる新規な分光分析手法としてフェムト秒表面熱レンズ分光法を開発した。
    フェムト秒表面熱レンズ分光法では、フェムト秒時間分解を得るためにポンプープローブ法を用いている。本手法では、過渡反射法など従来の時間分解分光法と異なり、ポンプ光とプローブ光の間に数十ミクロンのピント差を与え制御する必要がある。このためのレンズ対を設計して導入した。また、時間分解能を向上させるために群速度分散を補償するプリズム対も導入した。以上の工夫により、350フェムト秒の時間分解で電極溶液界面を計測できる分光法を開発できた。表面精度のよくない高分子基板上の単結晶金薄膜から、従来の反射法以上の信号雑音比で非平衡状態の電子を観測できた。従来法では、溶液の本質的な揺らぎが原因で溶液中の光路長は5mm以下にする必要があり実験上の制約となっていたが、本手法では、溶液中の光路長を40mm以上としても問題なく電極表面を計測できた。測定感度についても、従来の反射法と比較して2桁から3桁ほど向上した。本研究で開発に成功したフェムト秒表面熱レンズ分光法は、様々な形態の電極に適用でき、電極上の電荷移動・エネルギー移動を高感度に計測できるため我々の発見した超高速電荷移動過程の詳細を検討する上でおおきな利点をもつ手法である。
