    Methods for precisely constructing a β‐sheet assembly with number‐defined strands in solution remains quite limited due to its intense aggregation property. Here, we report the precise construction of a four‐stranded anti‐parallel β‐sheet by utilizing a non‐covalent approach. This was achieved by folding and assembly of Ag+ and a pentapeptide (1) with the Ala‐D3pa‐Gly‐3pa‐Val (3pa: β‐(3‐pyridyl)‐alanine) sequence, which was designed to form an interlocking Ag2(1)2 ring through metal cross‐linking of the side chains. NMR analyses and X‐ray crystallographic studies characterized the structure of the discrete β‐sheet assembly as well as the remarkable structural selectivity in terms of strands’ number, orientation and the sheet type.

    分子の集合体形成によって得られる様々なナノサイズの空隙(ナノ空間)を構築する自己集合化学と、アミノ酸配列に基づくペプチド鎖のとりうる配座とその機能解明を行うペプチド化学を融合させることにより、人工酵素の化学合成を目標に研究を実施している。すなわち、空間的に仕切られた人工のナノ空間内に、さまざまなアミノ酸側鎖や活性中心となる金属イオンを精密集積させた人工ナノ空間を独自に構築し、特異な化学反応場の構築を目指している。 本年度の研究では、八残基のペプチド配位子とヨウ化亜鉛の錯形成によって人工βバレル錯体を合成し、その溶液構造の解明を行った。まず、錯形成により固体状態での精密なβバレル構造形成を確認した。しかし各種溶媒に不溶であったため、ペプチド配列へジエチレングリコール鎖を複数導入することを検討した。これにより、βバレル錯体の可溶化とNMR測定による溶液状態の観測に成功した。DOSY NMR測定および結晶構造解析を共に実施することにより、βバレル構造が6本鎖と8本鎖の2種の構造の平衡状態にあることを見出した。以上により、本手法で構築したβバレル構造は、天然にみられる剛直な構造と異なり、ユニークな動的構造特性をもつことを明らかにした。

    A porous peptide crystal, spontaneously self-assembled by simple mixing of a short peptide and a metal ion, has a precise nano-cavity via the facile synthesis. In this work, we succeeded in syntheses of a series of such porous crystals from 1.5 to 2.8 nanometer-sized cavities via chemical modifications of peptides, and revealed their molecular inclusion properties based on their cavity sizes. Furthermore, we also succeeded in creation of uniquely entangled molecules and a precise mimic of natural β-barrels via peptide sequence designs.

    本研究では、超分子化学によるものづくりと生命分子の科学の融合領域を開拓し、生命分子に潜在する動的秩序形成を解明するための人工系ー生命系のハイブリッド材料を創出した。超分子錯体の合成技術をベースとし、生命分子であるオリゴペプチドに備わっている動的秩序形成を引き出し、新たなナノ材料の構築を行った。
    まず、三角形有機配位子(L)とパラジウムイオン(M)の水中での自己集合により得られる自己集合性錯体において、三角形有機配位子の設計を工夫することで、M18L6組成の六面体カプセル錯体を合成した。このカプセル錯体は、ゲストとなる有機分子を添加することにより、ゲスト分子を包接したM24L8組成の正八面体カプセル錯体へ動的変換されることを明らかにした。六面体ー八面体の構造変換のおこる、ユニークな動的超分子錯体の創出に成功した。
    さらに、柔軟なペプチドを素子として組み込むアプローチも行った。Pro-Gly-Pro配列のペプチドの両端に配位性のピリジル基を導入した配位子(L)と銀イオン(M)を自己集合させることにより、M12L12組成の球状の集合体の構築に成功した。得られた構造は、M3L3組成の大環状構造が4つ、知恵の輪状に複雑に絡まり合った構造であることを明らかにした。このとき、複雑さを示す交差数12は合成分子トポロジーの中で最高記録であり、柔軟な生命素子であるペプチドと人工系の配位結合を利用した自己集合を融合させることで、既存の合成手法では達し得ない複雑な分子構造を容易に作り出せることを明らかにすることが出来た。
    以上、本研究では、動的な超分子カプセル錯体の構築と、ペプチドと金属イオンからなる人工系ー生命系ハイブリッド分子の創出を達成した。

    In this work, a new artificial peptide-based nanomaterial was created by the strategy of coordination-driven self-assembly with concomitant folding and assembly processes. A tripeptide with coordinating groups at both termini was designed and successfully complexed with silver(I) ion to give a crystalline porous material. The nanochannel of this material was composed of polyproline II helix of the peptide ligand and its diameter reached more than 2-nm-size. Furthermore, unique molecular recognition properties of this new material were also revealed.

    This research project was intended to the integration of solution-state chemistry and solid-state chemistry through the nano-cavities of self-assembled hollow complexes. New chemistry was established by the successful integration of those two fields, which have been developed independently so far. New mechanistic insights on molecular recognition events or reactions in solution state were visualized by the single crystal X-ray analyses of the guest inclusion complexes. Furthermore, topochemical control in thermal and photochemical reactions was achieved in solution when substrates are well reorganized in the cavity before the events.

    生体内では、短いヌクレオチドやアミノ酸が水中で、特異的な水素結合することにより、精密な分子認識や酵素反応を実現している。しかしながら、水中でそれらを単純に混合するだけでは、無数の水分子により水和を受けて、特異的に水素結合を形成することは起こりえない。そこで、本研究では、水中で短いヌクレオチドを人工的な疎水性空間内に包接することにより、特異的に水素結合対を形成させることを目標としてきた。
    前年度までの研究では、水中において、かご状錯体の疎水性内部空間へのAとUのモノヌクレオチドのペア包接と水素結合対の形成を達成し、論文として報告していた。平成21年度の研究では、異なる配列のGとCのヌクレオチドを用いて水素結合対の誘起を実現し、全4種類の核酸塩基配列の挙動を解明した。GとCの配列のモノヌクレオチドをかご状錯体内にペア包接させ、水素結合対の形成挙動をNMR測定およびX線結晶構造解析から明らかにした。その際、これまでのAとUの塩基対と異なる挙動も明らかにした。特に、GとCの塩基対形成では3本もの水素結合が関与するため、人工疎水場中での塩基対形成に顕著な選択性が現れた。これにより、本研究課題の掲げる、「人工疎水場を利用した水中での水素結合の誘起」という概念を実験的に検証出来たと言える。また、核酸のみならず、本手法を短いペプチド分子に対して応用することで、人工疎水空間を利用したヘリックス構造へのフォールディングも達成し、論文として報告した。
    以上、本研究課題では水中での水素結合形成に関する基礎的な知見のみならず、水中で人工的な疎水性空間を用いた生体分子の精密な構造制御の手法も確立出来たと言える。今後、本研究を基に、長いオリゴヌクレオチドの配列認識やタンパク質の再生技術などへの発展が期待できる。
