    To gain insight into the host functions of a nanocavity encircled by both polyaromatic panels and heteroatoms, nitrogen-doped polyaromatic capsules were successfully synthesized from metal ions and pyridine-embedded, bent anthracene-based ligands. The new capsules display unique host-guest interactions in the isolated cavities, which are distinct from those of the undoped analogues. Besides the inclusion of Ag+ ions, the large absorption change of fullerene C60 and altered emission of a BODIPY dimer are observed upon encapsulation by the present hosts. Moreover, the N-doped capsule exhibits specific binding ability toward progesterone and methyltestosterone, known as a natural female and synthetic male hormone, respectively, in water.

    本研究では、「超硫黄分子による転写制御」を主軸に、超硫黄分子の中でも新たに生体分子としての機能が期待されている環状化硫黄S8に着目して、超硫黄分子依存のシグナル伝達におけるS8の役割を明らかにする。
    生物にとって毒である硫化水素は、生体内で生合成されて様々な生理機能の調節にも関わる「諸刃の剣」である。硫化水素による生体機能の制御では、ポリスルフィド化合物である「超硫黄分子」がシグナル分子の実体として機能する。一方で、申請者の研究を含めたほとんどの超硫黄分子関連研究では、細胞内の主要な超硫黄分子だと考えられてきたシステインパースルフィドやグルタチオンパースルフィドを主体とした有機超硫黄分子にのみ着目して解析が行われてきた。しかし、最新の研究から、無機硫黄分子であるS8が、様々な生体機能に深く関わることが示唆されてきた。したがって、超硫黄分子による生命機能の制御を真に理解するためには、これまでの知見の中での無機超硫黄分子S8の位置づけを明確化することが必要不可欠である。
    真核生物では馴染みのないS8だが、細菌では硫化水素酸化過程において、S8やポリ硫黄 (Sn)を主成分とした“硫黄顆粒”と呼ばれる直径0.5~5 μmの顆粒が細胞内あるいは細胞外に産生されることが知られている。そこで、申請者が細菌を用いた研究から明らかにした超硫黄分子応答性の転写因子SqrRの作用機序や超硫黄分子との関係性を踏まえて、1)細胞内での超硫黄分子の合成場所とS8への代謝過程、および、2)S8の細胞外排出経路と硫黄顆粒の形成機構を解明することで、超硫黄分子によるシグナル伝達の分子機構の詳細解明を試みる。得られる成果は、超硫黄分子から生体機能を理解するための重要な指針となり、これまで見過ごされてきたS8による新たな生理機能の発見を可能にすることが期待される。
