ABSTRACT

Tris (2,4,6‐trichlorophenyl)methyl (TTM) radicals are promising scaffolds for fluorescence imaging (FI) and magnetic resonance imaging (MRI) owing to their long‐wavelength emission and intrinsic paramagnetism. Incorporating donor‐acceptor (D‐A) architectures into TTM radicals has been an established strategy to enhance the brightness and photostability of TTM radicals. However, conventional D‐A‐type TTM radicals exhibit strong charge‐transfer (CT)‐dominated emission that is easily quenched in water due to accelerated non‐radiative decay known as the energy‐gap law. Here, we report water‐soluble TTM radicals (TTM‐ tris TP3B and TTM‐ tris TP9B) which exhibit hybridized locally excited and CT character through rational modulation of the donor units and their substitution positions. New radicals exhibited high photostability and deep‐red emission with photoluminescence quantum yields of 1.4% (TTM‐ tris TP3B, 690 nm) and 1.1% (TTM‐ tris TP9B, 685 nm) in water. Their dual functionality as both fluorescent probes and MRI contrast agents was demonstrated. TTM‐ tris TP3B was taken up by living cells, localized near the cell nucleus, and showed fluorescence. The new radicals also provided a clear longitudinal relaxation time ( T 1 )‐weighted MRI contrast. This work represents the first demonstration of trityl radical functioning as practical FI/MRI dual‐modal imaging probes, establishing a molecular platform without any surfactants or metals in which the fluorophore and electron‐spin source are unified.

ABSTRACT

Quantitative pH imaging is important for understanding pathological tissue environments and diagnosing diseases such as tumors and inflammation. MRI‐based pH quantification has been developed based on several approaches including chemical exchange saturation transfer and ratiometric probes. However, existing approaches involve trade‐offs in sensitivity, probe complexity, and reversibility, and robust quantitative pH imaging remains challenging. Previously, we reported ratiometric MRI probes based on a simple core–shell architecture in which a gadolinium‐loaded core and a pH‐responsive hydrogel shell independently contribute to longitudinal relaxivity ( r 1 ) and transverse relaxivity ( r 2 ), respectively. These probes enabled pH quantification using r 2 / r 1 ratiometry although their pH responses were not optimized for the physiological pH range and their usable probe concentrations were limited. Here, we optimized the crosslinked network of the shell using tetraethylene glycol dimethacrylate, achieving substantial pH‐dependent changes in r 2 around physiological pH while preserving r 1 and extending the usable probe concentration range. Electron microscopy confirmed the core–shell structure and elemental distribution of the probe. The resulting probe enabled quantitative pH imaging over pH 6.0–7.5 across probe concentrations from 0.5 to 1.5 wt.%. These findings establish a practical strategy for MRI‐based pH quantification, offering a step toward standardized non‐invasive pH measurements.

Abstract

Efficient delivery of boron is a critical determinant of the therapeutic efficacy of boron neutron capture therapy (BNCT). However, conventional boron-loaded liposomal systems often suffer from low encapsulation efficiency (EE), resulting in a significant loss of costly 10B-enriched compounds and limited practical applicability. In this study, we established a remote loading strategy for the efficient encapsulation of boron compounds into liposomes. By employing a transmembrane ion gradient, boron-encapsulated liposomes (BLPs) were prepared with an encapsulation efficiency of 42.1% and a boron-to-phospholipid (B/P) ratio of 2, demonstrating the feasibility of remote loading for boron delivery. To further enhance liposome–cell interactions, BLPs were modified with cancer cell membrane interfaces derived from MDA-MB-231 cells. The resulting CM-BLPs exhibited significantly enhanced cellular association, achieving a boron accumulation of 125 ng[B]/106 cells, and showed stable retention without detectable leakage under physiological conditions. In vitro BNCT irradiation experiments revealed that CM-BLPs exhibited greater therapeutic efficacy than both unmodified BLPs and 4-borono-l-phenylalanine (BPA). These results demonstrate the feasibility of remote loading as a practical and scalable strategy for boron delivery in BNCT with cell membrane interfaces serving as a complementary approach to enhance cellular interactions.

ABSTRACT
Reactive Oxygen Species (ROS), which play a critical role in cellular signaling pathways, are one of the chemical products caused by ultrasound irradiation. In general, ROS generation has been ignored in terms of high‐frequency ultrasound (>5 MHz) due to the limitation of cavitation induction. However, does high‐frequency ultrasound not affect the production of ROS in cells? Here, we propose comparing ROS in solution and intracellular ROS generation by ultrasound at high frequency (6.5 MHz) and at a common frequency (1 MHz) in medical ultrasound applications, thereby elucidating the capabilities of high‐frequency ultrasound on cells by ROS production. We discovered that 6.5 MHz ultrasound, as irradiated by a lithium niobate transducer, generates intracellular ROS more than conventional 1.0 MHz ultrasound. The phenomenon of ROS production by high‐frequency ultrasound having high spatial resolution demonstrated the possibility that ultrasound is capable of effectively generating intracellular ROS at the cellular level.

Glucose sensors for NMR relaxometry and magnetic resonance imaging (MRI) can be used for the direct measurement of glucose in turbid biological specimens. Here, we proposed a magnetic glucose sensor based on superparamagnetic iron oxide (SPIO) nanoparticles conjugated to a mannopyranoside derivative and concanavalin A (ConA). The binding of mannopyranoside groups to ConA produced a nanoparticle cluster that was dissociated by competitive binding of glucose to ConA, resulting in changes in the transverse relaxation time (T2) in a glucose-dependent manner. The sensor gave rise to significant T2 changes in physiological glucose levels of 3 - 8 mM at a nanoparticle concentration of 0.5 nM. Significant T2 responses were observed within 6 min of 5 mM glucose detection. Sensor-based MRI by a benchtop 1 tesla scanner permitted a measurement of multiple samples within 8 min. These results demonstrate that the relaxometric glucose sensor could lead to high throughput direct assay of blood samples by using a compact MRI scanner for point-of-care testing.

Cyclic RGD (cRGD) peptide-conjugated boronated albumin was developed to direct toward integrin αvβ3, which overexpresses on many cancer cells. A stepwise conjugation of c[RGDfK(Mal)] and maleimide-conjugated closo-dodecaborate (MID) to bovine serum albumin (BSA) afforded cRGD-MID-BSA, which was noncytotoxic toward both U87MG and A549 cells. As compared with l-BPA, selective antitumor activity of cRGD-MID-BSA toward U87MG cells overexpressing integrin αvβ3 was identified after thermal neutron irradiation. In vivo fluorescence live imaging of Cy5-conjugated cRGD-MID-BSA and MID-BSA revealed that both cRGD-MID-BSA and MID-BSA similarly reached the maximum accumulation during 8-12 h after injection. The selective accumulation and retention of Cy5-cRGD-MID-BSA was more pronounced than Cy5-MID-BSA after 24 h. An in vivo boron neutron capture therapy (BNCT) study revealed that the cRGD peptide ligand combination enhanced accumulation of MID-BSA into tumor cells in U87MG xenograft models. The significant tumor growth suppression was observed in U87MG xenograft models at a dose of 7.5 mg [10B]/kg after neutron irradiation.
