    A new form of micelle, which is composed of detergents with bent aromatic panels, has been created by Michito Yoshizawa and his colleagues at Tokyo Institute of Technology. Unlike traditional micelles, the new ‘aromatic micelles’ are photoactive, and capable of encapsulating dye molecules and showing unusual fluorescence in aqueous solutions. “The present micelles might be suitable for potential applications in the fields of photofunctional dyes, sensors, and materials owing to their ability to accommodate dye molecules and their efficient host-guest energy transfer in aqueous media,” explain the researchers. They also emphasise the straightforward synthesis, aqueous green chemistry and high stability of the aromatic micelles.

    Michito Yoshizawa, Zhiou Li, and collaborators at Tokyo Institute of Technology synthesized ~1 nanometer-sized molecular capsules with an isolated cavity using green and inexpensive zinc and copper ions. In sharp contrast to previous molecular capsules and cages composed of precious metal ions such as palladium and platinum, these nanocapsules emit blue fluorescence with 80% efficiency.Molecular nanocapsules have potential applications as photo-functional compounds and materials but so far molecular capsules synthesized by incorporating palladium ions and so on exhibit poor fluorescence. The researchers expect to be able to prepare multicolor fluorescence composites by the simple insertion of appropriate fluorescent molecules into the isolated cavity of the nanocapsules.

    東京工業大学資源化学研究所の吉沢道人准教授らは、溶液中でバラバラに存在する芳香族分子をナノメートルサイズ(用語1)の空間内で、「数」と「向き」をそろえて正確に積み重ねることに世界で初めて成功した(図1)。すなわち、自己組織化により組み上がった箱型化合物(図2)のナノ空間を活用することで、3つの極性芳香族分子(図3;用語2)の選択的な集積化を達成し、しかもそれらが集積体の双極子モーメント(用語3)をキャンセルして、120度ずつずれて配向することを明らかにした(図4)。さらに、この三重集積体では、個々の分子では見られない特異な分光学的性質が観測された。この分子集積法では、必要成分を加熱・撹拌するだけと簡便かつ汎用性が高いため、未開拓な分子間相互作用の誘起や新規な機能性材料開発に威力を発揮することが期待される。

    An expanded metalloporphyrin-based "black dye" Au-Pd oxohexaphyrin (AuPd-1) with absorption capabilities across the visible-to-near-infrared (NIR) range was synthesized. This black dye, AuPd-1, possessed efficient light-harvesting and photostable capabilities, which were indicative of superior photothermal (PT) conversion abilities. Encapsulation of AuPd-1 with a micellar nanocapsule resulted in a compound that demonstrated intense photoacoustic (PA) properties in the NIR region in water. This finding indicated how metal (d)-ligand (π) molecular orbital interactions in metalloporphyrins could aid in the design of visible-to-NIR light-harvesting black dyes for PT and PA applications.

    本年度は、有限な「芳香環カプセル」の優れた分子捕捉能を維持しつつ、それらの金属イオンによるクロスリンクで、無限の多空間骨格を有する「芳香環カプセル固体」の作製に挑戦した。その結果、カプセル外部に金属配位点を有する外面配位型のM2L4組成の芳香環カプセルの構築と外面配位型の芳香環カプセルの金属配位結合によるクロスリンクを達成した。まず、湾曲型3座ピリジル配位子は根岸・鈴木-宮浦カップリング反応を含む計3段階の反応で得た。次に、外面配位型のM2L4組成の芳香環カプセルの合成は、この新規配位子と金属イオンを加熱混合することで達成した。それらの構造は、NMRおよびMSで明らかにした。さらに、M2L4組成のクロスリンク芳香環カプセルの構築は、得られた外面配位型芳香環カプセル溶液に対して、2当量の金属イオンを加え加熱混合することで達成した。このクロスリンク芳香環カプセルは金属イオンの種類に依存して、シート状構造および粒子状構造を取ることをAFMで明らかにした。

    本年度は、高い集合安定性の芳香環オリゴマーミセルの構築を行った。2つのアントラセン環パネルを有する湾曲型両親媒性分子AAは、水中で5分子から成る芳香環ミセルを形成する。しかしながら、その希釈に対する安定性は中程度(臨界ミセル濃度(CMC)=1mM)で、高温条件で容易に解離する。そこで本研究では、超高希釈と超高温で安定な芳香環ミセルの構築に挑戦した。その戦略として、AA を直線型に「連結」することで「両親媒性オリゴマー」を合成した。実際には、湾曲型両親媒性分子AA の親水基をオリゴエチレングリコール鎖で連結した。連結鎖はパネルの集合を阻害しないため、目的の芳香環オリゴマーミセルの構築が期待できる。まず、湾曲型両親媒性分子の3量体 AT の合成と水中での集合体を達成した。次に、集合体の詳細な構造(DLS, AFM解析など)と集合安定性(NMR, UV-vis, 蛍光, ITC解析など)を解明した。

    We have established a methodology for encapsulating water-insoluble nanographenes in water-soluble micellar capsules and transporting the nanographenes to the surface of gold and/or graphite. Particularly, we succeeded to obtain the molecular resolution scanning tunneling microscopy (STM) images of the triangle-shaped C96H30 and the three-leafed C150H42, and hexagonal C222H42. We also succeeded to find the structural changes of the ovalene adlayer on Au(111) at the electrochemical interface and one of the thiolate molecules into the gap site, by the so-called “molecular planting”. In addition to the results related to the adsorption and organization of hydrophobic molecules, we also focused on metal complex molecules such as iron phthalocyanine (FePc). The FePcs-encapsulating capsules exhibit the function as a “nanoreactor” that promotes the oxygen reduction reaction (ORR).

    本研究の目的は、「多環芳香族ナノ空間」を有する金属架橋カプセルを基盤として、その有機配位子に異種の多環芳香族パネルを導入することで、新たな『異方性ナノ空間』を精密構築することである。また、その多環芳香族骨格に囲まれた特異ナノ空間の高い分子内包能を活用した、新空間機能と新空間材料の創出を目指す。これら目的の達成により、本領域研究が目指す独創的な「空間アシンメトリー」の化学の推進に貢献する。これまでに申請者は、アントラセン環を含む湾曲型の有機配位子と金属イオンの錯形成により、多環芳香族パネルに囲まれた約1 nmの孤立空間を有する金属架橋カプセルの構築を達成した。この孤立空間では、π-スタッキング・CH-π相互作用と疎水効果が三次元的および共同的に働くことで、特異機能が発現した。これらの知見を活かして、本研究では金属架橋カプセルの異方性展開を図った。
    「研究成果1:アニソトロピック型ナノ空間の異性化制御」本年度は、2つのナフタレン環を持つ新規な二座配位子を設計および合成した。これらと金属イオンの錯形成で、最大で42個の異性体を持つアニソトロピック型の金属架橋ケージを構築した。この金属架橋ケージにおいて、内包分子の種類に応じて異性化を制御することに成功した。具体的には、フラーレンC60を内包することで、1つの異性体構造に収束することをNMRや結晶構造解析で明らかにした。
    「研究成果2:扁球型ナノ空間によるボウル反転制御」本年度は、金属架橋カプセルの異方的な空間縮小を達成した。2つのアントラセン環をオルトフェニレン鎖で連結した湾曲型配位子を活用して、扁球型の金属架橋カプセルの構築に成功した。また、空間機能を解明した。具体的には、温度可変のNMR解析から、ボウル型のスマネンを1分子内包することでその圧縮効果により、ボウル反転運動の顕著な加速を達成した。

    For the development of selective sensing systems for various biomolecules in water, we designed and prepared polyaromatic nanopockets inspired by biological protein nanopockets. Using the synthetic nanopocket, we succeeded in the selective binding of male hormones such as testosterone from a mixture of male and female hormones in water. In addition, selective recognition of fatty acids with multiple unsaturated units was also achieved by the nanopocket.

    本研究では、多環芳香族パネルを有する動的ケージを新規に構築するとともに、その機能の開拓を行う。まず、配位結合やパイスタッキング相互作用、動的共有結合などを利用して、新規な動的ケージの構築に挑戦する。例えば、湾曲型の多環芳香族パネルにホルミル基を導入した前駆体を合成し、それとアミノ基を持つ架橋分子との脱水縮合で、狙いとする動的ケージを構築する。また、種々の多環芳香族パネルを含む分子パーツの自己組織化で、空間性質の異なる分子ケージも構築する。次に、得られた動的分子ケージを使って、特異な分子内包機能を開拓する。特に、難溶性で分離が困難な高次フラーレンの選択的な内包に挑戦する。さらに、水中で機能する外部刺激応答性の分子ケージやカプセルの開発にも挑戦する。
    本年度は、外部刺激に応答する新規な動的カプセルの構築と機能開拓を検討した。具体的には、湾曲型の多環芳香族骨格に酸化還元活性なフェノチアジン環を導入することで、新たなV型両親媒性分子を合成した。それらは水中で、疎水効果とパイスタッキング相互作用を駆動力に自己組織化することで、新たな球状の動的カプセルを形成した。その三次元構造をNMR, DLS, AFM 分析で解明した。また、動的カプセルの分光学的性質や電気化学的性質を調査した。その結果、酸化によりラジカルカプセルを可逆的に生成できることが判明した。さらに、種々のゲスト分子の内包と外部刺激により放出を達成した。水中で利用できる新規な動的カプセルを構築すると共に、その特異機能を明らかにした。

    本研究の目的は、我々がこれまでに開発した高い分子内包能の「多環芳香族ナノ空間」を有する金属架橋カプセルを基盤として、これと「異方性(アシンメトリー)」を組み合わせることで、新たな『 異方性ナノ空間の化学 』を創出する。具体的な研究課題として、対称性の高いナノ空間を活用して、非対称性ゲスト分子を内包することで、選択的に異方性ホスト-ゲスト錯体を作製する。また、新規な非対称性ナノ空間を配位結合を使って構築し、その空間が持つ特異物性や相互作用などを解明する。本研究領域のA03班において、他の班との共同研究により、「溶液系」と「多環芳香族骨格」と「孤立ナノ空間」を特徴とした独創的な「空間アシンメトリー」の化学の推進に貢献する。本年度は、芳香環に囲まれたナノ空間が、水中、瞬時にかつ100%の収率で、親水性で非対称性なオリゴ乳酸オリゴを内包できることを見出した。また、結晶構造解析や等温カロリメトリーでの詳細なメカニズムの検証から、内包体は多点の分子間相互作用に基づく負のエンタルピー変化により、水中で安定化することが明らかになった。さらに、両親媒性で対称性のオリゴエチレンオキシドが水中・室温で、瞬時かつ定量的に、金属架橋カプセルと結合することを見出した。結合様式は基質の長さに依存し、約3 nmまでのオリゴエチレンオキシドはカプセル内部に包み込まれ、それより長いオリゴエチレンオキシドはカプセル骨格を貫いて結合した。詳細な熱量分析から、この前例のない貫通型ナノ構造体の形成は、カプセル内面とひも状分子の多点相互作用が駆動力と判明した。水中での芳香環ナノポケットと生体関連分子の相互作用を明らかにした。新たな「多環芳香族ナノ空間」の機能を開拓した。

    Selective recognition of saccharides by artificial receptors in water is a challenging goal due to their strong hydrophilicities and complex molecular structures with subtle regio- and stereochemical differences. Here we report the selective and efficient encapsulation of D-sucrose within a coordination-driven molecular capsule from natural saccharide mixtures in water (~100% selectivity and >85% yield). Unlike previous artificial receptors and natural receptors that rely on multiple hydrogen-bonding interactions, theoretical calculations and control experiments indicate that the observed unique selectivity arises frommultiple CH-pi interactions between the sucrose hydrocarbon backbone and the shape-complementary polyaromatic cavity (~1 nm in diameter) of the capsule.

    The development of new fluorescent molecules and molecular assemblies is of importance for the preparation of unique photofunctional materials. In this work, we succeeded in the quantitative formation of new polyaromatic nanocapsules with fluorescent properties in water from V-shaped amphiphilic compounds with various polyaromatic rings. The nanocapsule could encapsulate a wide range of water-insoluble molecules (e.g., fullerenes, carbon nanotubes, and phthalocyanines) in the hydrophobic cavity to give aqueous host-guest nanocomposites in water. In addition, host-guest nanocomposites containing fluorescent dyes (e.g., DCM and Nile red) display unique photofunctional properties in solution and in the solid state.

    AIBN and its derivatives are useful reagents for polymer and organic syntheses that generate radical species on stimuli by light or heat. These radical initiators are unstable so that they should be kept in the dark at low temperature to avoid photochemical and thermal decomposition. In this work, we demonstrated the quantitative encapsulation of the radical initiators by a metal-linked molecular capsule in aqueous solutions. We also revealed the remarkable stability of the initiators toward light and heat in the cavity shielded by the polyaromatic capsule shell. The stabilized initiators could be utilized for the radical polymerization of olefins on spontaneous release of the initiators from the capsule.

    For studying the “Function of Soft Molecular Systems” in one of the “Creation” research groups, we have approached from organic and materials chemistry by the concerted use of “theory” and advanced spectroscopic “experiments”. Our target systems are mainly photo responsive supramolecular materials at different length scale from discrete capsule-like self-assemblies at nanometer scale to molecular liquid materials at bulk scale. The optical information/properties were positively utilized for understanding the formation mechanism of assemblies and the unique function.
    We have achieved (i) multifunctional molecular capsules and tubes, and (ii) optoelectronically-active molecular liquids. And many collaborations with theory and advanced experiments were successfully conducted in the KAKENHI project.

    比較的大きな芳香族分子は非局在化した複数のπ電子を有するため、興味深い分光学的性質を示す。これらを集積したカラム状構造体は、π共役系の三次元的な拡張により、分子単独では観測されない特異な物性を発現する。我々は最近、有機多座配位子と金属イオンの自己組織化により、芳香環の7重集積体を簡便かつ精密に組み上げることに成功した(M.Yoshizawa et al.,JACS,2008,130,5832-5833)。この7重集積体は、上下2つの芳香族分子を有する2つの箱型分子が3次元的にインターロックして、その間に3つの芳香族分子がインターカレーションした構造である。そこで本年度は、7重集積体の"側面官能基化"による1次元自己組織化を達成したので報告する。
    まず、芳香環7重集積体の構成成分である、官能基を導入した新規ピラー状配位子は、Diels-Alder反応により効率良く合成した。次に、合成したピラー状配位子、パネル状配位子、シス位をエチレンジアミンで保護したパラジウム錯体(4:6:12の比率)を過剰量のトリフェニレン存在下、水中で混合することにより、目的とする側面を官能基化した7重集積体を定量的に構築することに成功した。この構造は、詳細なNMRおよびX線結晶構造解析により決定した。その結晶構造では、7枚の芳香環が約3.5Aの面間距離で集積していることから、効果的なπ-π相互作用が働いていることが示された。^1HNMRにおいて芳香環に由来するシグナルは、低濃度でシャープに観測されるのに対して、高濃度では顕著にブロード化した。また、DOSYによる拡散係数の値も大きく変化した。さらに、7重集積体はリオトロピック液晶性を発現した。以上のことから、側面官能基化した7重集積体は、高濃度で分子間スタッキングして、1次元方向に自己集積することが明らかになった。

    「かご状有機金属錯体」は、自己組織化により特異なナノメートルサイズの疎水的空間を内部に形成し、様々な化学反応の場を提供する「分子フラスコ」としてはたらく。本研究では、この特異なナノ空間中における水クラスターおよびアダマンタンの光化学反応メカニズムを赤外および可視分光法で比較解析し、ナノ空間に束縛された水クラスターの水素結合ダイナミクスを明らかにすることを最終的な目的としている。
    これまでにアダマンタンを内包したかご状錯体に対する低温赤外分光解析により、光電子移動によるアダマンタンおよびかご状錯体の構造変化を検出した(Y.Furutani et al., J.Am.Chem.Soc., 2009)。このような構造変化を実時間で捉えるために、2009年3月に異動した分子科学研究所において、新たに時間分解赤外分光計測系を立ち上げた。光受容タンパク質であるバクテリオロドプシンについては、内部結合水のO-H, O-D伸縮振動の変化を含む4000-900cm^<-1>領域での計測を12.5マイクロ秒の時間分解能で実現した。また、アダマンタンからかご状錯体への光誘起電子移動に伴う可視吸収変化を実時間で捉えるために、東工大資源研の藤井正明教授、酒井誠准教授、井上圭一特任助教(現、名古屋工業大学助教)に協力頂いて、266nm紫外光励起、500-750nm可視光プローブのピコ秒過渡吸収計測を行った(A01班内共同研究)。さらに、A03班の須藤雄気准教授との共同研究として、光センサー蛋白質の分子機構について赤外分光計測を行い、3件の論文を発表した。以上の研究により、ナノ空間に束縛された水クラスターの水素結合ダイナミクスだけでなく、蛋白質場での特異な光誘起構造変化による情報伝達機構などについても様々な知見を得た。

    ボトムアップ型の機能性材料開発においては、新しい性質を有する分子の合成とともに、それらをナノメートルレベルで精密かつ簡便に「配置する」技術が重要な鍵を握る。前者は、従来の合成化学的な手法を駆使することで、これまでに大きな発展を遂げてきた。それに対して後者は、1つ1つの分子をつかんで置くことのできない私たちにとって、現在もなお困難な課題である。そこで本研究では、配位結合を駆動力とした有機配位子と金属イオンの自己組織化に着目し、この仕組みをπ共役系分子の有限集積に活用することで、効率的なπ共役系分子の高次有限集積化を達成し、その集積体を用いた新奇な物性の開拓を検討した。平面状金属錯体の代表格である、巨大な色素分子、ポルフィリン類は様々な金属を導入できることから、その高次集積構造に興味が集まっている。しかしながら、3分子以上のポルフィリン金属錯体をカラム状に集積し、複数の金属間で相互作用を誘起させた例はほとんどない。そこで本研究では、伸長したピラー状分子を用い、内部に3分子集積可能な箱型骨格を利用して、3重金属集積体の構築を検討した。その結果、ポルフィリン金属錯体とその類縁体であるアザポルフィリン金属錯体を組み合わせることで、同種の金属錯体の三重集積化だけでなく、2種類の金属錯体を交互集積することに成功した。具体的には、銅-銅-銅、銅-パラジウム-銅および銅-コバルト-銅からなる三重金属集積体を組み上げた。そこで、集積された3つの金属はお互いにコミュニケーションしているか?いないのか?を明らかにするため、電子スピン共鳴を測定した。その結果、銅錯体の三重集積体では、不対電子(=スピン)を持つ3つの銅間でのスピン-スピン相互作用に由来するΔm_s-1〜3シグナルを初めて観測することに成功した。

    ある種の芳香族分子は集積することで、分子単独では見られない特異な性質を示すことが知られている。例えば、芳香族分子が2分子集積した電荷移動錯体や無限集積した液晶などが、有機伝導体や動的機能性材料として盛んに研究されている。しかしながら、その中間に位置する芳香族分子の有限な集積体は、集積する分子の数や種類、順序に応じた特異な物性発現が期待されるにも関わらず、その有効な合成手段すら見出されていない。そこで本研究では、配位結合を駆動力とした有機配位子と金属イオンの自己組織化に着目し、この仕組みをπ共役系分子の有限集積に活用することで、効率的なπ共役系分子の高次有限集積化を達成し、その集積体を用いた新奇な物性・反応性の開拓を検討した。まず、合理設計した有機配位子と金属イオンの自己組織化により、必要な成分を混合するだけで狙いとする有限集積体を一義的に組み上げる。その合成戦略として、集積するn枚のπ共役系分子に対して、その上下に2枚のパネル状配位子を配置し、その距離に応じたピラー型配位子を用意し、それらを金属イオンで繋ぐことで有限集積体が構築できると考えた。実際、上述の合成戦略で4重の集積体が一義的に合成できることを見出した。ここでは、平面状のピリジン三座配位子、直線状のビピリジン配位子、シス位をエチレンジアミンで保護したパラジウム錯体、そしてπ共役系分子としてピレンを水溶液中で混合することで、かご型の4重集積体が定量的に生成した。その構造は、X線結晶構造解析によっても明らかにした。また、内部集積した2枚のピレンは、かご型骨格から容易に抽出でき、再び、他の芳香族分子を2分子集積できることを明らかにした。

    In this project, we have focused on the synthesis and function of nano-scale molecular materials by programmed molecular self-assembly utilizing transition metals. Here we summarize our research results that are divided into the five topics. (1)Self-assembly of endo-functionalized molecular spheres : we succeeded in the formation of a liquid-like fluorous droplet, composed of 24 perfluoroalkyl chains confined in the interior of a 5-nanometer-sized, roughly spherical shell. The structures were spontaneously assembled in solution from 12 palladium ions and 24 bridging ligands. (2)Photo-switchable molecular catenate : We achieved a precise, clean molecular lock that operates by light. Since the inert Pt(II)-py bond is considerably labialized upon UV irradiation, we demonstrated the photo-switchable molecular lock by the one-way catenation of coordination rings. (3)Selective recognition and folding of oligopeptides by self-assembled hosts : We anticipated that the large hydrophobic cavity of self-assembled hosts could replace the protein scaffolds and stabilize the secondary structures of oligopeptides. (4)Unusual chemical reactions within self-assembled hosts : We found that an aqueous self-assembled cage induces highly unusual regioselectivity in the Diels-Alder reactions of anthracene and phthalimide molecules, promoting reaction at a terminal ring. (5)Direct crystallographic observation of unstable transition species within self-assembled hosts : We succeeded in the in situ crystallographic observation of a coordinatively unsaturated metal complex within a self-assembled coordination cage.
