    Electrocatalysts which can operate for several years are required to produce hydrogen and commodity chemicals in an environmentally friendly manner. However, designing materials with long operational lifetimes is challenging, due to the lack of a conceptual framework to predict catalytic lifetimes quantitatively. Here, we report a microkinetic equation which quantifies the lifetime of an electrocatalyst undergoing dissolution. This equation was obtained by taking advantage of the fact that catalysis is much faster than deactivation, which allows the ordinary differential equations to be solved via the quasi steady-state approximation. All chemical reactions were modeled as irreversible, first-order elementary reactions. Under this assumption, the catalytic rate correlates linearly with the deactivation rate, leading to a trade-off relationship between activity and stability. Our model was supported by the correlation between theoretical and experimental lifetimes (r2 = 0.86) of a manganese oxide electrocatalyst during the oxygen evolution reaction.

    Hexavalent iridium (Ir VI ) oxide is predicted to be more active and stable than any other iridium oxide for the oxygen evolution reaction in acid; however, its experimental realization remains challenging. In this work, we report the synthesis, characterization, and application of atomically dispersed Ir VI oxide (Ir VI - ado ) for proton exchange membrane (PEM) water electrolysis. The Ir VI - ado was synthesized by oxidatively substituting the ligands of potassium hexachloroiridate(IV) (K 2 IrCl 6 ) with manganese oxide (MnO 2 ). The mass-specific activity (1.7 × 10 5 amperes per gram of iridium) and turnover number (1.5 × 10 8 ) exceeded those of benchmark iridium oxides, and in situ x-ray analysis during PEM operations manifested the durability of Ir VI at current densities up to 2.3 amperes per square centimeter. The high activity and stability of Ir VI - ado showcase its promise as an anode material for PEM electrolysis.

    The Sabatier principle states that catalytic activity can be maximized when the substrate binding affinity is neither too strong nor too weak. Recent studies have shown that the activity of several hydrolases is maximized at intermediate values of the binding affinity (Michaelis-Menten constant: Km ). However, it remains unclear whether this concept of artificial catalysis is applicable to enzymes in general, especially for those which have evolved under different reaction environments. Herein, we show that the activity of phosphoserine phosphatase is also enhanced at an intermediate Km value of approximately 0.5 mM. Within our dataset, the variation of Km by three orders of magnitude accounted for a roughly 18-fold variation in the activity. Owing to the high phylogenetic and physiological diversity of our dataset, our results support the importance of optimizing Km for enzymes in general. On the other hand, a 77-fold variation in the activity was attributed to other physicochemical parameters, such as the Arrhenius prefactor of kcat , and could not be explained by the Sabatier principle. Therefore, while tuning the binding affinity according to the Sabatier principle is an important consideration, the Km value is only one of many physicochemical parameters which must be optimized to maximize enzymatic activity.

    Amino acids in carbonaceous chondrites may have seeded the origin of life on Earth and possibly elsewhere. Recently, the return samples from a C-type asteroid Ryugu were found to contain amino acids with a similar distribution to Ivuna-type CI chondrites, suggesting the potential of amino acid abundances as molecular descriptors of parent body geochemistry. However, the chemical mechanisms responsible for the amino acid distributions remain to be elucidated particularly at low temperatures (<50°C). Here, we report that two representative proteinogenic amino acids, aspartic acid and glutamic acid, decompose to β-alanine and γ-aminobutyric acid, respectively, under simulated geoelectrochemical conditions at 25°C. This low-temperature conversion provides a plausible explanation for the enrichment of these two n-ω-amino acids compared to their precursors in heavily aqueously altered CI chondrites and Ryugu’s return samples. The results suggest that these heavily aqueously altered samples originated from the water-rich mantle of their water/rock differentiated parent planetesimals where protein α-amino acids were decomposed.

    Understanding how to tune enzymatic activity is important not only for biotechnological applications, but also to elucidate the basic principles guiding the design and optimization of biological systems in nature. So far, the Michaelis-Menten equation has provided a fundamental framework of enzymatic activity. However, there is still no concrete guideline on how the parameters should be optimized towards higher activity. Here, we demonstrate that tuning the Michaelis-Menten constant ([Formula: see text]) to the substrate concentration ([Formula: see text]) enhances enzymatic activity. This guideline ([Formula: see text]) was obtained mathematically by assuming that thermodynamically favorable reactions have higher rate constants, and that the total driving force is fixed. Due to the generality of these thermodynamic considerations, we propose [Formula: see text] as a general concept to enhance enzymatic activity. Our bioinformatic analysis reveals that the [Formula: see text] and in vivo substrate concentrations are consistent across a dataset of approximately 1000 enzymes, suggesting that even natural selection follows the principle [Formula: see text].

    Selective ion transport through nanochannels plays a crucial role in osmotic energy conversion in cellular systems. We show that this cell-essential process occurs in a submarine hydrothermal vent (HV) precipitate derived from a serpentine-hosted geological environment. Plate-like layered nanocrystals are aligned on the nano to millimeter scale, forming confined nanochannels in the HV precipitate. The nanochannels with surface charges function as a cation- and anion-selective ion transport membrane, allowing the precipitates to convert ionic gradients of Na+, K+, H+, and Cl- into electrical energy. Our findings suggest that osmotic energy conversion can occur spontaneously and widely through geological processes, offering valuable insights into the establishment of electrochemically coupled ion transport in early life as well as the creation of self-organized structures in engineering fields.

    In this research, encapsulation of Shewanella oneidensis MR-1 in the phospholipid polymer hydrogel and electrochemical property of the hydrogel with immobilized shewanella was investigated. Cytocompatible and electron transfer mediated polymer based on the water-soluble phospholipid polymer, poly(2-methacryloyloxyethyl phosphorylcholine (MPC)-co-n-butyl  methacrylate  (BMA)-co-p-vinylphenylboronic   acid (VPBA)-co-vinylferrocene (VFc)) (PMBVF) was synthesized by conventional radical polymerization method. Shewanella which have the ability to  generate  and  transfer extracellular electron without any external  mediator,  have  significant  potential  on  the fabrication of microbial fuel cell. The water-soluble PMBVF was dissolved in the cell culture medium containing shewanella cells. Poly(vinyl alcohol) (PVA) solution was added to the PMBVF solution with gently mixing, in which case, a reversible PMBVF/PVA hydrogel with encapsulated shewanella was obtained. The viability of the encapsulated shewanella was calculated by the fluorescence microscopy. The electrochemical property of the PMBVF/PVA hydrogel containing shewanella was measured by cyclic voltammograms (CV) and amperometric i-t curve. PMBVF/PVA hydrogel can  provide  mild  environment  for  the shewanella and maintain the cellular function for longtime. The generated current was higher than that cultured with normal medium.

    本研究では、【実験1】①「浄化能が強化される底質」、【実験2】②「電位操作により浄化が著しく促進される底質」のメカニズムを解明することを目的とする。令和3年度は、主に【実験1】①「浄化能が強化される底質」に取り組んだ。具体的には、養殖場から採取した底質を、ガラス電極(フッ素ドープ酸化スズ(FTO)電極)を用いて電位測定な可能なリアクター内に入れ、有機物を負荷した際の底質電位および底質汚染の重要な指標である酸揮発性硫化物量(AVS)について経時的に観測した。有機物を負荷した際の電位挙動とAVSの関係を詳細に検討した結果、AVSは電位の一時データよりも、積算値(10分間隔で測定した値の合計値)との相関性が高いことが明らかとなった。

    海洋の底質中に蓄積する有毒プランクトンのシストの発芽は、酸素や光など様々な環境因子によって制御されている。本研究ではシストの発芽応答に影響を及ぼす電位の特定および作用機序を明らかにすることを目的としている。昨年度、試験系の参照極から溶出する銀によるプランクトンへの毒性影響が当初の想定以上に高いことが明らかとなった事から、参照極を使用しない2極系の試験系を確立した。本試験系にて培養で形成したプランクトンシストのみを用い、底質を含まない条件にて電位操作試験を実施した結果、電位操作はシストの発芽に影響を及ぼさなかった。このことは、シストが直接的に電位を感知していないこと、また、海水中の酸化還元物質がシストの発芽応答へ影響をおよぼさないことを示唆するものである。底質中に含まれる酸化還元物質および電子伝達物質が発芽応答への影響をおよぼす可能性が残されていることから今後検証する予定である。一方、シストの発芽自体には影響が認められなかったものの、発芽後の一部の栄養細胞の行動に異常が確認された。
    電位操作がプランクトンに与える影響の作用機序の解明に向け、本年度はまずシストの形成から発芽にいたるまでの代謝物をメタボローム解析を用いて同定した。その結果、栄養細胞とシストでは脂肪酸含有量が変化すること、また、シストの形成および保存温度により糖質の含有量が異なる事等が明らかとなり、発芽機序解明に向けシストの代謝物に関する基盤となる情報を得た。

    酸揮発性硫化物量(AVS)を基準とした3段階(AVS:試験区1:0.04 mg/g、試験区2:0.24 mg/g、試験区3:1.21 mg/g)の底質を用いてマダイの底質曝露試験を実施した。なお試験方法は令和2年度に実施したクルマエビ底質毒性試験を参考にした。その結果、試験区3では13日後に、試験区2では20日後に7匹全て死亡した。試験区1は、20日後に7匹中1匹死亡したが、残りの6匹は28日間生残した。この結果から、底質汚染はマダイの生残に重大な影響を及ぼすことが改めて立証された。また、底質AVSは、マダイを収容している全ての試験区に比べ、マダイを収容していないコントロール区で有意に高い値を示した。この結果は、昨年度実施したクルマエビの結果と同様であった。電位操作による毒性試験を実施するため、電位を-300mV、+300mVに設定し、マダイを飼育した。その結果、電位操作の影響は認められなかった。なお、得られた成果は、2021年秋の水産学会で発表した(コロナ感染拡大のため要旨のみ)。

    As a method to introduce structural distortion into Mn oxides, we have studied gamma-type MnO2, which has a microtwinning structure and allows precise control of structural distortion. As a result, it was confirmed that the disproportionation reaction of Mn3+ is suppressed with the introduction of structural distortion, and the overpotential of oxygen evolution is reduced. By optimizing the electrolysis conditions taking into account the potential dependence of the disproportionation reaction, we found that the MnO2 catalyst can stably electrolyze water even in a strong acid environment.

    In-situ electrochemical measurements were carried out on hydrothermal fields in deep-sea and shallow sea. Especially, successful in survey of in-situ electrochemical measurement in alkaline hydrothermal vents was significant to consider about chemical evolution of metabolic pathways. Survey of deep-sea hydrothermal field also sufficiently achieved and understanding was deepened about relationship between electric field, electricity discharge, and minerals. Experiments for reproduction of metabolic pathways in electrochemical cells were carried out in laboratory. The results revealed that catalytic efficiencies of chemical reactions were influenced by types of sulfide minerals and exchange of minerals with electricity.

    To understand the newly hypothesized deep-sea electro-ecosystems, we investigated how electrical current affects the physiology of marine benthos. We found that (1) benthos generates weak electrical current, (2) core metabolisms of benthos is affected by direction of electrical current flow, and also (3) behavior of benthos is also regulated by environmental redox potential. These findings demonstrate the importance of “electrical current” as a new parameter to understand the ecology of deep-sea electro-ecosystems.

    Based on the new hypothesis that the difference in the stability of Mn3+ between biological and artificial Mn catalysts is the origin for the activity difference, we have attempted to design the efficient catalysts for water oxidation under neutral pH. It was demonstrated that the induction of coupled electron-proton transfer is effective strategy to stabilize Mn3+, leading to the large enhancement of the activity. It was also demonstrated that the exposure of metastable faces of and lowering the size of Mn catalysts stabilize Mn3+, thus enhancing the activity. Furthermore, by comparing the mechanisms of novel-metal catalysts, we have successfully generalize our concept of Mn3+, highlighting that the spin state regulation of intermediates is critical to generate next-generation O2 evolution catalysts with abundant 3d-block elements.

    In this project, enrichment of possible electrotrophic microbial communities has been tested under 600 mV redox potential in the laboratory. However, we have not successfully found biomass production of electrotrophic microbial community. Next, it has been conducted in natural deep-sea hydrothermal vent environments. In the Iheya North hydrothermal field, a fuel cell has been deployed for two weeks using platinum-coated titanium electrodes, deployed in a hydrothermal fluid and ambient seawater. During the deployment, generation of 600 mV redox potential has been achieved. After the recovery, dense biofilm has been identified on the surface of cathode. The efficient electron conductivity and the dense biofilm development both point to the possible enrichment and biomass production of microbial community significantly based on the electroautotrophy. This research project reveals the possible existence of electrotrophic microbial community in natural deep-sea hydrothermal environments.

    We exploited the ability of the deep-sea Fe-oxidizing bacteria, Mariprofundus ferrooxidans, to elevate the intracellular electrons into a higher energetic state as a built-in voltage multiplayer circuit. In-vivo electrochemical analysis of CO2 fixation processes revealed that the cells multiplies the voltage to 0.95 V, which enables to initiate the electrochemical CO2 reduction coupled with water oxidation at an external voltage of 0.19 V. The value of 0.19 V is one order of magnitude lower than the thermodynamic limit for producing organic compounds via CO2 reduction coupled with water oxidation. Thus, the integrated bioelectrochemical process could provide a way to utilize extremely low-voltage-electricity supplied by several sources such as hydro, solar, wind, geothermal, and others for the electrochemical conversion of CO2 to organic chemicals.

    The molecular mechanism of extracellular electron transfer (EET) was clarified through the direct observation of the dynamics, leading to the development of the methodologies for reversible regulation and collective activation of cellular metabolisms. It was also revealed that microorganisms with EET ability are widely distributed in natural environments. Furthermore, we successfully applied the concept of the EET to mammalian cells that do not possess EET ability by developing cytocompatible electron mediators. Based on the systematic understandings of the EET obtained through the present study, novel methodologies and perspectives were provided to various fields, including measurement/regulation of circadian clocks and cancer cells, and proposal of new ecosystem supported by electricity.

    To examine the roles of semiconductive minerals in the respiratory activity of microorganisms, we cultured the dissimilatory metal-reducing bacterium, Shewanella loihica PV-4, in an electrochemical cell and examined the influence of cell surface-associated Fe-oxide nanocolloids on the extracellular electron-transfer efficiency. The respiratory current was greatly improved (over 50 fold) when α-Fe_2O_3 were supplemented into the cell cultures. In vivo electrochemical studies have revealed that electrons injected from the cells to the conduction band of colloids to move long distances from cell surfaces without the use or requirement of soluble electron shuttles. This finding suggests the microbial ability to exploit semiconductive minerals for transporting and sharing electrons in anaerobic environments.

    The exposure of titanium dioxide(TiO_2) nanosized particle from TiO_2 photocatalytic materials was less than 1/50^th of permissible dose in NIOSH, suggesting that the risk of nano-particle exposure from the TiO_2 materials is very low level. The photocatalytic materials for soil and water purification were prepared respectively, and it was confirmed that polluted soil and agricultural wastewater were cleaned up by each system with the photocatalytic material in a level of practical use.

    金属酸化物半導体は、光エネルギー変換に適した光触媒として、多くの研究がなされている。本研究では、「表面反応中間体の分光学的in situ直接観察」および「金属酸化物表面構造の原子レベル制御」の2点から研究方法の開拓を進め、これを通じて反応機構の解明を行ってきた。特に光機能界面として最も重要であるTiO_2/水溶液界面における水の光酸化・酸素発生反応に焦点をあて、この反応の機構を原子・分子のレベルで解明を行った。また、新規可視光応答型の光触媒として光誘起MMCTで駆動するヘテロ金属分子光材料の開発も行ってきた。以下に主な結果を述べる。
    (1)新しい表面処理法であるHF浸漬-高温焼成法を用いることで、原子レベルで平坦でかつ安定なTiO_2ルチル表面を作製することに成功した。その結果、表面バンドエネルギーや価電子帯正孔の反応性がTiO_2の表面原子構造配列に依存することを世界で初めて明らかにした。さらに、in situ PL法を用いて、水の光酸化反応の前駆体である表面捕捉正孔を捉え、この原子レベルでの構造やエネルギーの面依存性を明らかにした。
    (2)金属酸化物上での結果を踏まえて、窒素含有・金属酸化物上における可視光照射下での光酸素発生反応の機構の検討を行った。TaON微粒子粉末を用いて薄膜光電極を作成し、種々の還元剤添加による光電流作用スペクトルの変化の詳細な検討から、可視光照射下での水の酸化反応は"電子移動機構"で進行するのではなく、"水分子の求核攻撃機構"によって進行することを明確に示した。
    (3)さらに、多電子移動触媒であるIrO_2、および光合成のPSII中心のモデル触媒としてMn二核錯体の合成を行い、水の酸化反応が上と同様に"水分子の求核攻撃機構"で反応が進むという重要な結果を明らかにしつつある。
    (4)ヘテロ金属からなる無機分子光材料の開発を行った。ヘテロ金属錯体の光誘起MMCTによって、可視光照射下で光触媒反応を駆動させることに成功した
