    Purified DNA plasmids traditionally used for microbial transformation have been supplanted by extracellular plasmids released via host bacterial lysis, offering an alternative approach for DNA-plasmid delivery. Specifically, shuttle vector plasmids liberated from host Bacillus subtilis were directly employed for the transformation of chemically competent cells Escherichia coli, eliminating the need for biochemical purification. This unconventional DNA delivery technique, referred to as 'Cell Lysis Technology to provide Transformable Extra-cellular DNA; CELyTED', has been successfully adapted for the transformation of microorganism Saccharomyces cerevisiae as well. The protocol includes optimized conditions for efficient cell lysis of the donor host cells. Notably, ' CELyTED ' enables the introduction of large-sized DNA plasmids exceeding 50 kb into target microorganisms mitigating the potential adverse effects of physical shearing during the purification process. This simplicity in the delivery protocol makes it versatile for both prokaryotic and eukaryotic microorganisms, establishing a fundamental platform in the synthetic genome field. Our study demonstrates the feasibility of introducing large DNA plasmids into cells E. coli and S. cerevisiae using the lysate of donor host cells, showcasing the potential of 'CELyTED ' as a streamlined approach in genetic transformation methodologies.

    Functional annotation of the vast noncoding landscape of the diploid human genome still remains a major challenge of genomic research. An efficient, scarless, biallelic, and gene-wide mutagenesis approach is needed for direct investigation of the functional significance of endogenous long introns in gene regulation. Here we establish a genome substitution platform, the Universal Knock-in System or UKiS, that meets these requirements. For proof of concept, we first used UKiS on the longest intron of TP53 in the pseudo-diploid cell line HCT116. Complete deletion of the intron, its substitution with mouse and zebrafish syntenic introns, and specific removal of retrotransposon-derived elements (retroelements) were all efficiently and accurately achieved in both alleles, revealing a suppressive role of intronic Alu elements in TP53 expression. We also used UKiS for TP53 intron deletion in human induced pluripotent stem cells without losing their stemness. Furthermore, UKiS enabled biallelic removal of all introns from three human gene loci of ~100 kb and longer to demonstrate that intron requirements for transcriptional activities vary among genes. UKiS is a standard platform with which to pursue the design of noncoding regions for genome writing in human cells.

    [Purpose] To verify the effect of a 12-week additional resistance training intervention in patients on hemodialysis who had been performing supine ergometer exercises alone during dialysis. [Participants and Methods] Overall, 18 patients undergoing hemodialysis were included. A 12-week intervention with additional resistance training was conducted in hemodialysis patients who had been performing supine ergometer exercise for 30 min during dialysis for over >3 months. Physical function before and after the period of bicycle ergometer exercise alone and before and during the additional intervention was compared. Resistance training consisted of 1-3 sets per day of five different exercises for the large muscle groups of the upper and lower limbs. [Results] The results of the 6-min walk test improved significantly after the additional intervention. The average driving distance in the supine ergometer exercise during the additional intervention was identified as an associated factor. [Conclusion] The addition of the resistance training to long-term supine ergometer exercisers improved walking endurance. When supine ergometer exercise alone does not change physical function, the additional use of resistance training subsequently may have a positive effect on walking endurance.

    BACKGROUND: Japanese cedar (JC) pollinosis is a common allergic rhinitis in Japan. JC pollen sublingual immunotherapy (SLIT) tablets are licensed for the treatment of JC pollinosis. OBJECTIVE: To assess the disease-modifying effects of JC pollen SLIT tablets over 5 years (2014-2019), comprising a 3-year treatment period and 2-year follow-up. METHODS: A total of 1042 patients with JC pollinosis (aged 5-64 years) were included in the study. An optimal dose-finding study was performed in the first 15 months, after which 240 patients in the placebo (P) group and 236 patients in the optimal active dose (A) group (5000 Japanese allergy units) were re-randomized to receive P or A for an additional 18 months (designated AA, AP, PA, and PP groups). Clinical efficacy was evaluated by the total nasal symptom and medication score (TNSMS) during the peak symptom period of each JC pollen season over 3 years of treatment and 2 years of observation after treatment cessation. RESULTS: The AA, AP, and PA groups exhibited significantly reduced TNSMS; however, the largest relative reduction was seen in the AA group both during the treatment period (third season, 46.3% vs PP, P < .001) and during the 2-year follow-up period (fourth and fifth seasons, 45.3% and 34.0% vs PP, respectively; P < .001). The most common adverse drug reactions were mild reactions at the administration site. CONCLUSIONS: JC pollen SLIT tablets show sustained clinical efficacy during 3 years of treatment and sustained disease-modifying effects for at least 2 years after treatment cessation.

    BACKGROUND: There have been no reports of treatment effect persistence after long-term sublingual immunotherapy (SLIT) in patients with Japanese cedar (JC) pollinosis. Therefore, we conducted a post-marketing clinical trial to investigate the efficacy, safety, and effect persistence of JC pollen SLIT drops after approximately 3 years of treatment. METHODS: This was an open-label trial of 233 patients with JC pollinosis who were treated with JC pollen SLIT drops for approximately 3 years (2015-2017) and followed-up for an additional 2 years (2018-2019). Efficacy and effect persistence were evaluated using nasal and ocular symptom scores, daily use of rescue medication, and Japanese Rhinoconjunctivitis Quality of Life Questionnaire scores recorded during the JC pollen dispersal season of each year. Safety was evaluated by monitoring adverse events and adverse drug reactions. RESULTS: The mean combined total nasal symptom and medication score (range 0-18) during the peak symptom periods of 2015 through 2019 were 5.47 ± 3.38, 4.52 ± 3.13, 3.58 ± 2.63, 5.28 ± 4.01, and 6.83 ± 4.65, respectively. The percentage of patients who used no rescue medications during the same periods was 64.8%, 75.2%, 80.3%, 63.7%, and 50.3%, respectively. A total of 138 adverse drug reaction incidents were recorded in 73 of the 233 patients (31.3%), of which 134 incidents (97.1%) were mild in severity. CONCLUSIONS: JC pollen SLIT drops demonstrated treatment duration-dependent efficacy with effects that persisted for 2 years after cessation of treatment. The drug had a favorable safety profile over the 5-year study period.

    Achondroplasia (ACH) is the most common form of short-limbed skeletal dysplasia. Patients with ACH sometimes undergo lower limb lengthening to get functional and psychological achievements. The periosteal resection (PR) is a known mechanism to increase longitudinal bone growth without osteotomy, although the results are not predictable. It could be alternative for limb lengthening in a minimally invasive technique. The purpose of this study is to evaluate the effect of PR on acceleration of bone growth in a mouse model of ACH (Fgfr3 ach). We performed a circumferential resection of periosteum on the proximal tibia to both wild-type and Fgfr3 ach mice at the age of four weeks. The second PR was done one week later in each mouse, which was subsequently sacrificed at the age of six weeks for micro-computed tomography (micro-CT) scan and histological examinations. We measured tibial bone length, bone volume, and metaphyseal trabecular bone parameters, including bone volume/tissue volume (BV/TV), trabecular thickness (Tb.Th), trabecular number (Tb.N) by reconstructed micro-CT images. We also quantified the entire width of the growth plate of the proximal tibial from the sections stained with hematoxylin and eosin. Tibial bone length and bone volume of the PR side were significantly larger than the sham side in wild-type mice, while they were not statistically significant in Fgfr3 ach mice. The BV/TV and Tb.N in the metaphysis were significantly decreased in the PR side of both mice. The histological analysis revealed that the growth plate of the proximal tibia was significantly wider in the PR side of wild-type mice while it showed no difference in width between the PR side and the sham side in Fgfr3 ach mice. PR promoted longitudinal bone growth in wild-type mice, but it exhibited only a marginal effect on bone growth in Fgfr3 ach mice.

    PURPOSE: The most appropriate procedure and at what type and stage of osteonecrosis of the femoral head (ONFH) these procedures had been argued. We attempted to clarify the trend in surgical operations with respect to the age of patients, type classification, and stage of ONFH over a period of 15 years by using the multi-center sentinel monitoring system in Japan. METHODS: We evaluated the hips of 3844 patients using this system in three phases of every five  years from 2003 to 2017. We classified the surgical procedures as osteotomy (OT), hemiarthroplasty (Hemi), and total hip arthroplasty (THA). We assessed the trend in age, type classification, and stage of ONFH over three time periods; "early," and the "late." We calculated the proportion of surgeries for ONFH in each period. We used the Cochran-Armitage test to evaluate trends in proportion of two levels of characteristics across three time periods. RESULTS: The proportion of younger patients significantly decreased. The proportion of OT and Hemi decreased over time, while the proportion of THA increased. The proportion of patients with types C1 and C2 who underwent OT and Hemi decreased over time. In contrast, that of THA increased. The proportion of patients who underwent OT and Hemi significantly decreased; the proportion of patients who underwent THA significantly increased over time at all stages. CONCLUSIONS: In Japan, the younger patients underwent surgery for ONFH decreased. The patients who underwent OT and Hemi for ONFH decreased, while that of THA increased over time.

    BACKGROUND: There have been no studies of dual administration of sublingual immunotherapy (SLIT) tablets for perennial and seasonal allergic rhinitis. This trial (JapicCTI-184014) was conducted to investigate the safety profile and immunological response during dual therapy with SQ house dust mite (HDM) and Japanese cedar pollen (JCP) SLIT tablets. METHODS: This was a multicenter, open-label, randomized trial of 109 Japanese patients with coexisting HDM and JCP allergic rhinitis who had positive tests for HDM- and JCP specific IgE (≥0.7 kU/L). Patients were allocated to receive HDM (N = 54) or JCP (N = 55) SLIT tablets alone for 4 weeks followed by 8 weeks of dual therapy with both SLIT tablets administered within 5 min of each other. Adverse events (AEs), adverse drug reactions (ADRs), and serum IgE and IgG4 specific for HDM (Dermatophagoides farinae, Dermatophagoides pteronyssinus) and JCP were recorded. RESULTS: The percentage of subjects with AEs and ADRs was similar between the two groups and between the two periods of monotherapy and dual therapy. Most AEs and ADRs were mild in severity, and no serious events were observed. The most common ADRs were local events in the oral cavity. Levels of IgE and IgG4 specific for HDM (D. farinae, D. pteronyssinus) and JCP were increased after treatment with HDM and JCP SLIT tablets, respectively. CONCLUSIONS: Dual therapy with both SLIT tablets administered within 5 min after 4 weeks of monotherapy with HDM or JCP tablet was well tolerated and induced the expected immunological responses.

    BACKGROUND: We conducted a randomized, placebo-controlled, double-blind clinical trial to investigate the optimal dose and long-term efficacy and safety of Japanese cedar (JC) pollen tablets for SLIT (JapicCTI-142579). Here, we report details of the effects of the JC pollen SLIT tablet on rhinitis and conjunctivitis symptoms over three pollen dispersal seasons. METHODS: A total of 1042 JC pollinosis patients (aged 5-64 years) were randomized to receive tablets containing placebo (P), 2000, 5000, or 10,000 Japanese allergy units (JAU) of JC pollen for 15 months to identify an optimal dose. Patients receiving P (n = 240) and the optimal dose (5000 JAU; A, n = 236) were then randomized to receive P or A for an additional 18 months (AA, AP, PA, and PP groups, allocation ratio 2:1:1:2). Nasal and ocular symptoms, rescue medication use, and quality of life (QOL) were assessed on quantitative scales. RESULTS: In the second and third seasons, the AA, AP, and PA groups exhibited significantly better improvements in nasal, ocular, and medication scores compared with the PP group in the order AA > AP > PA > PP during the second season and AA > PA > AP > PP during the third season. Rescue medication use and QOL scores were also significantly better in the AA, AP, and PA groups compared with the PP group. CONCLUSIONS: The JC pollen SLIT tablet relieved nasal and ocular symptoms and medication use and improved QOL in a treatment duration-dependent manner. Continuous dosing regimens appear to enhance the efficacy of the drug.

METHODS: A total of 1042 patients with JC pollinosis (aged 5-64 years) were equally randomized into 4 groups and received daily treatment with 2,000, 5,000, or 10,000 Japanese allergy unit (JAU) or placebo. The primary end point was the total nasal symptom and medication score (TNSMS) during the peak symptom period in the first season. Key secondary end points were TNSMS for the JC pollen dispersion season and total nasal and ocular symptom and medication score for the peak symptom period and JC pollen dispersion season. RESULTS: For the primary end point, absolute reductions and relative mean reductions in TNSMS compared with placebo were 1.50 and 21.4%, 2.24 and 32.1%, and 2.18 and 31.2% for 2,000, 5,000, and 10,000 JAU, respectively (P < .001 in all groups). For all key secondary end points, efficacy was confirmed for all doses (P < .001 in all groups). The treatment was well tolerated. Long-term efficacy of 5,000 JAU was shown over the 3 years. CONCLUSIONS: The optimal dose of the JC pollen SLIT tablet was 5,000 JAU, with good efficacy and safety over a 3-year treatment period. This sustained effect was dependent on treatment duration.

    BACKGROUND: Sublingual immunotherapy (SLIT) can improve the symptoms of allergic rhinitis and modify its natural history; however, its efficacy varies among patients. This study aimed to determine which factors modify the effect of SLIT through post hoc analysis of a previous phase 3 trial of standardized Japanese cedar (JC) pollen extract (CEDARTOLEN®). METHODS: The study included 482 patients who had previously completed a phase 3 trial during two seasons. The SLIT and placebo groups each contained 241 subjects. Because pollen dispersal differed in the two seasons, we identified good and poor responders from the SLIT group in the 2nd season. We compared patient baseline characteristics, changes in serum immunoglobulin, and severity of symptoms in the 1st season between good and poor responders, as well as between SLIT and placebo groups. RESULTS: When we compared the baseline characteristics of good and poor responders, a significant difference was observed in body mass index (BMI) such that the patients with BMI ≥25 presented with lower treatment efficacy. No significant difference was observed in correlation with any other factors or treatment-induced alterations of serum immunoglobulin levels. We found that 75.3% of the patients with moderate symptoms and 50.9% of the patients with severe or very severe symptoms in the 1st season met our criteria for good responders in the 2nd season. CONCLUSIONS: BMI might modify the effect of SLIT; however, other factors were not related clearly. The severity of symptoms in the 1st season of treatment does not predict that in the 2nd season.

    INTRODUCTION: Spinal tuberculosis (TB) is the most common manifestation of extra-pulmonary TB. TB of the lumbosacral junction is rare and occurs in only 1 to 2% of all cases of spinal TB. Moreover, isolated sacrococcygeal TB is extremely rare. Herein, we report a rare case of sacrococcygeal TB, which was difficult to distinguish from complex anal fistula. CASE PRESENTATION: A 93-year-old man presented with sacral pain and peri-anal discharge. He had pulmonary TB at 25 years of age. Fistulography revealed an abnormal tract that connected to the pre-sacrococcygeal area, which was not connected to the rectum. Computed tomography scan showed fluid collection in front of the sacrum, with a lytic destruction of the lower sacrum and coccyx. Cold abscess aspiration cytology was negative for acid-fast bacilli. However, real-time polymerase chain reaction was positive for Mycobacterium tuberculosis. His symptoms resolved immediately after the initiation of anti-TB chemotherapy. CONCLUSION: This case highlights the importance of considering tuberculosis as a diagnosis if the unusual sites are involved.

    BACKGROUND/OBJECTIVE: As few reports are available on treatment goal for Japanese cedar pollinosis treated with allergen immunotherapy, therapeutic effect has to be assessed based on subjective symptoms in clinical practice. This research was conducted to investigate the informative treatment goal in clinical practice by using the results of TO-194SL phase III clinical trial. METHODS: In this investigation, correlation between total nasal symptom medication score (TNSMS), which was predefined as the primary endpoint, and secondary endpoints evaluated by severe symptom day, quality of life (QOL) score, and overall evaluation by subject, was firstly investigated. Based on the results, stratified analyses of the secondary endpoints to confirm relationships on the stratified results, TNSMS, and cedar season were performed. RESULTS: Correlation between TNSMS and evaluations which were severe symptom day, QOL score, and overall evaluation by subject was reasonably observed. TNSMS generally showed 3 or below (classified as mild) throughout the entire pollen season when these evaluations reached each threshold. CONCLUSIONS: The results of this investigation suggest that a few evaluation items would help patients to realize the therapeutic effects of allergen immunotherapy if they could be the treatment goal in clinical practice.

    BACKGROUND: The severity of symptoms of pollen-induced allergic rhinitis is affected by the amount of scattered pollen. However, the relationships between the pollen dispersal pattern, symptom severity, and treatment efficacy are not clear. METHODS: Between 2007 and 2012, we performed 4 randomized, placebo-controlled studies of sublingual immunotherapy (SLIT) on patients with Japanese cedar-induced allergic rhinitis who lived in or around Chiba, Japan. The participants were asked to avoid using rescue medicines during the cedar pollen season as much as possible and to record their nasal symptoms in allergy diaries. The amount of pollen dispersed daily was quantified using the Durham method, and the season was divided into early and late periods based on the pollen count. RESULTS: A total of 721 patients were enrolled in the 4 studies during the 6-year study period. In the placebo group (n = 349), a correlation was observed between the amount of pollen dispersed and the severity of symptoms in the early but not late period of pollen dispersal. Treatment with SLIT (n = 372) significantly improved symptom severity in the late but not early period. CONCLUSION: For patients with Japanese cedar pollen-induced allergic rhinitis, the fluctuation of daily pollen dispersal had a minimal effect on the severity of symptoms during the late period. SLIT was remarkably effective in alleviating symptoms during this period but not in the early period.

    DNA mutations play a crucial role in the origins of cancer, and the clonal expansion of mutant cells is one of the fundamental steps in multistage carcinogenesis. In this study, we correlated tumor incidence in B6C3F1 mice during the period after exposure to N-ethyl-N-nitrosourea (ENU) with the persistence of ENU-induced mutant clones in transgenic gpt delta B6C3F1 mice. The induced gpt mutations afforded no selective advantage in the mouse cells and could be distinguished by a mutational spectrum that is characteristic of ENU treatment. The gpt mutations were passengers of the mutant cell of origin and its daughter cells and thus could be used as neutral markers of clones that arose and persisted in the tissues. Female B6C3F1 mice exposed for 1 month to 200 ppm ENU in the drinking water developed early thymic lymphomas and late liver and lung tumors. To assay gpt mutations, we sampled the thymus, liver, lung, and small intestine of female gpt delta mice at 3 days, 4 weeks, and 8 weeks after the end of ENU exposure. Our results reveal that, in all four tissues, the ENU-induced gpt mutations persisted for weeks after the end of mutagen exposure. Clonal expansion of mutant cells was observed in the thymus and small intestine, with the thymus showing larger clone sizes. These results indicate that the clearance of mutant cells and the potential for clonal expansion during normal tissue growth depends on tissue type and that these factors may affect the sensitivity of different tissues to carcinogenesis.     BACKGROUND: Japanese cedar (JC) pollinosis is the most common seasonal allergic rhinitis in Japan. Standardized JC pollen extract is available for subcutaneous immunotherapy, but this treatment is limited by potentially serious side effects. The aim of this double-blind, randomized comparative study was to evaluate the efficacy and safety of standardized JC pollen extract in a new oral formulation (CEDARTOLEN®, Torii Pharmaceutical Co., Ltd., Tokyo, Japan) for sublingual immunotherapy (SLIT) for JC pollinosis. METHODS: A total of 531 subjects with JC pollinosis were randomized into 2 groups at a ratio of 1:1 to receive daily sublingual administration of standardized JC pollen extract with a maintenance dose of 2,000 Japanese allergy units (JAU) or placebo for 2 consecutive pollen seasons. The efficacy was evaluated using the total nasal symptom and medication score (TNSMS) as the primary end point. Secondary end points included the total ocular symptom and medication score (TOSMS) and scores for individual symptoms and medication. RESULTS: The TNSMS was significantly lower (p < 0.0001) in the SLIT group than in the placebo group in the peak symptom period by 18 and 30% in the first and second seasons, respectively. All secondary end points were also significantly lower in the SLIT group in both seasons. No systemic anaphylaxis occurred. CONCLUSIONS: SLIT with daily administration of standardized JC pollen extract was effective for improving nasal and ocular symptoms of JC pollinosis and reducing the use of relief medication. The JC pollen extract was well tolerated with only local adverse events.

    We developed microfluidic devices for cell fusion of bacteria with low damages and for expressing or strengthening characteristics of fused bacteria. As for the cell fusion device, we developed microscaled gap opposing electrodes to reduce the voltage to induce cell fusion, and a single bacterium-sized narrow gap to transport a single bacterium to reduce the randomness of gene expression. As for expressing and strengthening characteristics, we developed techniques to control and apply different stresses of temperature by serially connected heater and concentration of antibacterial drug by gradient generator. For the cyclic operation of expressing and strengthening fused bacterial characteristics, we also achieved equal division of a droplet by a geometrical design, and sequential microfluidic transfer of equally divided droplets.

    Handling of DNA fragments comprising over several dozens of kb is difficult, because of their fragility when they are in solution. To avoid physical shearing in vitro, assembly of contig DNA fragments have been accomplished within Bacillus subtilis cell using one step protoplast transformation method. Moreover, to avoid physical shearing in the purification step, we demonstrated that large DNA plasmid over 50 kb in size can be introduced into Escherichia coli, or Saccharomyces cerevisiae using the lysate of donor B. subtilis cells. Conjugation protocol has also been established for transfer of large-sized DNA from E. coli to
    eukaryotic microorganisms (S. cerevisiae). Although transfer to animal culture cell remains, our designed horizontal transfer methods is a versatile procedure for delivery of large-sized DNAs into prokaryotic and eukaryotic microorganisms, which would be a basic platform in the synthetic genome area.

    Cell-to-cell direct cloning technology has established by designed horizontal transfer using extra-cellular DNA without purified DNAs. The purified DNAs in a test tube have been generally required to introduce into the host cell for molecular cloning technology in laboratory. In the natural environment, extra-cellular DNAs without purified step contribute to the gene delivery during horizontal gene transfer (HGT) dependent on the stability of DNA released from donor cells. Under such a concept we have developed that DNA delivery procedure using lysate of Escherichia coli lysed by virulent bacteriophage into competent Bacillus subtilis cells. The direct cloning technology has been applied for the DNA synthesis by fragment assembly within the B. subtilis genome. Furthermore we showed the transfer of plasmid DNAs from lysing E. coli into cyanobacteria, which is suggests the procedure is useful for general use.

    シイタケの子実体形成には光が必要である。現在までに光受容体をコードする遺伝子phrA, phrB, Le.cryを単離・同定しているが、光受容体は光を一番最初に受け取る分子であるため、これらの発現は分化ステージによらず、構成的であることが多い。そこで、きのこ類に存在する光応答メカニズムを含めた関連因子(タンパク質)を網羅するため、シイタケにおける子実体形成過程を①栄養増殖菌糸、②子実体原基、③成熟子実体に分け、各々の試料から細胞内タンパク質を抽出し、二次元電気泳動に供した。3種の蛍光染色(全タンパク質染色、リン酸化タンパク質染色、糖鎖付加タンパク質染色)を行い、モレキュラーイメージャーにて解析した。検出されたタンパク質は、各々の分化ステージにおいて約6000種類、リン酸化タンパク質は約300種類、糖鎖付加タンパク質は約30種類存在することが明らかとなった。
    発現に差異のあるスポットをゲルから切り出した後プロテアーゼ処理を行い、目的タンパク質の分解産物(ペプチド)画分を得た。サンプルはLC-MS/MSにてアミノ酸配列解析を行った後、タンパク質質量分析器データベース(マスコット・サーバー)に対し相同性検索を行い、目的産物タンパク質を同定・機能推定を行った。その結果、子実体形成期に特異的に発現するタンパク質は、その発現が上昇するもの(脂肪酸分解酵素、ATP合成タンパク質、G-タンパク質ファミリー、細胞骨格タンパク質、シグナル伝達関連因子等)と、一方減少するもの(カタラーゼ、アルコールデヒドロゲナーゼ、オキシドレダクターゼ等)が同定された。
    これらタンパク質は、現在までのトランスクリプトーム解析によるデータと照らし合わせ、きのこの子実体形成期間に細胞内で起こっている代謝やシグナル伝達経路を如実に反映しているものと示唆された。

    A designed horizontal gene-transfer system by natural transformation has been developed as novel cloning tool to introduce plasmid DNA into the objective cells without purification. Each culture condition for donor and recipient cells has been decided in this study. The bacteriolysis induction of donor cells by virulent phage is very important for universal use. Furthermore it was revealed that each aurintricarboxylic acid as nuclease inhibitor and exonuclease I as accelerator is effective for the DNA transfer by mixing it appropriately. In this condition it was found that the DNA transfer was extended to successful size increase up to 100 kb. The damage-free handling of DNA larger than 100 kb in test tubes is increasingly difficult. The designed transfer system named as Culture Mix Method (CMM) can apply various plasmid DNAs simply and conveniently, which should facilitate development of experimental tools to mobilize giant DNAs for genomic syntheses in synthetic biology.

    Fruiting body formation of mushroom requires light. The Le.cry gene in L. edodes encoded a cryptochrome photo receptor first isolated from basidiomycetes. The expression product, Le.CRY, was suggested to be involved in fruiting body formation via perception of blue light. As a result of proteome analysis, there existed metabolisms and signal transduction pathways involved in fruiting body formation, which were mediated by protein phosphorylation and glycosylation. Photo-responsive genes were comprehensively identified by transcriptome analysis.

    真核生物のゲノムを用いたBGMベクターへの直接的なターゲットクローニングを行うに当たってゲノムサイズが比較的小さいシイタケゲノム(約40Mb)を対象とし、ゲノム中に数百コピー存在するLTR(Long Terminal Repeat)領域、約400bpを組み込むための足場として利用した。マーカーであるcIカセット(cI-spc)を挟むようにして2つのLTRを同方向に組み込んだBGMベクターを構築し形質転換を試みた。この結果幾つかのコロニーは得られたものの残念ながら目的とするシイタケゲノムを組み込んだ株は今のところ得られていない。原因としてLTR領域同士で組み換えが起こり欠してしまった可能性と、用いたゲノム自体に多くのダメージが入っていた可能性が考えられる。この点を改良すべく、ゲノムの精製法を改良する必要があり、また組み換えが起っても欠失しない(逆位となるように)2つのLTRを逆向きに組み込んだBGMベクターを作製する必要がある。これらに関して現在も引き続き調整中であり、また他の足場として交配決定遺伝子座(MATA座、約10kb)の領域についでも随意調整中であるが、残念ながら平成19年度中に良好な結果は得られなかった。しかしながら2年間の本研究を通じて2つのcIカセット(cI-spcとcI-BSを用いた形質転換で198kbと220kbのマウスゲノムを含む2つBACクローンを一度に組み込むことができ、さらにこれらを連結させ、355kbに及ぶJmjゲノム領域を再構築できた事は効果的なクローニング技術開発の一環として-定の成果であると考えられ、現在学術論文として投稿準備中であるとともにBGMベクターの多様なクローニング技術の開発について今後とも進展が期待される。

    Etiology of coronary spasm is still unknown. Therefore, we investigated whether the autonomic activity or platelet activity related to the occurrence of the coronary spasm during perioperative period. For this purpose, we carried out the following.
    1.We constructed the simultaneous recording system and the particle and dense analysis in the regional area for interest on visual images of the transesophageal echocardiography, ECGs, and analog pressure wave forms on the mobile PC.
    2.We examined whether vascular endothelial dilatation was impaired with hypothermia during CPB, which might cause the prolonged hypoperfusion of myocardium after CPB. In conclusion, these results indicated that the blunting of the vascular response to Ach was more prominent than that to TNG in deep hypothermic CPB. This suggests that deep hypothermic CPB caused severe impairment to the endothelial function than to the smooth muscle function.
    3.We investigated whether even a smaller production of heparin and protamin complex(HPC) in patients with off-pump CABG would affect platelet function, by comparing in patients with on-pump CABG. Although a small dose of heparin activated the platelet function, a reduced production of HPC, estimated by smaller doses of hepain and protamin, did inhibit platelet function even in off-pump CABG.
    4.Parasympathetic activity augmented, but sympathetic activity did not change during anesthesia induction with using RR-inteirval analysis. Therefore, there was less possibility to induce coronary spasm during anesthesia induction. During the tracheal intubation, parasympathetic activity increased with accompanying augmentation of sympathetic activity. Although myocardial ischemia did not detect either situation, there was a possibility to induce the coronary spasm during tracheal intubation dependent on the balance of sympathetic and parasympathetic activation in the patients with ischemic heart disease. Further investigation is necessary for the determination of the other triggering of coronary spasm except anesthesia induction and tracheal intubation during anesthesia.

    To investigate the effects of edrophonium (Edr), which could produce the parasympathetic activation, on electrocardiogram (ECG) and hemodynamics, we studied 32 patients receiving elective coronary artery surgery. The patients were randomly divided into two groups ; control (C) group with only saline injection (n=14) and Edr (E) group with Edr followed by the saline injection (n=18). A bolus dose of Edr (0.25 mg・kg^<-1>) was injected through a central venous catheter during harvesting of the left internal thoracic artery . Edr induced mild sinus bradycardia in all patients. The RR interval prolonged after Edr (p<0.01 vs. C group) and the peak prolongation appeared at 2-4 min (p<0.01 vs. baseline). The maximum reduction of heart rate (HR) after Edr was amplified proportionally with increasing baseline HR (r=-0.74, p<0.001). The relationship between the baseline HR and the maximum reduction of HR was apparently different between the two groups (p<0.01). The PQ duration on ECG was elongated in 1 to 3 min after Edr (p<0.01 ) while QRS duration did not. The PQ duration quickly restored to the baseline value at 4 min after Edr, while the RR prolongation persisted for 20 min. There was no newly developed deviation of ST segments on the both leads ll and V_5 for 20 min-observation. Although mixed venous hemoglobin oxygen saturation (SvO_2) decreased for 15 min after Edr (p=0.01 vs. C group), none of the systemic and pulmonary arterial pressures, and central venous pressure changed during this period. A bolus injection of Edr showed a frequency -dependent, temporal depression on HR with little deterioration of hemodynamics except SvO_2. Edr could be expected to induce clinically tol erable sinus bradycardia without any severe A-V conduction block in patients with ischemic heart disease. We found no evidence of newly myocardial ischemia, which could be produced by the parasympathetic activation. Only the parasympathetic activation rarely produces the coronary spasm even in the patients with ischemic heart disease.

    研究の最初の目的である脳分離体外循環時の外科手技に伴う塞栓あるいは脳虚血の発生を早期発見するという点について術中モニタリングを以下の方法で行い実績を挙げた。
    【対象と方法】脳分離体外循環を用いた心臓手術14例において両側局所脳酸素飽和度(rSO_2)ならびに内頸静脈酸素飽和度(SjO_2)の同時計測を行った。フェンタニル、プロポフォールの気管内挿管麻酔下に、右内頸静脈から5.5Fr.のOpticath R(Abbott社)をセルジンガー法で挿入、先端が球部に達するように進めた。両側の前額部にrSO_2プローブ(INVOSR 3100A,Somanetics社)を装着し、手術終了まで連続モニタリングした。rSO_2<50%、SjO_2<40%を異常値と設定し、異常値出現と術後の脳神経機能障害との相関を検討した。
    【結果】rSO_2のみが術中50%以下に低下したのは14例中4例、またSjO_2のみが40%以下に低下したのは14例中2例と、rSO_2、SjO_2のどちらかが単独で異常低値を示したものは14例中6例であった。rSO_2、SjO_2の両者に異常低値が認められたのは14例中2例で、2例中1例に術後一過性の対麻痺がみられた。すなわちrSO_2<50%、あるいはSjO_2<40%の単独で異常低値が出現した症例は、術後脳障害に関してはすべてfalse positiveであった。しかし、rSO_2<50%、かつSjO_2<40%と両方に異常低値を示したのは14例中2例であり、うち1例に一過性の片麻痺を認めたが、他の1例では迅速に対応することで脳機能障害を回避できたと考えられた。
    【結論】脳分離体外循環時の外科手技に伴う塞栓あるいは脳虚血の発生を早期発見するという点について、rSO_2およびSjO_2の同時測定は脳分離体外循環時の外科手技に伴う塞栓あるいは脳虚血の発生を早期発見するという点について有用と思われる。
    現在症例を増やし、この研究を続行している。
