    By heating 1‐(ω‐azidoalkyl)‐2‐(2,2‐dihalovinyl)arenes in DMF, intramolecular Huisgen cycloaddition between the azido group and the 1,1‐dihalovinyl moiety proceeds smoothly to afford 5‐halo‐1,2,3‐triazole‐fused tricyclic benzocondensed compounds. Leveraging the remaining halogen groups, the carbon chain extension via the Mizoroki–Heck or Suzuki–Miyaura coupling reactions, followed by an intramolecular Friedel–Crafts reaction, furnish polycyclic systems that incorporate fused triazole rings. Furthermore, the transformation of the halogen groups into the 2‐nitrobenzene derivatives via Suzuki–Miyaura cross‐coupling, followed by a Cadogan reaction using triphenylphosphine, provides a series of fluorescent pentacyclic compounds.

    Abstract

    The treatment of nitro compounds with Grignard reagents in the presence of a catalytic amount of FeCl2/t‐BuMgCl produced various types of secondary amines in good yields. This reductive coupling most likely involves the compatibility of functional groups such as chlorine, benzyl ether, olefin, and acetylene. Moreover, the corresponding poly‐substituted amines were not observed under these conditions. When the Grignard reagents were replaced with organozinc reagents, functionalized secondary amines containing ester and nitrile groups were obtained in good yields.

    Abstract

    When a mixture containing 1‐halo‐1‐alkyne, a dinucleophilic agent, and a base (Cs2CO3 or K3PO4) was heated in DMF, regio‐ and stereo‐selective nucleophilic addition of the dinucleophilic agent to the acetylenes occurred. This reaction yielded (Z,Z)‐bis(haloalkene), which subsequently underwent Pd‐catalyzed cyclization through C−H bond activation, resulting in the formation of two‐oxygen‐ or ‐nitrogen‐containing hetero‐π‐conjugated compounds. N‐(5‐Hydroxynaphthalen‐1‐yl)methanesulfonamide was treated with 2‐phenyl‐1‐chloroethyne followed by 1‐chloro‐1‐hexadecyne. After these reactions, Pd‐catalyzed double C−H cyclization was employed to produce 1H‐2‐phenyl‐7‐tetradecylbenzofuro[7,6‐g]indole. Notably, this compound transformed into a fan‐shaped structure at 86 °C and transitioned into a smectic A phase characterized by a layered structure.

    Abstract

    Treatment of 1‐azido‐2‐(2,2‐dihalovinyl)arenes with a catalytic amount of Rh2(esp)2 afforded various 2,3‐dihaloindoles in 51–74% yields. This reaction progressed via the intramolecular cyclization of rhodium nitrene generated in situ and the rearrangement of one halogen group. For the 2‐chloro‐2‐iodovinyl group, the iodo group selectively rearranged to yield 2‐chloro‐3‐iodoindoles as single isomers. Moreover, 1‐azido‐2‐[(2,2‐dihalomovinyl)(Boc)amino]arenes were reacted with a catalytic amount of Rh2(oct)4, which produced 2‐haloquinoxaline in 55–92% yields.

    This research representative developed various synthetic methodologies of polifunctionalized compounds by using interaction of iron catalyst and functionalized carbon-carbon multiple bonds. Specifically, the following items were carried out. 1) Application of iron-catalyzed selective addition to functionalized dienes of Grignard reagents. 2) Iron-catalyzed selective addition to sulfonyl dienes or functionalized enynes of Grignard reagents. 3) Application of iron-catalyzed selective substitution to functionalized diene monoepoxides of Grignard reagents. 4) Reductive homocoupling reaction of functionalized diene monoepoxides with iron catalyst-stereoselective synthesis of cyclopropanes. 5) Iron-catalyzed selective synthesis of peroxyacetals from benzyl ethers and tert-butyl hydroperoxide. 6) Iron-catalyzed preparation of amines from nitro compounds and Grignard reagents via reductive coupling reaction.

    Iron reagent generated from FeCl_2 and Grignard reagent is useful for metallative cyclization of functionalized acetylenes and olefins, which subsequently enables the preparation of various kinds of polifunctionalized cyclic products.     本年度はジアミン誘導体とブロモアセチレンとの分子間カップリング反応によるイミダゾリジン誘導体の効率的な合成法の開拓および適用性の拡大を検討し,以下の結果が得られた.1)種々のジアミン誘導体とブロモプロピオール酸アミドをDMF溶媒中,塩基と加熱するだけで,様々なイミダゾリジン誘導体を得ることができた.2)本反応の進行の可否には,ジアミン上の保護基の選別が非常に重要であり,トシルアミドよりもNHプロトンの酸性度が低くても高くても,ダブル付加によるヘテロ環の生成はほとんど起こらないことが分かった.3)カップリングパートナーとして,ブロモプロピオール酸エステルを用いても,ブロモプロピオール酸アミドの時と同様に分子間カップリング反応が進行し,対応するエステル置換イミダゾリジン誘導体を得ることもできた.また,得られたエステル誘導体にDIBALを作用させると,収率良くアリルアルコール誘導体に導くこともできた.4)エチレンジアミンの片方のアミンをトシル基,もう一方をBoc基で保護したジアミン誘導体とブロモプロピオール酸アミドを,塩基と加熱反応させたところ,非対称のイミダゾリジン誘導体を単一の異性体として得ることができた.5)トシル基をアミノ基上に導入したアミノアルコール誘導体と,ブロモプロピオール酸アミドを塩基存在下で反応させたところ,二重結合の置換基の立体化学が制御されたオキサゾリジン誘導体を単一の異性体として得ることができた.
