    Here we studied cerium oxide nanoparticles (nanoceria) as an agent for the future treatment of oxidative damage by validating and evaluating its scavenging activity towards reactive oxygen species (ROS) in vitro. Nanoceria has been shown to mimic the activities of superoxide dismutase and catalase, degrading superoxide (O2•-) and hydrogen peroxide (H2O2). We examined the antioxidative activity of nanoceria, focusing on its ability to quench singlet oxygen (1O2) in an aqueous solution. Electron paramagnetic resonance (EPR) was used to determine the rates of second-order reactions between nanoceria and three ROS (1O2, O2•-, and H2O2) in aqueous solution, and its antioxidative abilities were demonstrated. Nanoceria shows a wide range of ultraviolet-light absorption bands and thus 1O2 was produced directly in a nanoceria suspension using high-frequency ultrasound. The quenching or scavenging abilities of nanoceria for 1O2 and hypoxanthine-xanthine oxidase reaction-derived O2•- were examined by EPR spin-trapping methods, and the consumption of H2O2 was estimated by the EPR oximetry method. Our results indicated that nanoceria interact not only with two previously reported ROS but also with 1O2. Nanoceria were shown to degrade O2•- and H2O2, and their ability to quench 1O2 may be one mechanism by which they protect against oxidative damage such as inflammation.

    Photoluminescent gold clusters are functionally variable chemical modules by ligand design. Chemical modification of protective ligands and introduction of different metals into the gold clusters lead to discover unique chemical and physical properties based on their significantly perturbed electronic structures. Here we report the synthesis of carbon-centered Au(I)-Ag(I) clusters with high phosphorescence quantum yields using N-heterocyclic carbene ligands. Specifically, a heterometallic cluster [(C)(AuI-L)6AgI2]4+, where L denotes benzimidazolylidene-based carbene ligands featuring N-pyridyl substituents, shows a significantly high phosphorescence quantum yield (Φ = 0.88). Theoretical calculations suggest that the carbene ligands accelerate the radiative decay by affecting the spin-orbit coupling, and the benzimidazolylidene ligands further suppress the non-radiative pathway. Furthermore, these clusters with carbene ligands are taken up into cells, emit phosphorescence and translocate to a particular organelle. Such well-defined, highly phosphorescent C-centered Au(I)-Ag(I) clusters will enable ligand-specific, organelle-selective phosphorescence imaging and dynamic analysis of molecular distribution and translocation pathways in cells.

    <jats:p>Elucidation of the static states and dynamic behavior of oxygen and nitrogen dissolved in water is one of the most important issues in the life sciences. In the present study, experimental trials and theoretical calculations were performed based on the hypothesis that the dissolution of gas molecules in water is related to excitation by the Earth’s magnetic field. Using quantum theories such as those used to describe electro magnetic resonance and nuclear magnetic resonance, this study investigated the states of oxygen, nitrogen and hydrogen dissolved in water. The results indicate that the Earth’s magnetic field is involved in the bonding and dissociation of molecules at the gas-liquid interface. These calculations assessed the effect of a field strength of 1.0 x 10<jats:sup>−4</jats:sup> T and reproduced the influences of temperature changes on dissolved gas concentrations. Molecular interactions caused by electromagnetic properties and the external geomagnetic field were found to affect intermolar bonding associated with water cluster structures. It is concluded that the binding between molecules typically attributed to Coulomb coupling by magnetic charge and van der Waals forces results from excitation in the Earth’s magnetic field.</jats:p>

    The present study identified the active radical species in acidic sodium chlorite and investigated the feasibility of quantifying these species with the diethylphenylenediamine (DPD) method. Electron spin resonance (ESR) spectroscopy was used to identify the active species generated in solutions containing sodium chlorite (NaClO2). The ESR signal was directly observed in an acidified sodium chlorite (ASC) aqueous solution at room temperature. This ESR signal was very long-lived, indicating that the radical was thermodynamically stable. The ESR parameters of this signal did not coincide with previously reported values of the chlorine radical (Cl●) or chlorine dioxide radical (O = Cl●-O and O = Cl-O●). We refer to this signal as being from the chloroperoxyl radical (Cl-O-O●). Quantum chemical calculations revealed that the optimal structure of the chloroperoxyl radical is much more thermodynamically stable than that of the chlorine dioxide radical. The UV-visible spectrum of the chloroperoxyl radical showed maximum absorbance at 354 nm. This absorbance had a linear relationship with the chloroperoxyl radical ESR signal intensity. Quantifying the free chlorine concentration by the DPD method also revealed a linear relationship with the maximum absorbance at 354 nm, which in turn showed a linear relationship with the chloroperoxyl radical ESR signal intensity. These linear relationships suggest that the DPD method can quantify chloroperoxyl radicals, which this study considers to be the active species in ASC aqueous solution.

    Wine lees, a major waste product of winemaking, is a rich source of polyphenolic compounds. LED-light irradiation at 400-nm elicited microbicidal activity of aqueous extract from wine lees (WLE) against Staphylococcus aureus, Pseudomonas aeruginosa, and Candida albicans, in addition to reactive oxygen species (ROS) formation, including hydroxyl radical (·OH) and hydrogen peroxide (H2O2). Although treatment for 20 min of photoirradiation alone exerted bactericidal activity with a 2- to 3-log reduction, photoirradiated WLE for 20 min achieved a 5-log or greater reduction in viable S. aureus and P. aeruginosa cells. Regarding C. albicans, a 1-log reduction (90 % reduction) of viable cells was achieved by photoirradiated WLE for 40 min, whereas photoirradiation alone did not show any fungicidal effect. ROS analyses revealed that approximately 170 μM ·OH and 600 μM H2O2 were generated in photoirradiated WLE for 20 min. Because the bactericidal activity of photoirradiated WLE was abolished by ·OH scavengers, ROS, especially highly oxidative ·OH, may be responsible for the microbicidal activity of photoirradiated WLE. In addition to its microbicidal activity, WLE may act as an antioxidant as it exerted radical scavenging activity against 2,2-diphenyl-1-picrylhydrazyl, a stable free radical.

    The reactions of three α-oxoaldehydes (methylglyoxal, glyoxal, and pyruvic acid) with hydroxyl radicals generated by sonolysis of water were investigated using an electron spin resonance (electron paramagnetic resonance) spin-trapping method, and their reaction kinetics were investigated. It is apparent from our experimental results that methylglyoxal exhibits the highest reactivity of the three α-oxoaldehydes. These α-oxoaldehydes can react with hydroxyl radicals faster than other well-known antioxidants can. The reactivity of hydroxyl radicals is higher than that of hydrogen peroxides.

    BACKGROUND: The cellular temperatures of microorganisms are considered to be the same as those of their surroundings because the cellular volume is too small to maintain a cellular temperature that is different from the ambient temperature. However, by forming a colony or a biofilm, microorganisms may be able to maintain a cellular temperature that is different from the ambient temperature. In this study, we measured the temperatures of bacterial colonies isolated from soils using an infrared imager and investigated the thermogenesis by a bacterium that increases its colony temperature. RESULTS: The temperatures of some colonies were higher or lower than that of the surrounding medium. A bacterial isolate with the highest colony temperature was identified as Pseudomonas putida. This bacterial isolate had an increased colony temperature when it grew at a temperature suboptimal for its growth. Measurements of heat production using a microcalorimeter showed that the temperature of this extraordinary, microcalorimetrically determined thermogenesis corresponded with the thermographically observed increase in bacterial colony temperature. When investigating the effects of the energy source on this thermal behavior, we found that heat production by this bacterium increased without additional biomass production at a temperature suboptimal for its growth. CONCLUSIONS: We found that heat production by bacteria affected the bacterial colony temperature and that a bacterium identified as Pseudomonas putida could maintain a cellular temperature different from the ambient temperature, particularly at a sub-optimal growth temperature. The bacterial isolate P. putida KT1401 increased its colony temperature by an energy-spilling reaction when the incubation temperature limited its growth.

    Enzymes, which catalyze a variety of reaction in living systems, have many specific features, such as high reactivity and safety for the environmental. However, enzymes are relatively expensive, and therefore discarding them after a single use is not economical. A variety of methods for the immobilization of enzymes have been eveloped, but strong interaction between enzyme and carrier causes denaturalization and deactivation of enzymes. In this work, we report a method of hydrogenase immobilization using polymer containing cationic side chains. Hydrogenases catalyze evolution hydrogen by proton reduction. This method prevented denaturalization of enzymes by using electrostatic interaction.

    The existence of an energy field that dominates the physiological mechanism of incorporating O2 in the atmosphere into the human body and excreting it outside the body as CO2 was investigated using computational chemistry techniques based on quantum theory. As a result, it was revealed that the binding, transportation, and metabolism of gas molecules depend on the weak magnetic energy induced by the earth's magnetic field (H0 = 4.5 × 10−5 T). Finally, it was concluded that the mechanism of oxygen transport and Christian Bohr's proposed”Bohr effect” depend on magnetic coupling between biomolecules.

    Cytochrome c3 has four hemes per molecule, and donates electrons to hydrogenase catalysing hydrogen evolution. In this study, Zn porphyrin intercalated cytochrome c3 was prepared in order to utilize high affinity of cytochrome c3 to hydrogenase in photoinduced hydrogen evolution systems. Photodriven reduction of Zn porphyrin intercalated cytochrome c3 occurs by visible light irradiation. An electron transfers from photoexcited Zn porphyrin to intrinsic Fe porphyrin. Procedures of Zn porphyrin was prepared by the intercalation of Zn porphyrin to apo type cytochrome c3. The apo type cytochrome c3 was obtained by removing one of four hemes in cytochrome c3. Photoinduced hydrogen evolution was observed by the irradiation, and photoinduced electron transfer occurs from Zn porphyrin to hemes and then to hydrogenase.

    Methane monooxygenase (MMO) catalyzes the hydroxylation of methane to methanol. MMO is a multicopper enzyme, and contains Type II and Type III copper ions. The role of the copper ions for catalysis was studied.
    When EDTA was added to MMO, the copper ion content decreased and MMO became inactive.
    EPR spectrum of MMO is derived from Type II copper (II) ions, and the signal intensity decreases when duroquinol is added as a reductant, suggesting that a part of those copper ions included in MMO was eliminated by EDTA, and that this copper ion is related to an electron transfer from a reductant.

    Cellular uptake and excretion of Zn-tetrasulfophthalocyanine (ZnTSPc) was determined by spectrophotometric analysis of the supernatant. After incubation time of 15, 30, 60, 1440 min. with ZnTSPc, cellular uptake was found to be 1.9 pico g per single Hela cell that corresponds to about 109 molecules of ZnTSPc localized in or on one cell. This indicates that cellular uptake rate of ZnTSPc is fast. Cellular excretion of ZnTSPc was observed after removal of this photosensitizer from the medium. Though 16 percent of ZnTSPc that was taken in or on cells was excreted after 72 hr., no significant differences in efficiency of Photodynamic effects on cell inactivation was observed.

    It is essential to understand the dynamic behavior of the excite triplet state of sensitizer in PDT (Photodynamic Therapy). Usually the Laser Flash Photolysis is used for measuring the triplet state in vitro, but the method for in vivo has not been reported. In this study we developed the system for in vivo measurement of the triplet state. The animal experiments revealed that the life time of HpD and NHpD in vivo is almost coincide with the life time of air saturated HpD and NHpD in vitro.

    ZnTSPc was administered to C3H/He mice via a tail vein (15mg/kg). At different times after injection, they were sacrificed. The tumor (transplanted MH-134 cells) as well as some organs was prepared for fluorescent assay. Accumulation of ZnTSPc in the tumor was higher than other organs except for the liver at 8 hours after administration. ZnTSPc was observed in the liver even after 50 days. The brain had no accumulation of ZnTSPc at any time.

    ZnPc [(OCH2CH2)3 OH]4 (ZnTEPc) was compared with zinc-tetra-sulfo-phthalocyanine (ZnTSPc) in their photochemical properties and ability to sensitize cells to red light in vitro. Both Pc were added to F-10 medium and incubated with HeLa cells for 30min. After wash-out of the dye, they were exposed to red light. Cells were damaged only by ZnTSPc. ZnTSPc has a longer triplet life time. It generates much singlet oxygen and is incorporated more to cells.

    メタン変換バイオ触媒はメタン-メタノール変換プロセスを大幅に省エネルギー化し、CO2 排出量を抑制できる。本研究ではメタン変換バイオ触媒の性能に有機無機ハイブリッドコーティングが及ぼす影響を明らかにし、このコーティング法を応用したメタン変換バイオ触媒が社会実装可能な性能を示すかどうかを検証する。本手法の有効性を実証することで、常温常圧,一段階での省エネ型メタン変換プロセスの実現に向けた研究展開をめざす。
    2023年度は、高分子電解質としてポリ(ジアリルジメチルアンモニウムクロリド)(PDADMAC)、金属酸化物として酸化ケイ素(SiO2)を用いたメタン変換バイオ触媒表面へのコーティング手法を検討した。メタン酸化バイオ触媒であるバクテリア細胞を培養し、このバクテリア細胞へ種々の条件(PDADMAC や TMOS 濃度、処理時間、温度)でPDADMACとSiO2を順に吸着させることで、コーティング状態の異なるバイオ触媒を調製した。得られた種々メタン変換バイオ触媒について、メタンからメタノールへの転化反応に対する触媒活性と寿命を評価した。その結果、SiO2はメタン酸化バクテリアのメタノール生産性能に影響を及ぼさないのに対して、PDADMCはその性能を低下させる効果を示した。そこでさらに、PDADMAC濃度や吸着溶液のpH、ならびに処理時間や分離方法を検討した。その結果、メタノール生産性能をほぼ維持した状態でメタン酸化バクテリア細胞表面をPDADMACおよびSiO2でコーティングできる条件を見出すことに成功した。

    Single cell analysis of lipid metabolism was investigate with phosphorescence lifetime confocal microscopy. 3T3-L1 cell was differentiated to adipocyte so that the oxygen consumption rate of adipocyte was higher than that of preadipocyte due to increase in the number of mitochondria in the cell. Lipid droplet degradation measurment was carried out with oxygen concentration measurement imaging to clarify the importance of oxygen,
    Effect of low oxygen stress on the oxygen consumption within cell was evaluated. Antioxidant enzyme systems were induced with the lowering oxygen concentration. Oxygen consumption rate was reduced by the acurate hipoxic stress.

    Site directed mutagenesis of cytochrome c3 from Desulfovibrio vulgaris was carried out for the addition of enzymatic activity to the electron transfer protein. Cytochrome c3 was tetraheme electron transfer protein and our previous study reveals the role of each heme. one of the heme plays an important role in electron donation to physiological electron acceptor, hydrogenase. So in this study, heme coordination sphere of the heme was changed by the site direct mutagenesis. Obtained cytochrome c3 can carried out both enzymatic activity and electron transfer activity.

    Cytochrome c3 from Desulfovibrio vulgaris has four hemes per molecule, and a redox change at the hemes alters the conformation of the protein, leading to a redox-dependent change in the interaction of cytochrome c3 with redox partners (an electron acceptor or an electron donor) .The redox-dependent change in this interaction was directly monitored by the high-performance electrochemical quartz crystal microbalance (EQCM) technique that has been improved to give high sensitivity in solution. In this method, cytochrome c3 molecules in solution associate electrostatically with a viologen-immobilized quartz crystal electrode as a monolayer, and redox of the associating cytochrome c3 is controlled by the immobilized viologen. This technique makes it possible to measure the access of cytochrome c3 to the electrode or repulsion from the electrode, and hence interconversion between an electrostatic complex and an electron transfer complex on the cytochrome c3 and the viologen as a mass change accompanying a potential sweep is monitored. In addition, simultaneous measurement of a niece change and a potential step reveals that the cytochrome c3 stores electrons when the four hemes are reduced (an electron pool effect), that is, the oxidized cytochrome c3 facilitates acceptance of electrons from the immobilized viologen molecule, but the reduced cytochrome c3 donates the accepted electrons to the viologen with difficulty.

    近年、遺伝子やタンパク質が生体内でどのような働きをしているかということに注目が集まっている。その中で、これら遺伝子やタンパク質の細胞内での働きを知るために、タンパク質や代謝産物などの網羅的解析技術の開発が望まれている。本研究では、細胞内全mRNAの網羅的な解析が可能な分離法の開発を行う。さらに、分離したmRNAの高感度塩基配列決定法についても検討した。
    本年度は2次元電気泳動によるmRNAの分離方法を確立するために、以下のような研究を行った。本研究では1次元目に通常の電気泳動を行い、分子量に基づく分離をおこない、2次元目として温度勾配電気泳動法2次元電気泳動をおこなった。mRNAから逆転写酵素により相補鎖DNAを合成する際に特殊なプライマーを用いることにより、GCクランプの付加したRNA-DNA2本鎖の合成を行った。まず、細胞から抽出したmRNAを鋳型に逆転写酵素を用いて相補鎖のDNAを合成した。このDNA合成に用いるプライマーとしては、一般的にmRNAのポリA鎖に対する相補鎖であるポリT鎖を用いた。本研究ではさらにGCクランプを分子内にもち、さらに分子内でヘアセン構造を形成するプライマーを用いた。つぎに、このプライマーを用いて合成したmRNAの相補鎖とmRNAをDNAとRNAに作用するリガーゼを用いて結合する(ライゲーション)。次に作成したRNA-DNA複合体を用いて温度勾配電気泳動を行い、最適分離条件についての検討を行った。これらの検討を行うことにより、目的とするmRNAの分離方法が確立できた。

    光線力学治療は光増感剤をガン患者に投与し、光増感剤が取り込まれた後、腫瘍組織に光照射を行い、腫瘍を壊死させる治療法である。現在、光線力学治療用の光増感割として用いられているフォトフリンは腫瘍選択牲が低いこと、組織透過性のよい長波長領域の光の吸光係数が小さく、深部の腫瘍の治療には適さないという欠点がある。そこで、本研究ではこれらの欠点を克服し、さらに抗体の高い腫瘍選択性と光増感剤の殺細胞効果を併せ持つ、抗体-光増感剤複合体の開発を目的としている。本研究では、抗体-光増感剤複合体の調製に疎水性相互作用を利用し、各種光線力学治療用光増感剤と抗腫瘍抗体とを疎水性相互作用により複合体とし、腫瘍に対する標的指向性を高めたミサイル光増感剤の創製を行った。これはモノクローナル抗体によって腫瘍部位に光増感剤を集め、可視光照射により腫瘍の治療を行うものである。
    これまでに、亜鉛カルボキシフタロシアニンおよびクロリンe6を調製し、抗体との結合に関して研究を行った。また、亜鉛シフタロシアニンの分子中に存在するカルボキシル基あるいはスルホ基の数を変えたフタロシアニンを調製し、疎水性の違いによる抗体との複合体形成効率を検討した。調製したフタロシアニンおよびクロリンe6を抗体と疎水性相互作用により複合体形成させ、光増感剤の疎水性を変化させることによる光増感剤の抗体に対する結合数を変化させ、疎水性相互作用による複合体形成能の検討した。これらの化合物の良好な光化学的性質を有していることが分かり、生体内において、良好な光毒性を示した。また、抗体の結合していない光増感剤に比べ、細胞内に良好に蓄積し、さらにガンに対する選択性が向上したことを明らかにした。

    本研究はヒドロゲナーゼとチトクロームc_3との間の電子移動速度に対するチトクロームc_3の電荷の影響及び、チトクロームc_3とヒドロゲナーゼの電子移動部位に存在するアミノ酸の同定を目的としている。
    まずチトクロームc_3とヒドロゲナーゼの間の電子移動速度をストップトフローを用いて測定したところ、天然の基質であるチトクロームc_3はヒドロゲナーゼとの間で効率の良い電子伝達が可能であることが分かった。
    チトクロームc_3とヒドロゲナーゼが効率よく電子伝達することが分かったので、ヒドロゲナーゼとチトクロームc_3との電子伝達複合体に関与する電荷の効果について検討した。チトクロームc_3中の主要な塩基性アミノ酸であるリジン残基を2,4,6-トリニトロベンゼンスルホン酸ナトリウムを用いた。このTNBS修飾チトクロームc_3とヒドロゲナーゼとを組み合わせた水素発生反応の結果、TNBS修飾チトクロームc_3を用いた場合、未修飾のチトクロームc_3に比べ活性が低下することが分かった。これは修飾されたリジン残基がヒドロゲナーゼとの電子伝達に重要な役割を果たしているためであると考えられる。
    さらにヒドロゲナーゼと組み合わせた水素吸収反応でも修飾されたリジン残基が電子伝達に重要な役割を果たしていることがわかった。
