    Cytochrome c_3 is a redox protein containing four hemes in one molecule and works as an electron mediator in vivo. There is intense interest in experiments which reveal, at molecular level, how electrons go into and out of cytochrome c_3. Here we report that specific heme which roles in an entrance or an exit during electron transfer, that is direction of electron transfer through cytochrome c_3, is clarified by electrochemical quartz crystal microbalance (EQCM) measurement. In vivo, cytochrome c_3 accepts/donates electrons from/to hydrogenase, which is catalyze proton reduction and hydrogen oxidation. In this study, electron transfer between cytochrome c_3 and hydrogenase was measured electrochemically, and the electron transfer complex formation was investigated by frequency change of the quartz crystal at the same time as electrochemistry.

    The aim of this study is the screening of novel laccases from metagenomic libraries of environmental samples and the directed evolution of the enzymes for synthesis of functional materials. The present results provided the detailed information about the recombinant expression of the laccases having a high activity. On the basis of this finding, we conducted the directed evolution of the laccases for the function improvement.

    Photoinduced hydrogen evolution with an enzyme and cytochrome c3 (an redox protein) was developed by addition photo-reactivity to cytochrome c3. Cytochrome c3 has four hemes in one molecule and one of hemes was exchange into Zn-porphryn, so that photoinduced electron transfer from Zn porphyrin to three hemes in cytochrome c3 occurred. The Zn porphrin cytochrome c3 applied to photoinduced hydrogen evolution and hydrogen production occurred.
