    理化学研究所(理研)ライフサイエンス技術基盤研究センター微量シグナル制御技術開発特別ユニットの小嶋聡一特別ユニットリーダー、ロナク・シュレスタ国際プログラム・アソシエイト(研究当時)と、加藤分子物性研究室の大島勇吾専任研究員、東京工業大学生命理工学院の梶原将教授らの共同研究グループは、肝臓に侵入した真菌(カビ)が活性酸素、特にヒドロキシルラジカルを作り、その酸化ストレスを介して肝細胞死を引き起こす分子メカニズムを明らかにしました。

    Based on the previous findings, we studied molecular mechanisms by which 1-1) a protein crosslinking enzyme, transglutaminase 2 (TG2) promotes cancer angiogenesis and 1-2) co-culture with fungi induces nuclear TG2 activity in hepatic cells, and 2) screened a food-derived compounds that control the above clarified mechanisms. We found 1-1) TG2 inversely regulates the expression of an endogenous angiogenesis inhibitor, vasohibin 1, in lung endothelial cell cultures, animal models, and patients with lung carcinoma, 1-2) C. albicans and C. glabrata but not S. cerevisiae induces increased nuclear TG2 activity in hepatic cells via producing reactive oxygen species, and 2) phenosafranine inhibits nuclear transportation but not activity of TG2. These results suggest phenosafranine will be promising to suppress nuclear TG2-mediated pathogenesis such as cancer angiogenesis and hepatic injury, but not affect cytosolic TG2-mediated physiological role of TG2 such as promotion of tissue remodeling.

    To clarify what kind of stress environments the fatty acid desaturases genes (Sk-OLE1,Sk-FAD2,Sk-FAD3) of yeast Saccharomyces kluyveri is induced by and where the cis-elements for gene induction on the promoter area exist in, the transcription of these desaturase genes was researched. The results are shown below.
    1)It was shown that these three fatty acid desaturase genes do not response by the addition of unsaturated fatty acid
    2)All the transcription of these three genes were induced by low oxygen environment and their induction patterns were different each other.
    3)Ii was suggested that Sk-FAD2 and SK-FAD3 are regulated by only low temperature stress. On the other hand, Sk-OLE1 are regulated by not only low temperature stress but also cell membrane situation
    4)It was suggested that the necessary area for the low-temperature induction exists between the upstream 228-172bp of the Sk-OLEI structural gene. Moreover, in this area, there is a CCCCT sequence that was called STRE (stress response element) and then the possibility that STRE took part in the low-temperature induction of Sk-OLE1 gene was thought.
    5)It was suggested that the necessary area for the low-temperature induction exists between upstream 326-209bp of the Sk-FAD2 structural gene. In the upstream 228-172bp of this gene, it was shown that there is the T-rich region.
    6)It was suggested that the necessary area for the low-temperature induction exists between upstream 207-145bp of the Sk-FAD3 structural gene. In this area, there is no common region with the low-temperature response areas in the upstream of the Sk-OLE1 gene and the Sk-FAD2 gene although there is a CCAAT-box in this area.
    The portion of the stress response mechanism of yeast S kluyveri became clear by the above-mentioned result of this research.

    Fruiting-body formation of basidiomycetes is one of the most typical and attractive morphological differentiation observed in microorganisms. To understand the molecular mechanisms of fruiting-body formation in Lentinula edodes (Shiitake), we have studied on the target genes of two DNA-binding transcription factors PRIB and Le.CDC5, both of which regulate the fruiting-body formation. The PRIB protein binds to a 16-bp sequence with the consensus sequence of 5'GGGGGGGACAGGANCC3' and the Le.CDC5 protein binds to a 7-bp sequence with theconsensus sequence 5'GCAATGT3'. MfbA is also interesting protein that regulates the fruiting-body formation and possesses RGD sequence, PHD finger and SET domain. So we studied properties of MfbA protein. Concerning the PRIB, Le.CDC5 and MfbA, we obtained the following results. (1)The three target genes were identified for PRIB protein, ie., priB, uck1 (UMP-CMP kinase genes) and mfbC, the homologue of Saccharomyces cerevisiae YJRO070c. The mfbC gene is considered to encode the protein interacting to a putative translation initiation factor eIF5A. Only mature fruiting body was shown to contain the transcript of the mfbC gene almost exclusively, suggesting that mfbC may play a role at the final stage of fruiting-body formation. The mfbC transcript showed a region-specific distribution in hymenophore (gill tissue). High level of the mfbC transcript was detected in the outer region of trama, which is the region branching out into subhymenium. (2)We isolated two (3.7-kb and 3.9-kb) genomic HindIII fragments, to which Le.CDC5 protein binds. The two fragments contained several 7-bp consensus-like sequences and Le.CDC5 protein bound to them. They contained two or three genes of which functions were unknown. (3)The PHD finger-containing fragment of MFBA protein was found to bind to a 7-bp sequence with the consensus sequence of 5'ANAAGGT3'. Computer searching gave the information that the SET domain may be related to the methylase activity for the lysine residues of histone H3. These imply the possibility that MFBA protein may be a sort of chromatin-remodelling protein.

  • 酵母の脂肪酸不飽和化酵素の構造特性とその生成物質の作用機構の解明

    昨年度の研究では、S.cerevisiaeのOLE1蛋白質で認められるC16:0に対する基質優先性が105番目から257番目のアミノ酸残基の間の領域に依存していることが分かった。そこで、本年度はこの領域を再度細分化したキメラ酵素遺伝子を作製して、基質優先性に関与するアミノ酸残基を特定することを試みた。その結果、105-257領域を細分化したキメラ酵素遺伝子のうち、C末端側の17アミノ酸残基をS.kluyveriのOKE1蛋白質のものに替えたものでは同様の基質優先性が認められたが、それ以上のアミノ酸残基の変更はS.cerevisiaeのOLE1蛋白質にみられる基質優先性を失うことが分かった。このことから、S.cerevisiaeのOLE1蛋白質の基質優先性には、ヒスチジンクラスターの2つの保存領域を含む細胞室側の遊離した領域以外に、第一および第二膜貫通領域も必要であることが分かった。一方で、Sc-OLE1遺伝子高発現株つまり膜脂質中の16:1や18:1の割合が増加した株では、エタノール発酵において細胞の増殖速度とエタノール生成速度が野生株と比べて高くなることが分かっている。この原因を明らかにするために、それぞれの酵母の全可溶性蛋白質を二次元電気泳動法により分画して各蛋白質の量的変化を解析することで、16:1や18:1の割合の増加により高発現されている蛋白質を同定することを試みた。現在、さらに詳細な解析を行っているが、今までのところ、Sc-OLE1遺伝子高発現株では解糖系に関わる酵素群の幾つかが増加していることが分かってきており、これらのことが酵母の増殖とエタノール生成を向上させている原因のひとつであることが示唆された。今後はSc-OLE1遺伝子高発現により、どうして解糖系の酵素量が増加するのかを分子レベルで調べる予定である。

    本研究は酵母Saccharomyces kluyveriの不飽和脂肪酸生合成経路及びその制御機構の解明を目的とし、多不飽和脂肪酸要求性株を相補する遺伝子のクローニング、Δ9デサチュラーゼ遺伝子(Sk-OLE1)及び脂肪酸生合成遺伝子(Sk-FAS1)のコードする酵素の基質特異性等の解析等を試みた。S.Kluyveriは、リノレン酸(18:3)を培地中に添加することで、その多不飽和脂肪酸生合成系が抑制されることが分かったことから、Differential Subtraction法を用いて、18:3を添加していないS.Kluyveriで多量に存在するmRNAを抽出し、cDNAライブラリーを作製した。このcDNAライブラリーから既知のΔ12デサチュラーゼで保存されている領域を基にしたプライマーに用いたPCRにより、3種類の増幅したDNA断片を分離した。これらを全て塩基配列決定に供したが、S.KluyveriのΔ12デサチュラーゼ遺伝子と推定されるDNA断片は得られていない。次に、クローニングした2つの遺伝子Sk-OLE1とSk-FAS1の基質特異性を解析するために、酵母S.cerevisiaeのSc-ole1-株及びSc-fas1-株を作製して、これらの株にSk-OLE1、Sk-FAS1遺伝子を各々導入し、得られた形質転換株の脂肪酸を分析した。その結果、S.KluyveriのΔ9デサチュラーゼはS.cerevisiaeのものに比べ、パルミチン酸(16:0)よりむしろステアリン酸(18:0)を基質とする傾向があることが分かった。一方、S.Kluyveriの脂肪酸合成酵素のβサブユニットはS.cerevisiaeのαサブユニットと複合体を形成し、機能することができることが分り、またS.cerevisiaeのものに比べ、パルミチン酸(16:0)をステアリン酸(18:0)よりも多く生合成する傾向があることが分かった。S.Kluyveriの生合成する脂肪酸には鎖長16と18が同程度含まれているので、予想外の結果となった。最後に、Sk-OLE1遺伝子の転写発現解析とそのプロモーター領域の構造解析を行った。S.Kluyveriを低温培養や不飽和脂肪酸を添加した培地での培養を行い、Sk-OLE1遺伝子の転写量を比較解析したところ、低温では転写量の減少が見られ、不飽和脂肪酸添加は転写量に影響しないことが分かった。
