    New peptide probes that interact with polymeric nanoparticles, a model for nanoplastics, and with water-soluble polymers to emit characteristic and distinguishable fluorescent signals were designed and synthesized. A machine learning method was found to classify the fluorescent signals (training data) obtained from the peptide probes. Using the results as an indicator, it was able to identify the test polymeric nanoparticles and water-soluble polymers, which are virtual unknown samples. Thus, by combining molecular probing and machine learning methods, we succeeded in creating a technological basis for the reliable detection and identification of nanoplastics and water-soluble polymers in water in a short time and in small quantities.

    Molecular self-assembly that proceeds spontaneously in water is useful as a low-energy and environmentally friendly material creation method. In this study, we focused on crystalline aggregates consisting of cellulose oligomers (cello-oligosaccharides) or their derivatives with a functional group at the reducing end, which can be easily synthesized by in vitro enzymatic reaction, and composited them with various versatile polymer materials such as filter paper and nonwoven fabrics based on the self-assembly process. We then evaluated and controlled the structure, physical properties, and functions of the resulting composite materials, and developed the potential application value as functional composite materials.

    The neutralization-induced self-assembly of cellulose oligomers produced physically cross-linked hydrogels composed of crystalline nanoribbon networks. The self-assembly proceeded at various temperatures and ionic concentrations. Furthermore, the self-assembly was remarkably extended in serum-containing cell culture media, followed by 3D cell culture through control of the gelation time using seed crystals. Model cells proliferated to produce cell aggregates (so-called spheroids) in the hydrogels due to the anti-biofouling properties of the nanoribbons. The spheroids were readily collected from the hydrogels by natural filtration after mechanical collapsing of the hydrogels, thereby proposing the potential applicability for 3D cell culture matrices. The self-assembly was applicable to cellulose oligomer derivatives with a functional group at the reducing end. Furthermore, the composite hydrogels were produced through the self-assembly in the presence of other functional polymers.

    The self-assembly of biomolecules into highly regular structures is essential in the construction of biological tissues and organs. Various biomolecular assemblies composed of nucleic acids, peptides, and lipids have been utilized as molecular building units for self-assembled materials. However, crystalline oligosaccharides have rarely been utilized in self-assembled materials. In this research, multidimensional self-assembled structures of alkylated cellulose oligomers were synthesized via in vitro enzymatic reactions. It was found that the alkyl chain length drastically affected the assembled morphologies and allomorphs of cellulose moieties. The modulation of the intermolecular interactions of cellulose oligomer derivatives by alkyl substituents was highly effective at controlling their assembly into multidimensional structures. This research proposed a new potential of crystalline oligosaccharides for structural components of unique molecular assemblies.

    Nature-based nanocelluloses are attractive one-dimensional nanomaterials due to their mass productivity and unique physicochemical properties. Our previous studies have reported that activated ester, monophosphate, and amide linkages of small organic substrates are hydrolyzed on the surface of the nanocelluloses. However, fundamental knowledge on the hydrolytic activities of nanocelluloses is limited. In this study, artificial sheet-like nanocelluloses composed of cellulose oligomers were synthesized by phosphorylase-catalyzed enzymatic reactions and their hydrolytic activities against small molecular substrates were characterized. As-prepared nanocelluloses showed relatively low hydrolytic activities. On the other hand, smaller nanocelluloses with greater surface areas, which were prepared by sonication-based mechanical treatment of as-prepared nanocelluloses, significantly showed great hydrolytic activities.

    Polymerization of L-alanine ethyl ester hydrochloride in the presence of cellulose nanocrystal (CNC) prepared by treatment of cellulose sources with hydrochloric acid was investigated. Alanine oligomer products, such as alanine dimer and trimer, were successfully detected by a liquid chromatography-mass spectrometer, suggesting the potential of CNC as a polymerization catalyst. The product amounts were maximal at ambient temperature under weakly acidic or neutral conditions. When several amino acid monomers were applied, CNC accelerated specifically the polymerization of hydrophobic amino acid monomers. Unfortunately, the conversions of monomers to oligomers were less than several %, which should be improved in the near future.
