    東京工業大学 工学院の山本貴富喜(たかとき)准教授らは、病原性ウイルスのセンサーにつながるデバイス構造を開発している。ナノ(10億分の1)メートル(nm)幅の流路を設け、流路を通るウイルスを電気的に検出する仕組み。この仕組みにより、ウイルスの大きさ・形状によって異なるインピーダンス(電気抵抗)スペクトルが得られることを確かめた。この結果はウイルスの構造や成分などからウイルスを識別できる可能性を示唆しており、既知・未知を問わない網羅的なウイルスセンシング法が実現できると期待される。

    Our living sphere is exposed to a wide range of pathogenic viruses?some known and others of novel origin.Currently, there is no method to continuously monitor the environment for viruses in general, and needless to say, a procedure for the rapid and sensitive identification of a wide variety of viruses for infectious diseases. Traditional approaches based on PCR and immunoassay only enable the detection of known viruses.Now, Ryuji Hatsuki and colleagues at have developed a simple sensing methodology that can potentially detect any virus between the spacing of two electrodes with an intervening several hundreds nanometer-sized gap by measurement of electrical properties of viruses known as impedance spectroscopy.The sensing device was fabricated on a quartz substrate patterned with two measurement electrodes of thin film of gold (250 nm thick) deposited on titanium (1 nm thick); a polydimethylsiloxane (PDMS) sheet forming the measurement chamber wall; and a glass plate as the chamber lid.The researchers discovered that the impedance of three kinds of virus solutions?differing in shape and size?responded differently depending both on the kind of virus and its concentration.Although further studies are required, the proposed method may achieve a sensitivity comparable to that of more traditional, but less versatile continuous virus detection methods, to monitor a wide range of environments for untroubled living conditions.

    Bacteria in various environments have very important roles as biological resources of human beings. However, more than 99% of the microorganisms that inhabit the Earth are difficult or impossible to culture and study experimentally using conventional biochemical techniques. In order to analyze the genes of bacteria that can’t be cultured, it is necessary to trap only one bacterium, break the cell membrane, and take out their DNA. Therefore, as the technique of trapping and breaking bacteria, We attempted the electrodynamic operation. By applying alternating voltages, dielectrophoretic force works and We trap bacterium. By applying pulsed voltage, we generate electroporation and breaking bacteria. We found the proper voltage for trapping E.Coli and demonstrated breaking.

    Much attention has been paid to the exosome which is a small vesicle working as a tool for cell-cell communication in our body. To clarify unexplained phenomena related with the exosome, a smart technique of exosome handling has been strongly demanded to treat such a delicate substance. For resolution of this issue, we used ion-depletion zone for treating bio-substances including exosome without physical loads. We fabricated a prototype device, and demonstrated on the device to form an ion-depletion zone. The repulsive force of ion-depletion zone was visualized by fluorescent imaging of exosomes pushed out from the ion-depletion zone, and it was evaluated by measuring the zone dimension. After the demonstration, the shape and amount of the exosomes were observed by using an atomic force microscope to estimate the effects of the technique on the sample.

    Isolation of environmental microorganisms for single cell analysis is one of the most challenging techniques to clarify the microbial characteristics. We developed an isolation device for microorganisms by dielectrophoretic-trapping with micro/nanogap electrodes. The device has 96 sites for single cell trapping on the substrate of 26 mm × 45 mm. Each site for trapping has an electrode-gap of which interval is 500 nm. Cells were trapped at the gaps by the dielectrophoretic force. To prevent trapping of extra cells, 1 μm wells were aligned on the sites, and only the gap in the wells was in contact with sample solution. By utilizing the device, trapping of Single cells of E. coli and bacteria in intestines of a termite which are environmental microorganisms was successfully demonstrated.

    Separation and condensation of biomolecule and bio-nanoparticles are the fundamental operations for various analyses and reactions. Here, we proposed and experimentally demonstrated a microfluidic device to separate and concentrate biomolecule and bio-nanoparticles using ion depletion zone as a barrier, and we evaluated the electric field strength and flow rate dependency on the ion depletion zone. As a result, the ion depletion zone was strongly depended on the applied field strength and flow rate by samples. The result suggest that it is required the optimization for samples, however there is a possibility to separate a target from a crude of real sample.

    Low-cost but precise fabrication technology for nano/micro devices is required in the field of lab-on-a-chip and Micro Total Analysis Systems (μTAS), etc. Micro molding using of silicone is a beneficial technique and widely applied to fabricate nano/micro device at low cost. However, the silicone has a drawback caused by thermal shrinkage by residual thermal stress during the curing process. To avoid the thermal shrinkage to realize more precise molding, we tried to use a photosensitive (photo-curable) silicone, which requires no heat to cure the material. In this study, it was evaluated the optimum curing condition, optical absorbance, tensile strength, and hardness as the basic properties of photosensitive silicone. The evaluated characteristics indicate that the photosensitive silicone has excellent optical and mechanical properties compared with conventional thermoset ones. Especially, the shrinkage rate of the photosensitive silicone was 0.02 %, whereas the conventional thermoset silicone was 2.9 %.

    Structural color is a way to create color by nanostructures and the related interference color without any pigments. The structural color has many advantage against the conventional pigments, such as no degradation without destroyed the structure, the use of any material to create any color, etc. It is however difficult to fabricate the structure by conventional technique. Here, we tried to fabricate the tree-shaped structural color structure by conventional MEMS fabrication technique. Multi-layered Si/SiO_2 laminar structure was sputter deposited a substrate then RIE etched to form tree-shaped structures. The result is still primitive and not really nanostructure, but the tree-shaped structures was fabricated successfully.

    Separation and concentration of micro/nano particles are the fundamental operations for various analyses and reactions. Here, we proposed and experimentally demonstrated a microfluidic device to concentrate and separate any charged particles using ion depletion zone. The ion depletion zone was generated in the microchannel applied by ion concentration polarization effect, which was controlled by microfluidic device with ion exchange membrane and applied voltage. This work is still primitive but we successfully demonstrated both the concentration and separation.

    &nbsp;&nbsp;&nbsp;Improvement of primary culture of hepatocytes is definitely important for in vitro toxicity tests for various chemicals. Various improvements in terms of culture system have been investigated, such as enhancement in oxygen supply to the cells, coculture with nonparenchymal cells or cell lines, 3D organization into spheroids, etc. However, it is still difficult to maintain in vivo-like high liver-specific functions in in vitro culture. In this report, we briefly explained both the related important research and latest new approach of our research that tried to mimic the in vivo diurnal changes of hepatic functions.

    We introduce a novel MEMS device for various biocells measurements by singular cell level with parallel analysis and high throughput. This paper deals with the details of its fabrication, including device packaging, and the practical measurement of red blood cell using a twin microcantilever type sensor array. Based on a simple measurement of the electrical impedance of a living single red blood cell and its suspension, an important set of data, which can be used to describe living matter, could be obtained. It was confirmed that the electrical impedance between normal and abnormal red blood cells was different in magnitude and phase shift.

    Experiments at a single molecular level provide spatial resolution and dynamic information for the study of biological processes under physiological environments. Here, we show several experimental results: 1) immobilization of a single-molecule in a microchannel using a combination of 3D nano-electrodes and an electrostatic force of dielectrophoresis; 2) drug delivery to a desired location, at which the molecules are immobilized, using a feature of laminar stream; 3) a microreactor for cell-free protein synthesis, as our continuous efforts to develop a micro platform for single-molecule research. For the time being, we are developing each components, however, a system including all the component will be developed in the future for an all-in-one type system to realize chip-based single-molecule analysis.

    The purpose of this study was to develop an automated in situ gene analysis device for use in extreme environmental microbiology. Bacteria or Archaea inhabiting extreme environments such as the deep-sea or the deep subsurface are major targets of gene analysis using this device. By developing a small, fully automated and integrated gene analysis device, a small ROV (Remotely Operated Vehicle) or AUV (Autonomous Underwater Vehicle) can bring the device into underwater extreme environments to analyze microbial communities without risk of contamination and time lag problems. Since a long-term analysis operation will also be available, dynamic properties of microflora can be revealed. In this study, "microfabrication techniques" originated in the field of semiconductor or micromachine engineering are applied to develop an in situ gene analysis device that is capable of flow-through PCR (polymerase chain reaction) analyses. The device is composed of a glass-based "temperature control chip", a silicone rubber-based "microchannel chip" and a pumping system. Microfabrication techniques enable the device to be miniaturized and functionally integrated. Results of evaluation experiments indicate that the developed device can amplify targeted specific DNA fragments from E. coli cells. Furthermore, flow-through PCR operations have been tried under high static pressure conditions up to 30 MPa to simulate analysis in deep-sea environments. PCR is thus possible under high pressure, and the possibility of gene analysis in deep-sea or deep subsurface environments is demonstrated.

    交流ナノポア法(ACナノポア法)は,ユニバーサルに様々な微生物を計測可能で,高速・高精度,小型で現場計測に適した技術である。本研究では微生物サイズの断面(数100nm〜数μm)で長さが数10μm以上のストロー型ナノポアの作製技術を確立し,AC測定の特徴を生かす広帯域でのインピーダンス測定能を生かした微生物センシングを実現する。さらに清掃現場での評価から、現場性能に磨きをかける。これらの開発によってACナノポア法の基盤技術を確立しつつ微生物同定能を高度化し、住環境における微生物センシングおよび業務清掃における衛生状態のオンデマンド計測サービスへの展開を図る。

    Through the demonstration of the measurement of a single bacterial particle flowing in a microfluidic channel, we have demonstrated the electrical impedance measurement of a single bio-nanoparticle under a high electric field. Furthermore, we developed an analysis method to identify bacterial species from the measurement data using machine learning, and pioneered the realization of a "growing sensor," in which artificial intelligence grows in accordance with the amount of measurement and improves the classification performance of bacterial species. As a result, it was clarified that even if the size and material are the same, impedance characteristics can be obtained depending on the surface condition. By utilizing these characteristics and also using machine learning, we have demonstrated that bacteria of almost the same size can be classified with an accuracy of more than 95%.

    空間のサイズを小さくしていくと体積に対する表面積の割合が大きくなり、ナノメートル近くにもなると内部の物理が表面の状態に支配されるようになります。本研究では、分子1個の大きさに近いナノメートルサイズの流路に生ずるこうした極微小効果を利用して、生体分子を1分子ずつ流しながら、途中に組み込んだ電極を使って分子の動きを制御したり、1分子を測定する、1分子分析法の創成を目指します。
