    A superfluid helium insert was developed for cryogenic microscopy of millimeter-sized specimens. An optical-interferometric position sensor, cryogenic objective mirror, and piezo-driven cryogenic stage were fixed to an insert holder that was immersed in superfluid helium. The single-component design stabilized the three-dimensional position of the sample, with root-mean-square deviations of ( x, lateral) 0.33 nm, ( y, lateral) 0.29 nm, and ( z, axial) 0.25 nm. Because of the millimeter working range of the optical sensor, the working range of the sample under the active stabilization was ( x, y) 5 mm and ( z) 3 mm in superfluid helium at 1.8 K. The insert was used to obtain the millimeter-sized fluorescence image of cell nuclei at 1.8 K without a sample exchange.

    The photosynthetic reaction center of heliobacteria (hRC) is a homodimeric chromoprotein responsible for light harvesting and photoelectric conversion. The fluorescence of the hRC is radiated from a bacteriochlorophyll (Bchl) g having the lowest energy level, called red-Bchl g. The homodimeric architecture of the hRC indicates that it includes two red-Bchls g arranged symmetrically in pairs. Red-Bchl g is a fluorescent probe useful for monitoring the energy transfer network in the RC. Here, we show the fluorescence polarization dependences of two red-Bchls g, individually measured with selective excitation of chlorophyll a serving as the primary electron acceptor. The two red-Bchls g exhibit almost the same polarization dependences. Based on the polarization dependence and structural data of the hRC, we propose a candidate molecule for red-Bchl g. The fluorescence spectra of single hRCs represent the spectral heterogeneity reflecting the local conformational inhomogeneity. A time series of the fluorescence spectra indicates occasional peak shifts between blue- and red-shifted states without significant changes in the fluorescence intensity. The spectral fluctuation is interpreted to be due to the local conformational dynamics around a Bchl g mediating the energy transfer, switching the terminal energy acceptor between two red-Bchls g. In conclusion, while the energy transfer network in the RC can be perturbed by microscopic dynamics, the total energy transfer efficiency, i.e., the light-harvesting function, is rather robust. The functional robustness may be due to multiple energy transfer pathways composed of many antenna pigments in the RC.

    Recently, we have been developing the cryogenic fluorescence microscope for nanometer-level imaging (cryo-nanoscope). Although the cryo-nanoscope could be localized the small biomolecules with a nanometer precision, the field-of-view was as small as the size of cell. Here we study a light-sheet excitation for cryo-nanoscope. We examine the two types of the light-sheet excitation methods by wave optical simulation. As a result, the light-sheet excitation using a two objective lens having a NA of 0.6 was much better than that using a one objective lens.

    生化学反応は複数の生体分子がつくる柔らかく複雑な系である。それにも関わらず、高い選択性で効率よく進む。このような反応には水素結合が関わっており、水素結合がつくるネットワークが、このような特異性を産み出していると考えられる。しかし、その実体はよく分かっていない。研究を困難にしている主な原因は、(1)反応に関わる水素結合以外にも無数の水素結合があるため、重要な働きをする水素結合を特定できず、(2)様々な準安定状態の平均が観測されるため、活性に重要な変化が埋もれてしまうからである。そこで、応募研究では、水素結合とタンパク質活性化の関係を研究するモデル系として、光活性化酵素(OaPAC, Oscillatoria acuminata Photoactivated adenylyl cyclase)を用い、そのクライオ1分子観察をおこなうことを目標としている。
    本年度は水素結合状態を正しく知るための蛍光分子の開発をおこなってきた。その結果、Cy7に代表されるような近赤外蛍光性のシアニン色素を用いると、450 - 600 nmの波長領域の発色団の吸収スペクトルを発光励起スペクトルとして観測できることを明らかにした。OaPACへの蛍光標識が予想以上に難しく、来年度でOaPACの研究をまとめることが困難になってきたが、この系を用いて、OaPAC以外の光捕集系タンパク質の研究も同時におこなっていきたいと考えがえている。具体的には、これまで我々が1分子分光の実績があるLight Harveting Complexや、これまで観察をしたことが無いフィコビリソームなどである。どちらも天然状態で発色団を持っており、この発色団周りの水素結合に関する研究を行う予定である。

    In 2017, we developed cryogenic fluorescence microscope equipped with off-focusing imaging system. The microscope have localized individual fluorophores with angstrom spatial precision. However, the off-focusing imaging is not suitable for a cellular imaging because of a strong autofluorescence of cells. In the present project, we have developed a new cryogenic fluorescence microscope equipped with mutifocus imaging system. We demonstrated the mutifocus imaging of an individual fluorophore. The three dimensional position was localized with 1 nm precision. The experimental precision is 1.5 times of the theoretical limit.

    In the present project, we demonstrated two experimental techniques. First, we demonstrated a solid-immersion-lens (SIL) microscope to improve thel resolution and the detection solid angle. We experimentally showed that by using the SIL, the resolution and the detection solid angle were improved by a factor of 2 and 3, respectively. Second, we demonstrated nanometer-accuracy fluorescence microscopy of individual two-color emitters. Depending on the orientation of the transition dipoles of the emitters with respect to the focal plane, their spots were spheroids whose major axes tilted from the axial direction. When acquired with the axial uncertainty of a conventional fluorescence microscope (700 nm), the tilt due to the dipole orientation caused a lateral shift of 20 nm. Improving the axial uncertainty to 18 nm enabled visualization of these two emitters with a 1-nm accuracy. The technique is necessary to accurately determine the 3D position of multiple rare-earth ions in crystal.

    Nuclear spin of rare-earth (RE) ion doped in an inorganic crystal is a solid-state system useful for processing quantum information. In order to utilize RE ions to build an integrated system of an individual spin qubits, the nuclear spins must be individually detected and controlled. However, only a few experiments were successful in resolving nuclear spin states of single RE ion in solid. The major obstacles in photo-luminescence detection of a single ion with a microscopic setup are weak emission of the ion and background emission from the whole crystal. One of the solutions is to use a crystal smaller than the focal volume. Here we took a different approach to develop a single-component aspheric reflecting objective. Owing to a larger solid-angle of a smaller focal volume, the SN ratio improved to a single-ion level.

    Physical and chemical properties of enzyme can be studied using a homogeneous solution of a purified sample of enzyme. However, in living cells, individual protein molecules of enzyme are in different locations to experience different environments, so that they function differently. For understanding enzymatic reactions on the basis of molecular science, observation of individual molecules is essential. In this study we realized spectroscopy of visible-fluorescent single protein molecules.

    A protein has numerous meta-stable structures. In the physiological conditions. The structural fluctuation was pointed out for long years to be important, because the structural fluctuation plays essential role in regulating function of proteins. However, there have been no methods to elucidate the structural dynamics. To break through, we are aiming at getting structural information of single proteins by performing optical spectroscopy of individual proteins. Because measuring a fluorescence spectrum takes at least several milli-seconds, troscopic measurements are performed at temperatures low enough for the structural dynamics to be followed.
    In this study we have succeeded to follow one pigment-protein complex to know the temperature dependence of structural dynamics. The experimental data, which are the very first data on the globe show clearly that substantial part of the structural movement is temperature-independent tunneling motion [Oikawa, et. al. J. Am. Chem. Soc. 130(20008)48501.].
    Spectroscopy at low temperature of single proteins has been limited to the near infrared region. The method was successfully applied to photosynthetic antenna complexes of photosynthetic bacteria. Nevertheless it is worth making an effort to extend the wavelength of the setup to cover the visible region, since many enzymes absorb or emit in the visible. In order to reduce the chromatic aberration of the objective, which is immersed into liquid helium together with a sample, we have developed a reflecting objective [Fujiyoshi, et. al. Appl. Phys. Lett. 91 (2007) 051125.]. With the reflecting objective we have made two-photon excitation and measured fluorescence spectrum of single GFPs. The spectra were taken in the background-free condition, and series of spectra of the same protein revealed structural dynamics of the protein [Fujiyoshi, et. al. Phys. Rev. Lett. 100 (2008) 168101.].

  • 単一分子分光を用いた一個の核スピンの量子状態の観測

    固体中の一個の核スピンの量子状態を光を使って読み出そうという実験に向けて、初年度は液体ヘリウム用の共焦点顕微蛍光分光装置をつくった。集光効率を重視して作成したが微弱蛍光検出の感度が、系として考えているLaF_3中のPr^<3+>イオンを検出するには少なくともあと一桁足らないことが分かった。製作した低温用の顕微分光装置で集光効率にとって最も重要で、しかも通常の室温用の装置に比べて著しく劣っているのが対物レンズなので、市販の顕微鏡用対物レンズの集光率に近づけるべく、液体ヘリウム温度で使える明るい対物レンズの開発に取り組んだ。主な問題点は組みレンズが使えないので色収差を消せないこと、液体ヘリウム(屈折率1.025)を考慮していないため結像能が悪いことである。屈折系ではなく反射系を用いることを検討した結果、レンズ製造の工作精度で技術的に可能な反射型対物レンズの設計を完成させることができた。
    ここで系に要求される発光特性について一考しておく。一個のイオンから発せられる微弱な蛍光を利用する実験なのでもっと良く光る系を選ぶべきであるが、今回の実験では以下のような制約がある。即ち良く光るということは励起状態の寿命が短く、光を吸って励起状態へ移るとすぐに光子を放出して次の光を吸えるようになることなのだが、核スピンの状態による磁気的エネルギーの違いを共鳴光の周波数の差として区別できる程度には寿命が長くなければならない。
    実験の目的に完全に合致した理想的なパラメータを持つ系が現実には存在しないが、理論的上限を知ることは実験を進める上で重要である。Pr^<3+>イオンを例に取ると、基底状態と励起状態における核四極子分裂の差は10MHz程度、励起状態^3Poの寿命は47μsである。スペクトルの寿命幅Δν(半値全幅fwhm)と寿命τとの関係はπΔντ=1であるからPr^<3+>イオンの寿命幅はおよそ10kHzである。核スピン状態をスペクトルで分離するには線幅の10倍の分裂が必要とすると、理論的限界までには数十倍から百倍程度のゆとりがある。

  • 光磁気二重共鳴を用いたPr^<3+>の状態選択NMRによるLaF_3の結晶場の研究

    Pr^<3+>の電子励起状態^3P_0は全角運動量J=0で真空中では核四重極子相互作用を持たない。しかし結晶中では結晶場の影響により核四重極子分裂をおこすため、結晶中での核四重極子分裂を調べることによりイオンの周囲の環境についての情報が得られる。LaF_3結晶中のPr^<3+>について、^3P_0からの発光を利用した光とラジオ波の二重共鳴分光を行った。単色のcwレーザー光をPr^<3+>の^3P_0への遷移に共鳴させてラジオ波を掃引する。励起状態内で磁気遷移を起こした原子からの発光は励起光と磁気遷移の分だけエネルギーがずれており、この発光を励起光と重ねると2つの光の周波数の差で強度に変調が生じる。この強度変調を検出することにより^3P_0状態の核四重極子遷移を測定した(ラマンヘテロダイン法)。核四重極子分裂はフォトンエコーの実験からは0.73MHzと1.12MHzと報告されているが、今回のcwの実験では全く異なった結果を得た。フォトンエコーの結果が示す1MHz近傍は、レーザー由来のノイズが大きいものの目だった遷移は見らず、2MHzより上の領域に多数の遷移が現れる。核スピンI=5/2のPr核の核四重極子遷移は2本なので、この2本の遷移が何らかの環境との相互作用でさらに分裂しているか、あるいは結晶中に複数の環境が存在することが考えられる。吸収スペクトルは結晶中の複数の環境の存在の可能性を示唆しているが、磁気遷移の励起波長依存性から少なくとも磁気的環境は単一であることが分かった。磁気的パラメータの情報を増やすために外部磁場中でのゼーマン分裂の測定を行ってその解析を進めており、この解析から相互作用についての手掛かりが得られると考えている。

    The quadrupole transitions of ^<139> La nuclei around Pr^<3+> doped in LaF_3 have been detected as an optical-rf induced coherent Raman beat on the Pr^<3+>^3H_4-^3P_0 transition (20925 cm^<-1>) at 1.5 K.Because the effective ^<141>Pr nuclear magnetic moment is smaller in the excited ^3P_0 state than in the ground state, the Pr-La interaction decreases upon Pr^<3+> optical excitation, which makes the La quadrupole transitions couple with the Pr optical transition. Four magnetically inequivalent La nuclei were observed, revealing different local environments of La atoms around impurity Pr^<3+> ions.

    凝縮相中で光磁気二重共鳴を行うためにラマンヘテロダイン検出法を用いた装置を立ち上げた。光とラジオ波による二重共鳴によって、準位1から2へ光遷移し電子励起状態内で準位2から3へ磁気遷移するとき、あるいは基底状態内で準位1から2へ磁気遷移し準位2から電子励起状態3へ光遷移するとき、選択則が許せば終状態3から始状態1への発光が起こる。この発光はレーザー光とはラジオ波の周波数分だけ周波数がずれているため、試料を透過したレーザー光にこの発光が重なると、透過光強度がラジオ波の周波数で変調を受ける。微弱な発光を透過光強度のラジオ波帯域の変調信号として感度よく捕えようというのが本研究で用いたラマンヘテロダイン法である。装置は、可視領域の波長可変単一モードレーザー、試料1.5Kの極低温に保つ液体ヘリウムクライオスタット、磁気遷移を起こさせるためのラジオ波源、そしてヘテロダインビ-ト信号を検出する高速のフォトダイオードからなる。LaF3結晶中にドープされたPr3+イオンの1D2状態と基底3H4状態の間の光遷移にレーザー光を共鳴させ、電子励起、基底両状態についてPr核の核四重極子遷移を観測した。ラジオ波の検出の際、入力信号との位相の相関をとることにより、レーザー光や検出器のもつ雑音を除去できることを示した。今後は、二重共鳴のもつ選択性を生かして、C60単結晶のように結晶中に複数のサイトがある系について吸収波長の違いを利用して各々のサイトを選択して磁気共鳴を行う、などの応用をめざす。

    標題のラマンヘテロダインビ-ト検出のESRを行うためには、1.極低温の試料に 2.単色光と 3.ラジオ波を印加し、4.前方散乱光に含まれる数百MHzの微弱なビ-ト信号を検出しなければならない。本年度はこれら4つの条件のうち1から3を達成し、4については改善の後、来年度には本測定ができる状況に達した。以下に本年度の進渉状況を具体的に述べる。
    1.試料を極低温に冷やすために液体ヘリウムのクライオスタットを設計、制作した。さらに液体ヘリウム容器を数Torrに減圧することにより、1.3Kの超流動ヘリウムを得た。クライオスタットの液体ヘリウム容量は5リッターで、1.3Kを10時間維持できる性能を持つ。
    2.単色光はArイオンレーザー励起のTi:Sapphireリングレーザーを用い、波長700〜1000nmで単一モード発振できる。今年度、発振波長を知るための波長計、単一モード発振を確認するためのスペクトラムアナライザーを導入した。以上、1と2の条件が揃ったので予備的測定としてC_<60>単結晶の1.3Kにおける発光スペクトルを測定した。
    3.ラジオ波を試料に効率好く照射するために、容量可変のコンデンサーとコイルの直列共振器を制作した。共振周波数は90〜200MHzの範囲で変化させることができ、Q値は150であった。ラジオ波が実際試料に印加されていることは、1.3KでC_<60>単結晶のODMR信号を測定することによって確認した。
    4.1GHzの帯域を持つシリコン光検出器の出力の中から、ラジオ波と同じ周波数を持つビ-ト成分をスペクトラムアナライザーで検出する。現在、予備的な測定で、数mWの定常光に含まれる10_<-6>の強度のビ-ト成分を検出できることが分かった。実際の測定にはもう数十倍の感度の向上が必要であり、次年度にラジオ波の小信号増幅器を購入して本測定を行う。
