The present studies obtained provided with new concepts for "Supramolecular Allosteric Signal-amplification Sensing" proposed by us. A new series of reporter-modified curdlans showed very high cooperativity and positive homotropic allosterism with high limit of detection. All of the results thus obtained can be expanded to other oligosaccharides and analytes that are difficult to sense in biomolecular media.

Photoexcitation of donor-acceptor or charge transfer complex leads to the excited charge transfer complex as reactive intermediate, which has been recently revealed different from the conventional exciplex in its structure as well as its reactivity. In this study to utilize the excited charge transfer complex for the asymmetric photoreaction, we explored additional supramolecular or weak interaction as precisely stereo-controlling factor. We also examined the photoreactions in the excited triplet state, where a longer excited-state lifetime and thus better control can be attained. Extended work on the relevant photochemical behavior of chiral charge transfer complex is now underway in the following project.

Circularly polarized luminescence (CPL) of organic molecules has been less explored until recently. We focused in this study on the variety of chiral organic molecules including helical chirality in helicenes, propeller chirality in hexaarylbenzenes, as well as Mobius chirality in expanded porphyrins. Through our systematic investigation, we successfully obtained the structure-property relationship for the chiroptical responses, which can be utilized for design principle for advanced organic CPL materials. Results have been appeared in 36 international original papers. In addition, a relevant review article is under preparation and a book related on CD and CPL on small organic molecules is edited, both of which should be distributed shortly.

The author has elucidated the mechanism of dynamic glucan-polythiophene hybridization by using curdlan and schizophyllan as representative unbranched and branched linear polysaccharides and polycationic PyPT as a chromophoric hybridization partner. The in situ hybridization to hetero-triplex [(Cur)2-PyPT] upon addition of an aqueous solution of PyPT to a DMSO solution of Cur at ambient temperatures turned out to be far from completion. It should be emphasized that polycationic PyPT can unzip Cur homo-triplex and hybridize with the resulting homo-duplex in neutral aqueous solutions, despite that Cur homo-triplex does not show any sign of disassembling even at high temperatures in the absence of PyPT or in the presence of neutral PT. Interestingly, the hetero-triplex disassembles to hetero-duplex at very high pHs (>11) through further deprotonation from Cur in the hetero-triplex.

Photochirogenesis has rapidly developed in recent years as an alternative methodology to complement the conventional asymmetric syntheses using chiral catalysts and enzymes. In the present study, we have achieved the simultaneous enhancement of reactivity and stereoselectivity in an enantiodifferentiating photocyclodimerization of 2-anthracenecarboxylic acid through the formation of a novel higher-order (2:2) complexation with L-prolinol, which was shown to be expandable to a wider range of photosubstrates with carboxylic acid moiety/ies. This enabled us to accomplish the organocatalytic supramolecular photochirogenesis for the first time, which is inherently difficult and hence has never been achieved in asymmetric synthesis mediated by a chiral template.

In this study on the asymmetric photoreactions of chiral charge-transfer complex, we explored the generality of wavelength effect and mechanistic investigations. We employed three different photoreactions (two intermolecular and one intramolecular systems) to reveal the different excited complexes are always formed in all systems studied. We have published some of the results in three J. Am. Chem. Soc. papers (Impact Factor = 11.4), delivered the invited lectures in international conferences such as GRC Photochemistry, as well as contributed as invited review article in Chem. Soc. Rev. (Impact Factor = 24.9).

The applicant has expanded his work to polymer-based supramolecular sensing of bio-related materials. To develop a chirality sensing system based on polythiophene, a permethylated cyclodextrin was attached to monothiophene unit with a hexamethylene spacer, which was then polymerized to give novel functional polythiophenes with chirality recognition sites. The cyclodextrin-appended polythiophene showed high enantioselectivity of 13.7 for phenylalanine dipeptide.
To further achieve a more difficult task by constructing a saccharide sensor operative in aqueous media, the applicant employed polysaccharide curdlan (Cur) as a hydrogen-bonding saccharide receptor. Thus, the saccharide sensing by using native Cur/polythiophene complex hybridized in situ as a signal-amplifying reporter enabled us to selectively sense acarbose among 24 mono-, di-, tri, tetra-, and pentasaccharides.

In this study, we first elucidated the chiral properties, guest affinities, and photochirogenic performances of inorganic, organic, and biological supramolecular systems, and then developed various novel chiral supramolecular combination systems and established the multidimensional control strategies, which were jointly applied to several photochirogenic reactions to ultimately achieve unprecedentedly high optical yields of up to 99%, which has been a sort of dream for researchers working in this area of science. We published 97 original papers and reviews as well as the first book on this new interdisciplinary area at the intersection of photochemistry, supramolecular chemistry, and asymmetric synthesis to enhance its visibility in science community.

これらの結果はMacromolecules(ACS)に投稿準備中である。
